Project description:Reversible MHC class I deficiency on tumour cells is commonly caused by coordinated silencing of antigen-presenting machinery genes and restorable by IFNγ. Here we describe association of DNA demethylation of selected antigen-presenting machinery gene regulatory regions located in the MHC genomic locus (TAP-1, TAP-2, LMP-2, LMP-7) upon IFNγ treatment with MHC class I upregulation on tumour cells. Our novel findings demonstrate that IFNγ acts as an epigenetic modifier upregulating the expression of antigen-presenting machinery genes through DNA demethylation. Our data also cast more light on the role of DNA methylation in tumour cell escape from specific immunity.

Inhibition of PARP14 restores response to α-PD1 immune checkpoint inhibition in tumours with resistance driven by IFNγ. Mouse tumour samples

Project description:While immune checkpoint blockade therapy (ICBT) benefits cancer patients, many fail to maintain their response due to adaptive resistance mechanisms. IFNγ is critical for cellular immunity, but also promotes adaptive resistance to ICBT. We have established a role for PARP14 in mediating IFNγ-induced adaptive resistance. We confirm that chronic pre-treatment of tumour cells with IFNγ confers resistance to α-PD-1 antibodies in syngeneic mouse tumour models. We identified that PARP14 was consistently upregulated in cancer cells treated chronically with IFNγ as well as in IFNγ-high melanoma samples. Further, we showed that PARP14 knockdown or pharmacological inhibition restores sensitivity to α-PD-1 antibodies accompanied by increased immune cell infiltration into tumours but the decreased presence of regulatory T cells. RNA sequencing analysis of tumours and cultured cells treated with PARP14 inhibitor showed an upregulation of inflammatory-related pathways. In conclusion, PARP14 is an actionable target for reversing IFNγ-driven adaptive resistance to ICBT.

Whole-organ analysis of TGF-b-mediated remodelling of the tumour microenvironment

Project description:Tissue clearing technique may be one of the most powerful strategies for a comprehensive and unbiased analysis of disease progression. Here, we improved a tissue clearing protocol and explored its application in a mouse tumour model of experimental lung metastasis. We established an integrated pipeline for 3D profiling of the tumour microenvironment in combination with our new clearing protocol and machine learning. This pipeline distinguished each cellular component within the tumour microenvironment, and provided information on their spatial relationships. We further explored the role of TGF-b in the tumour microenvironment using this integrated pipeline. TGF-b-stimulated cancer cells enhanced metastatic colonization of unstimulated-cancer cells in vivo when both cells were mixed. RNA-sequencing analysis showed that expression of the genes related to coagulation and inflammation were up-regulated in TGF-b-stimulated cancer cells. Further, 3D profiling of the tumour microenvironments revealed accumulation of platelets or macrophages with TGF-b-stimulated cancer cells, suggesting that TGF-b might promote remodelling of the tumour microenvironment through the activation of these cellular components at the metastatic sites, which further enhances the colonization of cancer cells. Hence, an integrated pipeline that combines tissue clearing, 3D imaging, and machine learning will help further the understanding of the role of the tumour microenvironment.
