Project description:Integration of human papillomavirus (HPV) DNA into the host genome is a critical aetiological event in the progression from normal cervix to intraepithelial neoplasm, and finally to invasive cervical cancer. In this study, we want to know how HPV DNA physical status relates to treatment outcome for cervical carcinomas. Cervical cancer samples were compared with normal sample. And also, we divided 39 cervical cancer patients into four groups according to HPV DNA physical status and investigated differentially expressed gene profiles in these groups using Agilent two-color experiment.

Differentially Expressed Genes in the Peripheral Blood of Patients in Cervical Cancer

Project description:We prospectively investigated gene expression profiles of cervical cancer patients undergoing concurrent chemoradiation. Upregulated genes in the circumstance of anemia were analyzed. Peripheral blood of 20 patients (bulky stage IB –IVA cervical squamous cell carcinomas) undergoing concurrent chemoradiation at four time-points was collected. Total RNA extracted by PaxGene Blood RNA System was analyzed with microarrays and MetaCoreTM functional network analyses. Fifty-three genes were significantly differentially expressed during concurrent chemoradiation. Fetal and embryonic hemoglobin genes were upregulated when patients had been severely myelosuppressed. Twenty-eight genes significantly correlated with the hemoglobin genes are involved in responses to hypoxia and oxygenation, TGF-β signaling, cell cycle suppression, G-protein signaling, and transcriptional regulation. C-Myc has the highest rank in transcriptional co-regulation. Besides, IGKV1D-13 was significantly downregulated in patients with severe hematologic toxicity. These approaches identified biological processes in peripheral blood modulated by concurrent chemoradiation and subsequent anemia. Keywords: chemoradiation, microarray, network analysis

RNA profiling of cervical cancer exosomes using next- generation sequencing.

Project description:In the preset study we isolated and characterized exosomes from both HPV positive and negative cervical cancer cell lines and identified a subset of mRNA’s enriched specifically in HPV positive exosomes through deep sequencing methods. Potential target prediction gene ontology, KEGG pathway analysis revealed possible functions associated with differentially expressed genes of CaCx exosomes. Profiling of these exosomes also revealed the export of HPV 16 associated transcripts (E1,E2, E5, and L2) within SiHa exosomes, however no such export was observed for major oncogenic transcripts (E6 and E7). Our Reverse transcription PCR experiments could validate our deep sequencing results for the absence of E6, E7 transcripts in exosomes. Interestingly, PCR could amplify a truncated version of E6 transcript in HeLa exosomes, which was not detectable in our sequencing results.
