Project description:Directional cell migration plays a central role in a wide range of physiological and pathological conditions, such as inflammation and cancer. Steps involved in cell migration include cell polarization, formation of membrane protrusions at the cell front side and adhesion disassembly at the rear side, and a general cytoskeletal rearrangement. However, there are cell-specific and context-specific molecular events acting in the process. Here, we show how to screen for genes and miRNAs involved in migration by the direct integration of a high-throughput gene editing method, the CRISPR-Cas9 knockout pool screening, and a well-established functional assay, the transwell migration assay. We applied this methodology to human fibroblasts migrating in the presence of a biochemical gradient. The results confirm known genes involved in migration, but also highlight new candidates, like the ectopically expressed family of olfactory receptors. This work establishes a methodological advancement in the use of CRISPR technology for functional screening, and represents a resource for candidate genes and miRNA playing a role in human fibroblast directional migration.

Fibroblast-tracks are topographical cues that direct cancer cell migration

Project description:Fibroblasts rely on adhesive structures to migrate. It is generally thought that adhesive structures disassemble once at the rear of the cell to release the cell and allow further movement. However, a significant portion of adhesions is not disassembled and is instead left on the substrate. We observed that virtually all the non-resorbed adhesions remain connected to the substrate by the means of integrin β5. Forward cell migration results in plasma membrane pulling giving rise to tubular structures that are then left on the ground, forming a network of lipid tracks that persist for several days, thus altering substrate’s topography. Cancer cells of different origin exploit fibroblast-tracks as migration railways. The adhesion of cancer cells along tracks is not mediated by focal adhesions, but rather by frustrated clathrin-coated structures (CCSs). Analysis of the composition of track generated by cancer associated fibroblasts allowed to identify integrin αv as the receptor on cancer cells responsible for track recognition. Some cancer cell lines are not able to perform durotaxis (rigidity driven migration), while fibroblasts are very good at it. We thus tested whether, in the presence of fibroblast-tracks, non-durotactic cancer cells would become durotactic by following tracks and this was indeed the case. Hence, fibroblast-tracks are a novel structure capable of physically directing cell migration of different cell types.

Project description:Wound healing is a complex process that repairs organ-tissues including skin after injury. Cell migration is an important process of wound healing and fibroblast growth factor bFGF has been reported to accelerate cell migration. However, knowledge of how bFGF regulates cell migration is limited. Here, we used human foreskin fibroblast primary cells and RNA-Seq based transcriptome analysis to isolate bFGF-regulating signal pathways that regulate cell migration. Among the many pathways identified, an inflammatory response pathway was further examined for its role in fibroblast cell migration through analysis of function by the key regulator NF-κB. Application of Bay11-7082 and LPS, a typical inhibitor and inducer of inflammatory response, promoted and inhibited cell migration, respectively. Biochemical data showed that Bay11-7082 treatment induces the phosphorylation level of JNK，but PI3K inhibitor LY294002 application did not alter the IκBα phosphorylation level. In addition, Bay11-7082 and JNK inhibitor SP600125 together inhibit while LY294002 together with Bay11-7082 maintains normal cell migration, indicating that NF-κB is independent of the PI3K pathway for regulation of JNK activity during cell migration. Taken together, the present study broadens our understanding of the bFGF-regulation mechanism and further identifies a new bFGF-regulating mechanism by which NF-κB regulates JNK during human fibroblast cell migration.

Targeting the Epigenome Reduces Keloid Fibroblast Cell Proliferation, Migration and Invasion

Project description:Keloids are pathological fibroproliferative scars resulting from abnormal collagen deposition within and beyond the margins of the initial cutaneous insult. Keloids negatively impact quality of life cosmetically and functionally and have unsatisfactory treatment modalities with adverse side effects and high reoccurrence rates. Recent studies indicate that epigenetic dysregulation is involved in the development and progression of keloids, suggesting that this could be a viable therapeutic target. The purpose of this study was to evaluate epigenetic targeting drugs, HDACi, LSD1i, Corin, and A-485, as potential therapies for keloids using in-vitro model systems with patient-derived keloid fibroblasts. Results demonstrated that both dual-acting CoREST inhibitor, corin, and HDAC inhibitor, MS-275, reduced fibroblast proliferation more than the LSD1 inhibitor. Corin was the most effective inhibitor of keloid fibroblast migration and invasion across keloid cell lines and primary cells. RNA-seq analysis showed that corin treatment upregulated a significant number of genes with an enrichment in neural differentiation and cell adhesion gene sets. Specifically, corin increased the expression of claudins, which may cause increased cell adhesion and contribute to corin-induced reductions in migration and invasion. Corin downregulated gene sets involved in cell cycle progression, confirmed by reductions in Cyclin A1 and Cyclin B2 at the protein level compared to DMSO. These results highlight the significant role for epigenetic regulation in pathogenic keloid fibroblasts properties and indicate that the epigenetic inhibitor corin may be useful in the prevention and/or treatment of keloids.

Project description:The present study aimed to investigate the role of Thbs4 in skin regeneration and elucidate its effects on cellular responses. Here we show that Thbs4 expression is upregulated in healing skin wounds and wound healing can be promoted by the application of recombinant Thbs4 protein. Thbs4 was shown to promote keratinocyte proliferation and fibroblast migration by activating β-catenin signalling cascade. This indicates that incorporating Thbs4 into novel wound healing therapies can be a promising therapeutic strategy for hard-to-heal chronic wounds.

Project description:Migration is essential for the reproduction and survival of many animals, yet little is understood about its underlying molecular mechanisms. We used the salmonid Oncorhynchus mykiss to gain mechanistic insight into smoltification, which is a morphological, physiological, and behavioral transition undertaken by some juveniles that culminates in a seaward migration. This species is experimentally tractable and, unlike common model species, displays intra- and inter-population variation in migration propensity. Migratory individuals can produce non-migratory progeny and vice versa, indicating a high degree of phenotypic plasticity. One potential way that phenotypic plasticity might be linked to variation in migration-related life history tactics is through epigenetic regulation of gene expression. To explore this, we quantitatively measured genome-scale DNA methylation in fin tissue using reduced representation bisulfite sequencing of F2 siblings produced from a cross between steelhead (migratory) and rainbow trout (non-migratory) lines. We identified 57 differentially methylated regions (DMRs) between smolt and resident O. mykiss juveniles. DMRs were of high magnitude, ranging from 20-62% differential methylation between life history types, and over half of the gene-associated DMRs were in transcriptional regulatory regions. Many of the DMRs encode proteins with activity relevant to migration-related transitions (e.g. circadian rhythm pathway, nervous system development, protein kinase activity). This study provides the first evidence of a relationship between epigenetic variation and life history divergence associated with a migration-related transition in any species. Comparing global DNA methyldation profiles (via RRBS) of resident and smolt O. mykiss siblings using caudal fin tissue.
