Reprod. Fert. Deveopl., 2011, 23, 889-898) and although Risk sows maintain higher litter growth in the weaned litter, this was at the expense of lower embryonic weight in their subsequent litter compared with Non-Risk sows. To understand the underlying molecular mechanisms involved, we investigated the gene expression profiles of embryos from Risk and Non-Risk litters in this experiment.

Project description:Purpose: RNA-seq method was applied to select genes expressed in feed conversion in pigs and pinpointed genes potentially associated with this feature. Methods: The whole pituitary samples were collected from 16 Puławska pigs (8 pigs per two groups - with high and low feed conversion) maintained at the same environmental and feeding condition. The RNA-seq analysis was performed in 90 single-end cycles on HiScanSQ platform (Illumina) and differentially expressed genes (DEGs) were determined using Deseq2 software. Validation was performed for 10 randomly selected genes using real-time PCR method. Results: The study showed that in pigs characterized by better feed conversion 274 genes were differentially expressed. Genes showed decreased expression encode proteins associated with cholesterol metabolism (APOA1, CH25H, APOE) and regulation of hormone levels (POMC, CTSZ, ADM, AGTR1, SOX8, AGT, CPT1, APOA1, GJA1, MYRIP, CYP1B1, NR1D1). Four genes showed over the 10-fold decrease of expression (CRYM, ISM1 and WNK4), where the CRYM protein binds thyroid hormone for regulatory and developmental roles and WNK4 regulates the balance between NaCl reabsorption and K(+) secretion. In turn, genes showed an increase of expression in the pituitary of pigs having better feed conversion encode proteins were related to cellular response to corticotropin-releasing hormone stimulus (NR4A1, NR4A2, NR4A3) and regulation of type B pancreatic cell proliferation (BIRC5, NR4A3, NR4A1). Conclusions: The present study indicates new genes potentially related to feed conversion, which is one of the most economically important traits in pig breeding. The selected genes should be investigated in future in the terms of their association with fattening traits. Project description:To identify the molecular markers of early pregnancy in pigs, we compared global gene expression profiles of the maternal peripheral blood in pregnant sows with the non-pregnant sows. Peripheral blood sample was collected at 14 days after insemination from the submandibular vein of pregnant and non-pregnant sows respectively, and total RNA was isolated, purified and sent for microarray analysis. This study identified 127 up-regulated and 56 down-regulated genes (FC >= 1.5 and P < 0.05) in peripheral blood from pregnant sows versus non-pregnant sows. Of the differently expressed genes, nine (including LPAR3, RXFP4, GALP, CBR1, CBR2, GPX6, USP18, LHB and NR5A1) were found to exert function related to early pregnancy processes. Seven differentially expressed genes (CHGB, USP18, VWF, LPAR3, NR5A1, PPARD, BIN1) were selected to perform qRT-PCR in the same RNA samples.The expression profiles of these genes detected by qRT-PCR were consistent with those by microarray, which confirmed the reliability of our microarray data.

Project description:The Chinese Erhualian is one of the most prolific pig breeds in the world, which farrows at least five more piglets per litter than Western pig breeds partly due to a greater ovulation rate. Differences in the transcriptome of Chinese Erhualian and Large White ovaries directly result in variation of ovulation rate. To understand the molecular basis related to ovulation rate in Chinese indigenous and Western breeds, samples were collected and used to hybridized. This study reveals many potential avenues of investigation for seeking new insights into ovarian physiology and the genetic control of reproduction. Expression profiling experiments were conducted to identify differentially expressed genes in ovarian follicles at the preovulatory stage of a PMSG-hCG stimulated estrous cycle from 3 Chinese Erhualian and 3 Large White cycling sows by using the Affymetrix Porcine Genechip™.

Project description:Embryonic survival rate is an important factor for the fecundity of sows, and it is affected by endometrium secreting histotroph. A higher concentration of calcium ion has been observed in the uterus of the high prolific Erhualian sows (EH) compared with low prolific ones (EL), which suggests EH sows had a greater establishment and maintenance of pregnancy, and thus increase embryonic survival rate during peri-implantation period. In order to understand the mechanisms of endometrium secreting histotroph affecting embryonic survival rate during Erhualian peri-implantation period, the expression patterns of endometrial mRNA in the EH and EL on day 12 of gestation were analyzed using RNA sequencing technology. 164 differentially expressed genes (DEGs) were identified (Padj < 0.05, |log2(FC)| ≥ 1)，including 46 upregulated and 118 downregulated genes in the EH group compared with EL group. Among these DEGs, 17 DEGs have been previously reported as DEGs in other pig breeds for litter size. Gene Ontology (GO) enrichment indicated that a subset of DEGs were involved in the calcium ion binding and cell adhesion. Solute carrier family 8 member A3 (SLC8A3) and solute carrier family 24 member 4 (SLC24A4) were upregulated genes (Padj < 0.05) in EH group and they were considered as key candidate genes expressed in endometrium to affect embryonic survival rate during peri-implantation period. The present results will aid in understanding the variation of litter size in Erhualian breed during peri-implantation period.

Project description:Slow-growing Korat chicken (KR) is an alternative to broiler chickens that has been used as a national tool to support smallholder farmers due to a higher selling price of KR meat. However, the individual variability of feed efficiency (FE) within a KR stockbreeding results in a lack of competitiveness. Therefore, improvement of FE of KR is of major importance to improve the profitability of livestock production enterprises. Here, we selected two groups of KR with divergent feed conversion ratios (FCR). We performed RNA-sequencing in order to profile KR jejunal transcriptome and to identify the transcriptional variations and biological pathways implied in response to divergent FCR. The biological pathways involved were revealed by enrichment of the Gene Ontology (GO) terms, and the Kyoto Encyclopedia of Gene and Genome (KEGG) pathways. The results showed that main pathways involved in KR FCR divergence were related to immune response, glutathione metabolism, vitamin transport and metabolism, lipid metabolism, and maturation, development and growth. This is the first study to investigate the molecular genetic mechanisms affecting the FCR values in jejunum of slow-growing chicken. This study will be useful in the line-breeding programs for slow growing chickens to improve FE in the stockbreeding and its profitability.
