Project description:Artemisia argyi Lev. et Vant., a common ancient compositae species, is widely utilized in traditional Chinese medicine. The underlying mechanism of terpenoid biosynthesis in leaf has been suggested to play an important role in this medicine. However, the transcriptome of A. argyi has not been established. Here, we performed RNA sequencing in leaf, root and stem tissues to identify all possibly transcribed genes. We assembled a total of 99,807 unigenes by analyzing the expression profiling that were generated from the three tissues. Of them, 67,446 unigenes (67.58%) were annotated from public databases including GO, KEGG, COG. We further performed differential gene expression analysis between leaf with stem and root tissue. Our findings revealed that a total of 7,725 unigenes were specified transcribed in leaf. In particular, we determined multiple genes, which encode significant enzymes including HMGR, MVD, DXS, DXR, HDS and HDR, and transcription factors related to terpenoid synthesis. This study established a valuable resource of transcriptome and identified many transcribed genes related to terpenoid biosynthesis, providing the genomic basis for further studies on the molecular mechanism of the medicine for this species.

Project description:To shed light on the biosynthesis mechanisms of these diterpenes, we performed the first high-throughput RNA-seq for A. paniculata leaves with time-serialized MeJA treatments. The transcriptomic approach not only defined a comprehensive transcriptome (comprised of 20,629 highly expressed genes), but also profiled time-serialized gene expression patterns with 4,463 up-regulated genes and 6,517 down-regulated genes responding to MeJA induction. These up-regulated genes significantly enriched in biological processes like stress responses, metabolic pathways, and secondary metabolites biosynthesis, especially terpene/terpenoid metabolic process. Consistent with the gradual accumulation of andrographolide, the expression of most genes involved in MEP pathway showed progressive increase. In the hypothetical terpenoid metabolism pathway, ApCPS2, which was putatively involved in andrographolide synthesis, dramatically increased its expression. Among other co-regulated genes, 154 genes encode cytochrome P450s (CYP450s); 42 genes were predicted as UDP glycosyltransferases (UGTs); and 363 genes belong to transcription factors.

Project description:In previous work, cephalotaxine, harringtonine, homoharringtonine were shown to be accumulated differentially after various stimuli. Especially, after MeJA treatment, the concentration of 3 cephalotaxus alkaloids all showed decreasing. We speculated that the genes expressed lower after MeJA treatment might encode some enzymes responsible for the biosynthesis of cephalotaxus alkaloids. Therefore, choosing the sample treated with MeJA and the control sample for comparative iTRAQ analysis will greatly facilitate dissection of the genes involved in the biosynthesis of cephalotaxus alkaloids and even the acyl portions of cephalotaxus ester alkaloids. This approach is widely used for mining and identifying novel genes in the biosynthesis of secondary metabolites without genome data in plants.

Project description:Dendrobium plants are perennial herbs in the family Orchidaceae (Dendrobium Sw.). Due to protocorm can also produce plant-specific useful metabolites, protocorm is becoming a good substitute. MicroRNAs play essential roles in plant growth, development, and the response to environmental stresses, and they are widely used for prediction of molecular functions for biosynthesizing active comportments in medicinal plants. To obtain insight into the function of miRNAs in Dendrobium plants. Illumina sequencing of D. nobile protocorm, D. officinale protocorm and D. nobile leaf were conducted. A total of 439, 412 and 432 miRNAs were identified in three samples, and their expression levels were significantly different. Specially, 2, 12 and 4 specific miRNAs were identified. Through integrated GO and KEGG function annotation, miRNAs mainly involved in metabolic pathways, plant hormone signal transduction, biological regulation and protein binding. AACT, MK, DXR and HDS as important enzymes in synthesizing basic precursor isoprene pyrophosphate (IPP). were predicted controlled by 6 different miRNAs in terpenoid backbone biosynthesis pathway. 26 miRNAs participated in Auxin, Cytoklinine, Abscisic acid, Jasmonic acid and Salicylic acid signal transduction pathway. Our results could provide valuable information about miRNAs involved in terpenoid biosynthesis and plant hormone signal transduction pathway in D. nobile and candidate genes for increasing the yield of dendrobine.

Project description:We studied the MeJA response processes at four time points in Sindora glabra, which accumulates abundant sesquiterpenes in the tree trunk. Across the time course, differentially expressed genes involved in terpenoid backbone biosynthesis were significantly enriched, while JAZ genes involved in jasmonic acid pathway and TGA genes involved in salicylic acid pathway were significantly enriched at later time point after treatment.

Project description:In total, 5856 proteins were identified via proteomics analysis, 97 of which were significantly differentially expressed in the liver and affected fatty acid metabolism, unsaturated fatty acid biosynthesis, the peroxisome proliferator-activated receptor (PPAR) signalling pathway, pyruvate metabolism, and terpenoid backbone biosynthesis. Screening identified 10 target proteins, including perilipin‐2 (Plin2), Pdk4, farnesyl diphosphate synthase (Fdps) and Fdft1. Among these, the key proteins were Plin2 and Fdps, which were found to be associated with the PPAR signalling pathway and terpenoid backbone biosynthesis via the relationship networks

Project description:The pyrite mine of Lousal, Grândola, Portugal, was discovered in 1882 and extensively exploited from 1900 until its closure in 1988 when the mining ores were not viable economically. Aiming at becoming a cultural-touristic center, the mining area was rehabilitated raising a Mining Museum and a Science Centre along with the reopening of a mining gallery for public access. In recent times, aerobiological studies on subterranean environments such as caves, mines and archeological necropolis revealed to be an important source of novel bacteria, from which their biotechnological potential are under study. Here we present a Pseudomonas sp. isolated from the mine air with potential secondary metabolite biosynthesis, resistance to antibiotics and virulence factors.
