Project description:Intrauterine growth restriction (IUGR) is a leading cause of neonatal morbidity and mortality in humans and domestic animals. Developmental adaptations of skeletal muscle in IUGR lead to increased risk of premature muscle loss and metabolic disease later during life. Although transcriptome-wide profiles in muscle associated with IUGR have been reported, their regulation by miRNAs is not well understood. The aim of this study was to identify differences in miRNA expression in porcine skeletal muscle of IUGR and normal-weight (NW) littermates during late foetal development (day 90 of gestation).

Project description:Piglets cloned by somatic cell nuclear transfer (SCNT) show a high incidence of malformations and a high death rate during the perinatal period. To investigate the underlying mechanisms for abnormal development of cloned pig fetuses, we compared body weight, amniotic fluid (AF) metabolome, and placental transcriptome between SCNT- and artificial insemination (AI)-derived pig fetuses. Results showed that the body weight of SCNT pig fetuses was significantly lower than that of AI pig fetuses. The identified differential metabolites between the two groups of AF were mainly involved in bile acids and steroid hormones. The levels of all detected bile acids in SCNT AF were significantly higher than those in AI AF. The increase in the AF bile acid levels in SCNT fetuses was linked with the downregulation of placental bile acid transporter expression and the abnormal development of placental folds (PFs), both of which negatively affected the transfer of bile acids from AF across the placenta into the mother's circulation. Alteration in the AF steroid hormone levels in cloned fetuses was associated with decreased expression of enzymes responsible for steroid hormone biosynthesis in the placenta. In conclusion, cloned pig fetuses undergo abnormal intrauterine development associated with alteration of bile acid and steroid hormone levels in AF, which may be due to the poor development of PFs and the erroneous expression of bile acid transporters and enzymes responsible for steroid hormone biosynthesis in the placentas.

Project description:Impaired muscle growth as a result of IUGR is a major contributor to lifelong reductions in muscle mass (sarcopenia) and metabolic disease risk. We use an ovine model of chronic placental insufficiency which restricts nutrient supply from mother to fetus and results in intrauterine growth restriction. In our model of placental insufficiency and IUGR, fetal hindlimb muscles weigh less than normally-grown control fetuses and have smaller myofiber diameters. Given the frequent correlation between functional changes and transcriptional changes, we investigated the effect of chronic placental insufficiency and IUGR on fetal skeletal muscle gene expression. We found that gene expression in the skeletal muscle is significantly altered by chronic placental insufficiency. In gene ontology analysis, we found that genes involved in cell cycle regulation were most significantly affected, with downregulation of several cyclins. These observations may in part account for decreased muscle weight relative to brain weight observed in the late gestation IUGR fetus.

Project description:Intrauterine Growth Restriction (IUGR) is a pathological condition that hinders the correct growth of the foetus during pregnancy due to oxygen or nutrient deficiency. Consequently, the foetus adapts its metabolism and physiology to survive in such scarce environment. The major adaptation is the so called “brain sparing”; this effect gives priority to brain development to ensure the individual survival. Nevertheless, this does not warrant the normal development of the brain and the risk exists of neurological and cognitive deficits at short or long term. In turn, this adaptation leads to other systemic alterations that affect the energetic metabolism, thus inducing the emergence of a fairly characterized phenotype called “thrifty phenotype”. This phenotype is responsible for the metabolic alterations that last up until adulthood, which increase the incidence of some diseases like diabetes and metabolic syndrome. On the other hand, diets rich in fat are known to exert pernicious effects Using a pig model of IUGR, animals are classified as normal birth weight (NBW) or low birth weight (LBW). We have studied the effect of a long-term high fat diet (HFD) on the neurological alterations regarding the neurotransmitter profile in several brain areas, the morphology of the hippocampus and the proteome of this same brain area. Our hypothesis is that those animals that were affected by IUGR during their gestation, and therefore were born LBW, will present a different susceptibility to a HFD than NBW animals when they become adults. Our results indicate that HFD had a significant effect on the neurotransmitter profile of the hippocampus, amygdala, hypothalamus, striatum and prefrontal cortex. The most affected neurotransmitter was serotonin (5-HT), thus affecting the indolamine pathway. On the other hand, HFD does not provoke relevant changes in the morphology of the hippocampus. Finally, the proteomic analysis revealed that, in some instances, NBW and LBW animals respond to HFD in different ways. In particular, NBW animals present changes in the mitochondrial respiratory chain and oxidative phosphorylation, and in the extracellular matrix and its interaction with the cell. LBW animals present differences in RNA splicing, anterograde and retrograde transport and the mTOR pathway.

Project description:Breast milk has shown neurodevelopmental advantages compared to infant formula, especially in IUGR infants, which may relate to the fat source. This study aimed to compare neurodevelopmental outcomes in normal birth weight (NBW) and intrauterine growth restricted (IUGR) piglets fed a formula diet with either a vegetable oil (VEG) or bovine milk fat source (MILK). Results showed that total plasma lipids were increased by feeding the MILK diet. 82% and 11% of lipid species were differentially expressed between dietary groups in plasma and hippocampus, respectively, with slight agreement between the plasma and brain lipidome. The MILK diet was most effective in increasing brain lipid accretion, mainly comprising phospholipids. Absolute regional brain weights, grey and white matter volumes, and behavior and motor function scores were lower in IUGR piglets with no effects of diet. Cognitive function and gene expression profiles were similar for dietary and weight groups, and overall only minor interactive effects between diet and weight were observed. In conclusion, dietary fat source influenced the plasma and to a lesser degree the hippocampal lipidome, but was unable to improve on IUGR induced brain structural and functional impairments.

Project description:Intra-uterine growth restriction (IUGR) and fetal overgrowth increase the risk of postnatal health. Maternal nutrition is the major intrauterine environmental factor that alters fetal weight. However, the mechanisms underlying maternal nutrition affect fetal development is not entirely clear. We developed a pig model and used isobaric tags for relative and absolute quantification (iTRAQ) to investigate alterations in the placental proteome were obtained from gilts in normal-energy-intake (Con) and high-energy-intake (HE) group, respectively.At 90 d of gestation, the heavy fetuses were found at the tubal ends and light fetuses at the cervical ends of the uterus in Con group and the heavy fetuses had higher glucose concentration than the light fetuses. However, a higher uniformity was noted in HE group. Placental promote between these two positions indicated that a total of 78 and 50 differentially expressed proteins were detected in Con and HE group, respectively. In Con group, these proteins related to lipid metabolism (HADHA, AACS, CAD), nutrient transport (GLUT, SLC27A1) and energy metabolism (NDUFV1, NDUFV2, ATP5C1). However, the differentially expressed proteins in HE group were mainly participation in transcriptional and translational regulation and intracellular vesicular transport.
