Project description:Endometrial stromal cell decidualization is required for pregnancy success. Although this process is integral to fertility, many of the intricate molecular mechanisms contributing to decidualization remain undefined. One pathway that has been implicated in endometrial stromal cell decidualization in humans in vitro, is the Hippo signaling pathway. Two previously conducted studies showed that the effectors of the Hippo signaling pathway, YAP1 and WWTR1, were required for decidualization of primary stromal cells in culture. To investigate the in vivo role of YAP1 and WWTR1 in decidualization and pregnancy initiation, we generated a Progesterone Cre mediated partial double knockout (pdKO) of Yap1 and Wwtr1. Female pdKOs exhibited subfertility, a compromised decidualization response, partial interruption in embryo transport, blunted endometrial receptivity, delayed implantation and subsequent embryonic development, and a unique transcriptional profile. Bulk mRNA sequencing revealed aberrant maternal remodeling evidenced by significant alterations in extracellular matrix proteins at 7.5 days post coitus in pdKO dams, and enrichment for terms associated with fertility compromising diseases like pre-eclampsia and endometriosis. Our results indicate a required role for YAP1 and WWTR1 for successful mammalian uterine function and pregnancy success.

Knockdown of Yap1 and Wwtr1 in normal and fibrotic murine alveolar type II cells

Project description:Yap1 and Wwtr1 were knocked down via siRNA in primary murine alveolar cells derived from normal or fibrotic mice (PBS treated as a control for bleomycin instilled animals, harvested at 14 days). Isolated cells from three independent isolations were plated on tissue culture plates and allowed to adhere for 2 days followed by incubation with siRNA via forward transfection. Samples were harvested 48 hours later for microarray analysis.

Regulation of posterior body and ectodermal morphogenesis in zebrafish by localized Yap1 and Wwtr1

Project description:The vertebrate embryo undergoes a series of dramatic morphological changes as the body extends to form the complete anterior-posterior axis during the somite-forming stages. The molecular mechanisms regulating these complex processes are still largely unknown. We show that the Hippo pathway transcriptional coactivators Yap1 and Wwtr1 are specifically localized to the ectoderm and notochord, and play a critical and unexpected role in posterior body extension by regulating the assembly of Fibronectin underneath the ectoderm and surrounding the notochord. We also find that Yap1/Wwtr1, also acting through Fibronectin, have an essential role in the ectodermal morphogenesis necessary to form the initial dorsal and ventral fins, a process that had been thought to involve bending of an epithelial sheet, but which we now show involves active cell migration. Our results reveal how the Hippo pathway transcriptional program, localized to two specific tissues, acts to control essential morphological events in the vertebrate embryo.

Project description:Purpose: The goals of this study are to compare control Dorsal Forerunner Cells (DFCs) transcriptome (RNA-seq) to the one of embryos depleted for either Vgll4l or Yap/Taz (Yap1 and Wwtr1) and to evaluate the transcriptional contribution of these transcription cofactors to the formation of the zebrafish left right organizer Methods: Knockdown (KD) of Vgll4l and Yap/Taz were performed by injection of morpholinos in Tg(sox17:GFP) embryos at the one cell stage. Single DFCs cluster were dissected out at 90% epiboly. Extraction of Total RNA and cDNA libraries construction were done using SMART-Seq® v4 Ultra® Low Input RNA Kit for Sequencing from clontech and NEB DNA Ultra library construction kit. Whole genome sequencing was done on illumina Hiseq 2000 sequencer. Results/Conclusions: Vgll4l and Yap/ Taz play and essential role in the formation of the left right organizer. They regulate a large number of genes important for differentiation of the organ. They are required for proper expression of major transciprition factors involved in Kupffer vesicle formation as well as for genes involved in ciliogenesis. Both Vgll4l and Yap/Taz act upstream of major pathways known to regulate the formation of the left right organizer. Additionally, Vgll4l is required for the spatio-temporal expression of writters and readers of DNA methylation marks (DNA methylation enzymes and methyl CpG binding proteins) during Kupffer's vesicle differentiation

Multiple roles for Wwtr1 in cardiac wall maturation

Project description:Cardiac trabeculation is a highly regulated process that starts with the delamination of cardiomyocytes from the compact wall to form stereotypical muscular ridges in the developing ventricle. The Hippo signaling pathway has been implicated in cardiac development but many questions remain. We investigated the role of Wwtr1, a nuclear effector of the Hippo pathway, in zebrafish and find that its loss results in hearts with reduced trabeculation. However, in mosaic animals, wwtr1-/- cardiomyocytes contribute more frequently than wwtr1+/- cardiomyocytes to the trabecular layer of wild-type hearts. To investigate this paradox, we examined the myocardial wall at early stages and find that loss of Wwtr1 leads to disruption of the compact wall architecture, as evidenced by the disorganized cortical actin structure and abnormal cell-cell junctions. The mutant compact wall is not able to support trabeculation as, in mosaic animals, wild-type cardiomyocytes are more frequently in the compact layer of mutant than heterozygous hearts. Therefore, we propose that Wwtr1 establishes the compact wall architecture necessary for trabeculation and that it also modulates a cardiomyocyte’s decision to enter the trabecular layer.

Transcriptome modulation by loss-of-YAP1/TAZ in SK-N-MC Ewing Sarcoma cells

Project description:YAP1 (Yes-associated protein 1) and TAZ (transcriptional coactivator with PDZ-binding motif, or WWTR1) are nucleo-cytoplasmic shuttling proteins that can function in the nucleus as transcriptional coactivators. Here we sought to evaluate which genes are regulated by endogenous levels of YAP/TAZ. We compared SK-N-MC cells transfected with a control non-targeting siRNA with cells transfected with a mix of siRNA targeting both YAP and TAZ.
