Project description:Enhanced vertical stratification brought about by warming of the ocean surface is expected to reduce vertical circulation and nutrient input with knock-on effects for phytoplankton. Increased nutrient limitation is one predicted outcome, but the response of phytoplankton is uncertain because long-term adaptation to nutrient limitation has not been studied. We used Cu as a model catalytic nutrient to explore the adaptive response of an oceanic diatom to continuous nutrient deprivation. Thalassiosira oceanica was maintained under Cu-limiting and sufficient conditions for more than 2000 generations and the evolved populations evaluated for physiological traits in a reciprocal transplant experiment. Adaptation to low Cu concentration increased Cu use efficiency, so that under Cu-limiting conditions T. oceanica maintained significantly faster rates of net C assimilation and growth than the control and ancestral populations.

Project description:Background: Ocean temperatures are projected to increase over the coming century, with dramatic consequences for the marine biosphere. Diatoms are important contributors to marine primary production and the ocean carbon cycle, yet the molecular mechanisms that regulate their acclimation and adaptation to temperature are poorly understood. Method: Here we use a transcriptomic approach to identify the molecular mechanisms associated with temperature acclimation and adaptation in closely related colder- and warmer-adapted diatom species. Results: We find contrasting patterns of differential expression at sub- and supra-optimal temperatures across the two species, which may be due to adaptive changes in baseline expression. Frontloaded and divested pathways indicate protein processing machinery, membrane structure, and the balance between temperature-independent photosynthesis and temperature-dependent metabolism are key elements of adaptation to temperature changes. Conclusions: Our findings suggest that transcriptional frontloading and divestment may provide a framework to interpret diatom acclimation and adaptation to temperature and success under future warming.

Project description:Microbes are responsible for cycling carbon (C) through soils, and predicted changes in soil C stocks under climate change are highly sensitive to shifts in the mechanisms assumed to control the microbial physiological response to warming. Two mecha-nisms have been suggested to explain the long-term warming impact on microbial physiology: microbial thermal acclimation and changes in the quantity and quality of substrates available for microbial metabolism. Yet studies disentangling these two mechanisms are lacking. To resolve the drivers of changes in microbial physiology in response to long-term warming, we sampled soils from 13- and 28-year- old soil warming experiments in different seasons. We performed short-term laboratory incubations across a range of temperatures to measure the relationships between temperature sensitivity of physiology (growth, respiration, carbon use efficiency, and extracellular enzyme activity) and the chemical composition of soil organic matter. We observed apparent thermal acclimation of microbial respiration, but only in sum-mer, when warming had exacerbated the seasonally-induced, already small dissolved organic matter pools. Irrespective of warming, greater quantity and quality of soil carbon increased the extracellular enzymatic pool and its temperature sensitivity. We propose that fresh litter input into the system seasonally cancels apparent thermal acclimation of C-cycling processes to decadal warming. Our findings reveal that long-term warming has indirectly affected microbial physiology via reduced C availability in this system, implying that earth system models including these negative feedbacks may be best suited to describe long-term warming effects on these soils. Project description:Studies have demonstrated that marine phytoplankton can adapt to the warmer environment. However, the underlying mechanisms remain largely unknown. Here, we quantified the capacity of a globally distributed marine diatom Skeletonema dohrnii, for rapid evolution under the moderate (24 ℃) and severe (28 ℃) warming scenarios. Whole-genome re-sequencing analysis revealed that the evolutionary adaptation of S. dohrnii to moderate warming was slow (i.e., 700 generations), whereas it was rapid (i.e., 300 generations) under severe warming but suffered a substantial loss of genetic diversity within the population. Genes associated with energy production and lipid metabolism evolved rapidly, particularly under severe warming, suggesting their vital roles in thermal adaptation. Proteomic results also showed the enhanced expression of proteins involved in energy production and lipid metabolism, especially under severe warming. Furthermore, particulate organic carbon and nitrogen production was greatly enhanced in the moderate warming-selected population but increased insignificantly in the severe warming-selected population, indicating more rapid adaptation driven by severe warming. Our results provide molecular insights into the rapid but limited evolution of thermal adaptation in marine diatoms and highlight energy production and lipid metabolism as the most important adaptive strategy. Future warming will affect genetic diversity and population dynamics of diatoms in the ocean.

Project description:The earth’s climate is warming, and warming-induced biological feedbacks to climate threaten to further destabilize ecosystems. In a 30-year soil warming field experiment at the Harvard Forest in central Massachusetts, microbial isolates from heated (+5 degrees C above ambient) show signs of irreversible adaptation to warming in traits associated with altered soil biogeochemical cycling. Our labs have documented physiological adaptation in all three dimensions of microbial activities: growth, resource acquisition, and stress tolerance. We will use metabolomics to investigate the nature of adaptation due to long-term warming, where reduced soil organic matter, reduced soil water holding capacity and potentially increased niche partitioning may be a selective pressure. Specifically we hypothesize that increased drought tolerance of Actinobacteria exposed to long-term warming is due to production of more or different compatible solutes compared to isolates from controls.

Project description:We isolate the cultivable microbiome of a diatom and show that different bacteria have commensal, antagonistic, or synergistic effects on the diatom. One synergistic bacterium enhances growth of the diatom by production of auxin, a phytohormone. The diatom and its synergistic bacterium appear to use auxin and tryptophan as signaling molecules that drive nutrient exchange. Detection of auxin molecules and biosynthesis gene transcripts in the Pacific Ocean suggests that these interactions are widespread in marine ecosystems.

Project description:The Southern Ocean diatom Fragilariopsis cylindrus was grown under half-saturating concentrations of silicate (0.3 uM) and in the blue (480 - 540 nm) and red (550 - 700 nm) light spectrum to investigate gene expression responses using RNA-seq relative to optimal growth conditions (deposited under accession E-MTAB-5024). RNA-seq was performed on three replicate samples for each experimental condition. Total RNA was extracted using a guanidinium thiocyanate-phenol-chloroform extraction protocol, followed by DNase I treatment and RNA purification (Quiagen). First strand cDNA synthesis was performed using random hexamers. Library preparation was performed using the RNA-seq Sample Prep Kit (Illumina) and sequencing was conducted according to the TruSeq RNA sequencing protocol (Illumina) All samples were sequenced together in one flowcell on one lane.
