Project description:The mechanisms underlying natural variation in lifespan and ageing rate remain largely unknown. We performed microarray experiment to characterise genome-wide expression patterns of a long-lived, natural variant of Drosophila melanogaster resulting from selection for starvation resistance (SR) and compare it with normal-lived control flies (C). We sampled adult females at two time points representing middle age (90% survival) and old age (10% survival) respectively, in three adult diets (malnutrition, optimal food, and overfeeding).

Project description:To make the human liver accessible to metabolic treatments, we employed a liver-specific humanized mouse model in which approximately 50% of the mouse hepatocytes were replaced by human ones. For the dietary treatment, the humanized mice were allowed free access to food (AL, n=4 for donor1, n=3 for donor2) or subjected to a twenty-four hours food withdrawal (Fast, n=4 for donor1, n=3 for donor2). For the transcription factor agonist treatments, the humanized mice were injected with DMSO (n=4), fenofibrate (n=4, 50mg/kg, Sigma-Aldrich, Cat. F6020), rosiglitazone (n=4,10mg/kg, Sigma-Aldrich, Cat. R2408) and GW4064 (n=4, 30mg/kg, Sigma-Aldrich, Cat. G5172) by i.p. injection. The livers were collected after 6 hours fasting and stored in liquid nitrogen immediately after mice sacrificed.

Project description:Genomic and behavioral investigations were performed to determine the effects of a mutation in a Drosophila soluble guanylyl cyclase gene. A mutant DGCalpha1[3] third chromosome was crossed into a natural rover (for[R]) or natural sitter (for[s]) genetic background. (See Osborne et al. 1997; PMID: 9242616.) First instar larvae were collected and grown on 60mm Petri plates containing 10 mL of food until mid-third instar. (Approximate density was 3 animals per mL food). Larvae were collected and washed quickly with distilled water and were flash frozen in liquid nitrogen. Co-reared larvae were tested for behavioural effects. Four independent collections were made for each of the two conditions (Rover_DGCalpha1[3] or sitter_DGCalpha1[3]). Project description:The typical human diet differs substantially in a number of ways from that of other primates. For instance, although many humans consume meat on a regular basis, non-human primate diets are typically dominated by plant foods. In addition, most human populations cook the majority of their foods, whereas all other free-living primate species eat exclusively raw diets. Such differences in food substrates and food processing are hypothesized to exert a large influence on metabolism. If maintained over evolutionary timescales, dietary differences may have contributed to shaping important human-specific features. To index the effect of food substrate and food preparation on metabolism we measured liver gene expression in mice fed diets of meat or tubers served either raw or cooked.

Project description:To compare total RNA levels in miR-124 and mock-transfected cells (Figure S3), 5-10 ug of total RNA from miR-124-transfected cells or mock-transfected cells or universal reference RNA (Stratagene Cat# 740000) was reverse transcribed with Superscript III (Invitrogen Cat# 18080085) in the presence of aminoallul-dUTP 5-(3-aminoallyl)-dUTP (Ambion Cat# AM8439) and natural dNTPs (GE Healthsciences Cat# US77212) with 10 ug of N9 primer (Invitrogen). Subsequently, amino-allyl-containing cDNAs from miR-124 and mock-transfected cells were covalently linked to Cy5 NHS-monoesters, and universal reference cDNA was covalently linked to Cy3 NHS-monoesters (GE HealthSciences Cat# RPN5661). Cy5- and Cy3-labeled cDNAs were mixed and diluted into 50 ul of solution containing 3x SSC, 25 mM Hepes-NaOH (pH 7.0), 20 ug human Cot-1 DNA (Invitrogen Cat# 15279011), 20 ug of poly(A) RNA (Sigma Cat# P9403), 25 ug of yeast tRNA (Invitrogen Cat# 15401029), and 0.3% SDS. Project description:Background: Different diets result in significantly different phenotypes through metabolic and genomic reprogramming. Epigenetic marks, identified in humans and mouse models upon caloric restriction, high fat diet or the intake of specific bioactives, suggest that genomic reprogramming drives this reprogramming and mediates the effect of nutrition on health. Histone modifications encode the epigenetic signal that adapts genome functions to environmental conditions, including diets, by tuning the structure and properties of chromatin. So far, the effect of different diets on the genome-wide distribution of critical histone marks has not been determined Methods: We investigated by chromatin immunoprecipitation sequencing the distribution of the trimethylation of lysine 4 of histone H3 in the liver of mice fed for one year with five different diets including: chow containing corn powder as extra source of plant bioactives or specifically enriched with the cyanidin-3-O-glucoside, high fat enriched obesogenic and caloric restricted pro-longevity diets Conclusions: The comparison of the resulting histone mark profiles revealed functional food containing cyanidin determines broad effect.
