Project description:Plants can respond to insect oviposition but little is known about which responses directly target the insect eggs and how. Here, we reveal a mechanism by which the bittersweet nightshade, Solanum dulcamara, kills the eggs of a generalist noctuid herbivore. The plant responded at the site of oviposition by Spodoptera exigua with neoplasm and chlorotic tissue formation, accumulation of reactive oxygen species and induction of defence genes and proteins. Transcriptome analysis revealed that these responses were reflected in the transcriptional reprogramming of the egg-laden leaf. The plant-mediated egg mortality on S. dulcamara was not present on a genotype lacking chlorotic leaf tissue at the oviposition sites on which the eggs are exposed to less hydrogen peroxide. As exposure to hydrogen peroxide increased egg mortality, while catalase supplementation prevented the plants from killing the eggs, our results suggest that ROS formation directly acts as an ovicidal plant response of S. dulcamara.

Project description:Although a seemingly harmless developmental stage of herbivores, insect eggs trigger efficient plant defenses that include necrosis, callus formation, accumulation of ovicidal compounds and release of volatiles to attract egg predators. The large white butterfly Pieris brassicae deposits batches of 20-30 eggs onto Arabidopsis leaves, causing a large transcriptional reprograming that is drastically distinct from the expression profile triggered by larval feeding. Also, P. brassicae eggs induce localized cell death, accumulation of reactive oxygen species (ROS) and salicylic acid (SA), and expression of PTI-related genes, suggesting that egg-associated molecular patterns (EAMPs) activate a response that is similar to the response induced by microbial pathogens. We previously reported that a crude P. brassicae egg extract (EE, soluble fraction from crushed eggs) induced similar responses as oviposition, including ROS and SA accumulation, cell death and defense gene induction. In order to compare oviposition and EE treatment at the transcriptome level, we analyzed changes in transcipt abundance with P. brassicae EE or after natural oviposition. After 5 days, hundreds of genes were significantly upregulated by each treatment and their induction was highly similar between treatments. This conserved transcriptomic signature thus strongly supports our previous observations that oviposition and EE treatment trigger comparable responses in Arabidopsis.

Subsequent exposure to atmospheric oxygen upon oviposition initiates recommencement of embryonic development. Arrest can be artificially extended for several days after oviposition by incubation of the egg under hypoxic conditions, with development recommencing in an apparently normal fashion after subsequent exposure to normoxia. To examine the transcriptomic events associated with embryonic arrest, RNA-sequencing analysis was performed on embryos from freshly laid eggs and eggs incubated in either normoxia (oxygen tension ~159 mmHg) or hypoxia (<8 mmHg) for 36 hours (h) after oviposition (n = 5 per group). The patterns of gene expression differed markedly among the three experimental groups. Normal embryonic development in normoxia was associated with up-regulation of genes involved in DNA replication, the cell cycle, and mitosis, but these genes were commonly down-regulated after incubation in hypoxia. Many target genes of hypoxia inducible factors, including insulin-like growth factor binding protein 1, were down-regulated by normoxic incubation but upregulated by incubation in hypoxia. Notably, some of the transcriptomic effects of hypoxia in green turtle embryos resembled those reported by others to be associated with hypoxia-induced embryonic arrest in diverse taxa, including budding yeast (Saccharomyces cerevisiae), the annelid Caenorhabditis elegans, and zebrafish (Danio rerio). Thus, while among oviparous species, hypoxia-induced pre-ovipositional embryonic arrest appears to be a unique adaptation of turtles, mechanisms underlying hypoxia-induced embryonic arrest per se may be highly conserved across the diverse taxa in which this mechanism operates. We report the transciptional changes in Chelonia mydas embryos incubated in either normoxia or hypoxia comapred to freshly laid eggs.

Project description:Replicate populations of Aedes albopictus were reared under diapause-inducing short day photoperiod (8h light: 16h dark) and diapause-averting long day photoperiod (16h light:8h dark). Eggs were collected from each replicate and snap-frozen 11d post-oviposition. We hope to characterize the metabolomic and lipidomic profiles of diapausing eggs relative to non-diapause eggs.

How prior oviposition affects plant responses to larval herbivory is largely unknown. Combining bioassays and defense protein activity assays with microarray analyses and metabolite profiling, we investigated the impact of preceding oviposition on the interaction of Solanum dulcamara with the generalist lepidopteran herbivore Spodoptera exigua at the levels of the plant's resistance, transcriptome and metabolome. We found that oviposition increased plant resistance to the subsequently feeding larvae. While constitutive and feeding-induced levels of defensive protease inhibitor activity remained unaffected, pre-exposure to eggs altered S. dulcamara's transcriptional and metabolic response to larval feeding in leaves local and systemic to oviposition. Particularly, genes involved in phenylpropanoid metabolism were stronger expressed in previously oviposited plants, which was reflected by reciprocal changes of primary metabolites upstream and within these pathways. Our data highlight that plants integrate signals from non-threatening life stages of their natural enemies to optimize their response when they become actually attacked. The observed transcriptional and metabolic reshaping of S. dulcamara's response to S. exigua herbivory suggests a role of phenylpropanoids in oviposition-primed plant resistance.

Project description:Transcriptional plant responses are specially under the scope on herbivore oviposition studies. However, most of the information comes from Lepidopteran models, where egg laying and feeding are separate events, and little is known regarding the plant reaction to those pests where females feed and oviposit simultaneously. The present study unveiled the oviposition-induced transcriptomic response of plants to Tetranychus urticae eggs. Transcriptional evidence indicated that the early events on the plant response to the egg extract involved ATP, Ca2+ and ROS disbalances. The main phytohormones triggered were JA and ET, with which also participated secondary metabolites, volatiles and glucosinolates as defence mechanisms. Many of the regulated genes are also involved during pathogen-defence mechanisms. Negative regulation of defence mechanisms happened at 72 h. Female fertility was significantly reduced when fed on plants pre-exposed to the eggs, while feeding increased on those plants pre-exposed for 72 h.

Project description:Gené organ (GO) is an exclusive apparatus of female ticks that secretes a protective wax coat onto the egg surface. This wax coat makes eggs more viable in the environment due to its waterproof, stickiness and antimicrobial proprieties. In this work, a combined transcriptomic and proteomic approach shows that GO is engaged in synthesis, modification and oxidation of lipid compounds, not solely secrets compounds uptaked from haemolymph onto the egg surface. GO were analysed at two distinct stages: (i) in the day haematophagy is concluded and ticks detach from host and (ii) during egg laying, precisely at the second day of oviposition. Data presented here show that before oviposition begins, GO undergoes a maturing process in order to be ready to play its role during oviposition. Before oviposition, GO content is directed to machineries of cell division, transcription and protein synthesis, including proteins participating in the cytoskeleton structure. Whereas during egg laying, the content of proteins related to secretion, transporting and metabolism of wax is overrepresented, though protein synthesis is also preserved. Since GO is a wax secretion organ, the work was focused on lipid metabolism, examining a full machinery to synthesize, modify and oxidize fatty acids. In addition, proteins involved on sterols modification, transport and degradation were addressed. Further than highlighting GO importance in tick reproduction physiology, data reveals molecules and pathways crucial to egg wax secretion, and consequently, its development in the environment. Tools targeting these molecules and pathways would impair egg development in environment, and so could be potential targets for novel tick control methods.
