Project description:The cell-surface glycoprotein melanotransferrin (MELTF) is a member of the iron-binding transferrin superfamily expressed in human melanocytes and other cell types. Dysregulated MELTF-expression in malignant melanoma and other malignancies has been reported before but its specific role in melanomagenesis remains elusive. We performed CRISPR/Cas9-based MELTF deletion in human A375-luc2 malignant melanoma cells followed by transcriptomic analysis in order to explore the mechanistic role of MELTF in malignant melanoma.

We have employed NanoString nCounterTM transcriptomic analysis of isogenic human malignant melanoma cells that differ only by NRAS mutational status (BRAFi-sensitive A375-BRAFV600E/NRASQ61 versus BRAFi-resistant A375-BRAFV600E/NRASQ61K), identifying modulation of specific gene expression networks as a function of NRASQ61K-status.

Whole Transcriptome RNA-seq Analysis: the Tumorigenesis and Metastasis of Melanoma

Project description:To better understand the tumorigenesis and metastasis of human metastatic melanoma(MM) , we analyzed the transcriptomes of three cell lines that represent the three distinct stages of MM pathogenesis: the normal stage (HEMn), the onset of MM (A375), and the metastasis stage (A2085). Using the next generation sequencing (NGS) technology, we detected unsymmetrical expression of genes particularly in Chr9, 12, and 14 among three cell lines, an indication of the involvement of these genes in the tumorigenesis and metastasis of MM. These genes were assigned into 41 categories by their expression patterns and their biological functions were subsequently analyzed by Ingenuity Pathway Analysis software. In the top category associated with cancer, HIF1A, IL8, TERT, ONECUT1 and FOXA1 had direct interaction with the transcription factors or cytokines known to be involved in the tumorigenesis or metastasis in other malignant tumors. Our findings suggest that pathway about cytokines regulation in macrophages may play a more prominent role in the pathogenesis of MM. This study provides not only new targets for downstream mechanistic studies in the tumorigenesis and metastasis of MM, but also a new strategy to study the progression of other malignant cancers. Next generation sequencing of A375 melanoma cell line revealed a complex regulatory network when IGFBP5 overexpressed

Project description:IGFBP5, a critical regulators of insulin-like growth factors, has been reported to be involved in many kinds of carcinogenesis and cancer metastases. The role of IGFBP5 in human malignant melanoma (MM), however, remains largely unknown. In this study, we demonstrated that IGFBP5 was aberrantly expressed in human melanoma cells and cancer tissues. Overexpression of IGFBP5 dramatically inhibited the proliferation, migration and invasion of human melanoma cells, whereas knockdown of IGFBP5 by shRNA resulted in the opposite effects, enhanced the cell proliferation, migration and metastasis. In addition, IGFBP5 overexpression suppressed the growth and metastasis of melanoma xenograft tumor in vivo and IGFBP5 overexpression inhibited epithelialâmesenchymal transition (EMT) phenotype and stem cell property of tumor cell, with decreased expression of HIF1Î±, E-cadherin and stem cell markers NANOG, SOX2, OCT4, KLF4 and CD133. Moreover, IGFBP5 exhibited its growth inhibitory activity through inhibition of extracellular signal-regulated Kinase (ERK) and P38-MAPK signaling pathway. Taken together, our findings indicate that IGFBP5 acts as tumor suppressor roles in MM through the modulation of ERK1/2 and P38-MAPK signaling pathway as well as EMT procession and cell stemness, suggesting IGFBP5 as a novel target for human melanoma diagnosis and therapy. mRNA profiles of IGFBP5 over expression (OE) in A375 and A375 cell line were generated using Ion torrent
