Project description:In physiological conditions, few circulating hematopoietic stem/progenitor cells (cHSPC) are present in peripheral blood (PB) and their role remain unsolved. By integrating advanced immunophenotyping, high-throughput transcriptome profiling and functional assays we characterized the biological properties of human cHSPC. We found distinct composition but comparable bone marrow (BM) homing and differentiation potential after xenotransplantation of cHSPC with respect to their BM counterpart. Trafficking HSPC are in a low-cycling pre-activated state with primitive and committed progenitors being transcriptionally and functionally skewed towards erythroid differentiation. Moreover, a subset of circulating lymphoid progenitors are primed for seeding lymphoid organs and can actively contribute to T-cell production in patients treated with gene therapy. In case of hematopoietic impairment, cHSPC content and composition correlate with BM-HSPC state. Altogether, our data unveiled the cHSPC contribution to steady-state hematopoiesis, with a key function in sustaining extramedullary lympho- and erythro-poiesis, and supported their clinical exploitation as biomarker for BM-HSPC.

Diverse Targets of the Transcription Factor STAT3 Contribute to T Cell Pathogenicity and Homeostasis

Project description:STAT3, an essential transcription factor with pleiotropic functions, plays critical roles in the pathogenesis of autoimmunity. Despite recent data linking STAT3 with inflammatory bowel disease, exactly how it contributes to chronic intestinal inflammation is not known. Using a T cell transfer model of colitis we found that STAT3 expression in T cells was essential for the induction of both colitis and systemic inflammation. STAT3 was critical in modulating the balance of T helper 17 (Th17) and regulatory T (Treg) cells, as well as in promoting CD4+ T cell proliferation. We used chromatin immunoprecipitation and massive parallel sequencing (ChIP-Seq) to define the genome-wide targets of STAT3 in CD4+ T cells. We found that STAT3 bound to multiple genes involved in Th17 cell differentiation, cell activation, proliferation and survival, regulating both expression and epigenetic modifications. Thus, STAT3 orchestrates multiple critical aspects of T cell function in inflammation and homeostasis. Project description:Regulatory T (Treg) cells derived from the thymus (tTreg) and periphery (pTreg) play central and distinct roles in immunosuppression, but mechanisms governing the generation and activation of Treg subsets in vivo remain uncertain. Here, we report that mechanistic target of rapamycin (mTOR) unexpectedly supports the homeostasis and functional activation of tTreg and pTreg cells. Further, mTOR signaling is crucial for programming activated Treg cell effector function to protect immune tolerance and tissue homeostasis. Treg-specific deletion of mTOR drives spontaneous TH2 responses and altered barrier tissue homeostasis, associated with reductions in both thymic derived effector Treg (eTreg) and pTreg cells. Mechanistically, mTOR acts downstream of antigenic signals to drive IRF4 expression and mitochondrial metabolism, and accordingly, disruption of mitochondrial metabolism severely impairs Treg cell suppressive function and their homeostasis in tissues. Collectively, our results demonstrate that mTOR coordinates transcriptional and metabolic programs in activated Treg subsets to mediate tissue homeostasis
