Project description:The extracellular matrix (ECM) is a crucial component of solid tumours. Using digital pathology in human and mouse tissues, we define 3 matrix regions: the tumour body (TB), the proximal invasive front (PIF)- and a further distal invasive front (DIF) Increased matrix density coupled to larger fibre thickness, length and alignment are all features of both invasive areas. Importantly, radial fibre orientation is a characteristic of the distal invasive front. Using several in vivo and in vitro 3-dimensional (3D) models, we show that matrix density and alignment induces RhoA/C-ROCK-Myosin activity in cancer cells, while maximal amoeboid directed migration correlates with areas of radial fibre orientation. Importantly, matrix topographical features are enough to prime cancer cells for successful local invasion and metastatic colonisation. Using several spatial OMICs, we describe a mechano-inflammatory transcriptional program linked to matrix topographical features in the invasive fronts of primary tumours that is preserved in metastatic lesions and strongly linked to worse prognosis in patients.

Surface topography guides morphology and spatial patterning of induced pluripotent stem cell colonies

Project description:The relevance of topographic cues for commitment of induced pluripotent stem cells (iPSCs) is largely unknown. In this study, we demonstrate that groove-ridge structures with a periodicity in the submicrometer range induce elongation of iPSC colonies, guide the orientation of apical actin fibers, and direct the polarity of cell division. Elongation of iPSC colonies impacts also on their intrinsic molecular patterning, which seems to be orchestrated from the rim of the colonies. BMP4-induced differentiation is enhanced in elongated colonies, and the submicron grooves impact on the spatial modulation of YAP activity upon induction with this morphogen. Interestingly, TAZ, a YAP paralog, shows distinct cytoskeletal localization in iPSCs. These findings demonstrate that topography can guide orientation and organization of iPSC colonies, which may affect the interaction between mechanosensors and mechanotransducers in iPSCs. Project description:We studied RSV infection in an appropriate in vitro model of respiratory epithelium, a pseudostratified and fully differentiated mucociliary epithelium of normal human bronchial epithelial (NHBE) cells. RSV infection increased actin cytoskeleton without compromising adherent-, tight-, and tricellular-junctions as well as ciliary functions and epithelial tissue barrier integrity. This increased cytoskeleton depends on actin polymerization and the induction of proinflammatory cytokines and chemokines. Thus, we observed a novel signature “increased cytoskeleton” termed “cytoskeletal inflammation” in RSV-infected respiratory epithelium that presumably lacks classical antigen presenting cells, such as resident dendritic cells and macrophages. Our results suggest that RSV-induced cytoskeletal inflammation is a noncanonical earliest host response to the pathogen and contributes to airway inflammation.

Topography-dependent gene expression in migratory glioma cells

Project description:The goal of this experiment was to compare the gene expression of glioma cells migrating on substrates of different topographies and to determine a migration-associated gene profile. Human malignant glioma U251MG cells were cultured on highly-aligned versus randomly-oriented electrospun nanofibers of poly-caprolactone. Cells migrated actively on aligned nanofibers but motility was very restricted on randomly-oriented fibers. Results of gene expression profiling indicated upregulation of a JAK/STAT signature in actively migrating glioma cells. Total RNA was extracted from 3 independent cultures of U251 glioma cells cultured on aligned versus random nanofibers and applied to Affymetrix HG-U133 plus 2 chips. Analysis of gene expression was used to determine a gene-signature correlated with different substrate topography
