Project description:As a historical nomadic group in Central Asia, Kazaks have mainly inhabited the steppe zone from the Altay Mountains in the East to the Caspian Sea in the West. Fine scale characterization of the genetic profile and population structure of Kazaks would be invaluable for understanding their population history and modeling prehistoric human expansions across the Eurasian steppes. With this mind, we characterized the maternal lineages of 200 Kazaks from Jetisuu at mitochondrial genome level. Our results reveal that Jetisuu Kazaks have unique mtDNA haplotypes including those belonging to the basal branches of both West Eurasian (R0, H, HV) and East Eurasian (A, B, C, D) lineages. The great diversity observed in their maternal lineages may reflect pivotal geographic location of Kazaks in Eurasia and implies a complex population history. Comparative analyses of mitochondrial genomes of human populations in Central Eurasia reveal a common maternal genetic ancestry for Turko-Mongolian speakers and their expansion being responsible for the presence of East Eurasian maternal lineages in Central Eurasia. In addition, our analyses indicate maternal genetic affinity between the Sherpas from the Tibetan Plateau with the Turko-Mongolian speakers.

Project description:Highly pathogenic avian influenza (HPAI) in wild birds is a major emerging disease, and a cause of increased mortality during outbreaks. The Common buzzard (Buteo buteo) has a considerable chance of acquiring the infection and therefore may function as bio-sentinel for the presence of virus in wildlife. This study aimed to determine the virus distribution and associated pathological changes in the tissues of Common buzzards that died with HPAI H5 virus infection during the 2020-2021 epizootic. Eleven freshly dead, HPAI H5 virus-positive Common buzzards were necropsied. Based on RT-PCR, all birds were systemically infected with HPAI H5N8 virus, as viral RNA was detected in cloacal and pharyngeal swabs and in all 10 selected tissues of the birds, with mean Ct values per tissue ranging from 22 for heart to 32 for jejunum. Based on histology and immunohistochemistry, the most common virus-associated pathological changes were necrotizing encephalitis (9/11 birds) and necrotizing myocarditis (7/11 birds). The proventriculus of two birds showed virus-associated necrosis, indicating tropism of this virus for the digestive tract. Our advice is to collect at least a miniset of samples including brain, heart, liver, and spleen, as these tissues were positive both by RT-PCR and for virus-antigen-associated lesions.

Project description:In this study, our aim was to determine the common sources of origin of 18 elements in the livers of Common Buzzards collected during the breeding season in an extensive agricultural landscape in south-east Poland with respect to age (adults and immatures) and sex (males and females). In all 34 specimens collected, the element concentrations followed the pattern of S > Na > Fe > Mg > Zn > Si > Cu > Mn > Ba > Se > B > Pb > Hg > Cd > Cr > Ni > Sr > V. Among the heavy metals examined, only the concentration of Pb was relatively high. Given the prevalence of farmland in the studied area (and the wide use of fertilizers), common use of lead-hunting ammunition and moderate concentration of Pb in fertilizers, the indirect influence of hunting ammunition ingested with food or as gastroliths was apparently responsible for the elevated levels of Pb in the livers of Common Buzzards. In our study, no significant sex-related differences were detected in the hepatic concentrations of any element. However, a significant age effect was observed for three elements, which had elevated levels in adults (Hg) and immature birds (B, Pb), and a significant age x sex interaction was found for S and Fe. These results might be explained by the importance of these elements in bone growth in immature birds (B), variable strategies of foraging between adults and immature birds (Pb), and possible intersex differences in the immature cohort in response to the presence of lead (S, Fe).

Project description:Balancing selection is a major mechanism to maintain colour polymorphisms over evolutionary time. In common buzzards, variation in plumage colour was reportedly maintained by a heterozygote advantage: heterozygote intermediates had higher fitness than homozygote light and dark morphs. Here, we challenge one of the basic premises of the heterozygote advantage hypothesis, by testing whether plumage colour variation in common buzzards follows a one-locus two-allele inheritance model. Using a long-term population study with 202 families, we show that colour variation in buzzards is highly heritable. However, we find no support for a simple Mendelian one-locus two-allele model of inheritance. Our results rather suggest that buzzard plumage colour should be considered a quantitative polygenic trait. As a consequence, it is unlikely that the proposed heterozygote advantage is the mechanism that maintains this genetic variation. We hypothesize that plumage colour effects on fitness might depend on the environment, but this remains to be tested.

Project description:Second-generation anticoagulant rodenticides (SGARs) are widely used to control rodent populations, resulting in the serious secondary exposure of predators to these contaminants. In the United Kingdom (UK), professional use and purchase of SGARs were revised in the 2010s. Certain highly toxic SGARs have been authorized since then to be used outdoors around buildings as resistance-breaking chemicals under risk mitigation procedures. However, it is still uncertain whether and how these regulatory changes have influenced the secondary exposure of birds of prey to SGARs. Based on biomonitoring of the UK Common Buzzard (Buteo buteo) collected from 2001 to 2019, we assessed the temporal trend of exposure to SGARs and statistically determined potential turning points. The magnitude of difenacoum decreased over time with a seasonal fluctuation, while the magnitude and prevalence of more toxic brodifacoum, authorized to be used outdoors around buildings after the regulatory changes, increased. The summer of 2016 was statistically identified as a turning point for exposure to brodifacoum and summed SGARs that increased after this point. This time point coincided with the aforementioned regulatory changes. Our findings suggest a possible shift in SGAR use to brodifacoum from difenacoum over the decades, which may pose higher risks of impacts on wildlife.
