Project description:The study's objective is to investigate the mode of action of a bispecific CD19-targeted CD28 agonist (RG6333, CD19-CD28) in combination with the glofitamab through bulk RNA sequencing of tumor tissue from humanized mice over time. This analysis aims to describe the molecular and immunological changes induced by the combination therapy, which is designed to potentiate T cell-mediated antitumor activity. Glofitamab, a T cell bispecific antibody, targets CD20-expressing malignant B cells and engages CD3ε to deliver a robust TCR signal. To enhance this effect, CD19-CD28 was developed to provide a necessary costimulatory signal by binding to CD19 on tumor-infiltrating T cells. This agent is engineered to require concurrent TCR signaling and CD19 target presence, avoiding the issues associated with previous superagonistic antibodies. In this study, we explore the synergistic potential of CD19-CD28 with glofitamab in activating T cell responses and inducing tumor regression in humanized mouse models. The RNA-Seq analysis identified gene expression signatures and immune pathways activated by the treatment. These findings underscore the promise of CD19-CD28 as an effective combination partner to glofitamab.

To capture transcriptomes reflecting pathophysiology and therapeutic development of metabolic diseases, we subjected the humanized mice to dietary intervention and the key metabolic transcriptional factor agonist treatments. We then performed rna exoression profiling analysis using data obtained from nanopore direct RNA sequencing of these humanized mice.

ATAC-Seq experiment of humanized liver mice with metabolic treatments

Project description:To make the human liver accessible to metabolic treatments, we employed a liver-specific humanized mouse model in which approximately 50% of the mouse hepatocytes were replaced by human ones. For the dietary treatment, the humanized mice were allowed free access to food (AL, n=4 for donor1, n=3 for donor2) or subjected to a twenty-four hours food withdrawal (Fast, n=4 for donor1, n=3 for donor2). For the transcription factor agonist treatments, the humanized mice were injected with DMSO (n=4), fenofibrate (n=4, 50mg/kg, Sigma-Aldrich, Cat. F6020), rosiglitazone (n=4,10mg/kg, Sigma-Aldrich, Cat. R2408) and GW4064 (n=4, 30mg/kg, Sigma-Aldrich, Cat. G5172) by i.p. injection. The livers were collected after 6 hours fasting and stored in liquid nitrogen immediately after mice sacrificed.

Project description:The major goal of this experiment was to identify the overall changes in gene expression of human CD4+CD28 null T-cells that develop after repetitive [xeno] antigen stimulation, in comparison to normal CD4+CD28+ T-cells. These cells were derived by adoptive transfer of human CD4 T-cells from a normal donor (all CD28+) into immunodeficient mice. Chimeric human CD4+CD28+ were isolated from 4 mice, and chimeric human CD4+CD28 null cells from two of these animals after expansion and T-cell differentiation in vivo. Animals were euthanized, and single cell suspensions of splenocytes derived. CD4 T-cell subpopulations were isolated by fluoresence activated cell sorting, RNA was extrated, labeled and hybridized to whole human gene expression arrays. Overall changes in the gene expression were identified using GEDI (gene expression dynamic inspector). 603 genes were found to be statistically significant (P<0.05) between the CD4+CD28+ and CD4+CD28null cells. Gene expression in self-activating- and non-self-activationg CAR-T cells with CD28 or 4-1BB costimulatory domain.

Project description:We observed self-activation of SKM-CAR-T cells which target novel mesothelioma antigen. CAR-T with CD28 costimulatory domain showed signs of exhaustion while those with 4-1BB costimulatory domain did not. To elucidate molecules responsible for self-activation-induced exhaustion, we performed RNA-seq analysis of SKM-CAR-T cells with CD28 or 4-1BB and CD19-CAR-T cells with CD28 or 4-1BB.
