Project description:An integrated approach was used to identify genes associated with resistance to Aeromonas hydrophila, an important bacterial pathogen causing aeromoniasis in rohu Labeo rohita. DNA polymorphism and gene expression profiling in lines of L. rohita selected for resistance or susceptibility to challenge with A. hydrophila, and grown in a common environment, were studied using Illumina mRNA-seq of selectively pooled RNA samples.

Identificatrion of marker peptides for the oilseed species chia, coconut, flaxseed, hemp, pumpkin, rapeseed, sesame, and sunflower

Project description:Food fraud is a common issue in the modern food industry. The undeclared use of foreign pro-teins in meat products is a major concern in this context. Oilseeds are ideal for this purpose due to their high protein content and since huge amounts of oil meals are obtained as a by-product of oil production. Therefore, a UHPLC-MS/MS method was developed for the simultaneous de-tection of chia, coconut, flaxseed, hemp, peanut, pumpkin, rapeseed, sesame, soy, and sunflower proteins in meat products. Potential tryptic peptide markers were identified by high-resolution mass spectrometry. The final twenty peptide markers selected, which are specific for one of the ten species targeted were each measured by multiple reaction monitoring. To the best of our knowledge, twelve new heat-stable marker peptides for chia, coconut, flaxseed, pumpkin, rape-seed, sesame and sunflower have not been reported previously.

Early Modulation of the Gut Microbiome by Female Sex Hormones Alters Amyloid Pathology and Microglial Function

Project description:It is well-established that women are disproportionately affected by Alzheimer’s disease (AD). The mechanisms underlying this sex-specific disparity are not fully understood, but several factors that are often associated-including interactions of sex hormones, genetic factors, and the gut microbiome-likely contribute to the disease's etiology. Here, we have examined the role of sex hormones and the gut microbiome in mediating A amyloidosis and neuroinflammation in APPPS1-21 mice. We report that postnatal gut microbiome perturbation in female APPPS1-21 mice leads to an elevation in levels of circulating estradiol. Early stage ovariectomy (OVX) leads to a reduction of plasma estradiol that is correlated with a significant alteration of gut microbiome composition and reduction in A pathology. On the other hand, supplementation of OVX-treated animals with estradiol restores A burden and influences gut microbiome composition. The reduction of A pathology with OVX is paralleled by diminished levels of plaque-associated MGnD-type microglia while estradiol supplementation of OVX-treated animals leads to a restoration of activated microglia around plaques. In summary, our investigation elucidates the complex interplay between sex-specific hormonal modulations, gut microbiome dynamics, metabolic perturbations, and microglial functionality in the pathogenesis of Alzheimer's disease.

Proteomic analysis of gut tissue of Labeo rohita infected with Aeromonas hydrophila

Project description:This study aimed at performing the quantitative proteomics analysis of gut tissue, one of Labeo rohita, one of the important aquaculture fish species. Data was acquired using high resolution mass spectrometry followed by Label free quantification. After performing statistical analysis, a panel of differentially expressed proteins were identified which include extracellular matrix proteins, cytoskeletal proteins, immune related proteins and metabolic enzymes. This analysis revealed important signaling pathways and protein-protein interaction networks which could help in understanding disease pathogenesis. The data acquired in the study would provide a basis for further omics research and can help in elucidating biological processes in healthy or diseased fish.
