Project description:Heavy metals have been postulated as significant nitrification inhibitor in wastewater treatment plant. The effect of heavy metals such as Cd2+, Cu2+ and Hg2+ to nitrifying bacterium, Nitrosomonas europaea, was studied in pseudo-steady state batch reactor. Under incubation of Nitrosomonas europaea with 1 ?M CdCl2 for 1 hour, transcripts for 66 of 2460 genes were found at high level, yet transcripts of 50 genes were found at low level. Mercury resistance genes (merACDPT) showed 277-fold up regulation. Keywords: cadmium, stress response, global transcription, mercury resistance genes, merA,

Project description:Transcriptional analysis of the cyanotroph Pseudomonas pseudoalcaligenes CECT5344 in response to cyanide present in wastewaters from industrial activities, such as jewelry and electroplating activities Four-conditions experiment, including three different nitrogen sources (ammonium, sodium cyanide or a cyanide-containing wastewater). One experiment without nitrogen added to the media (nitrogen limited condition). Four biological replicates for each condition (added nitrogen source to the media) plus the experiment without nitrogen source added.
Project description:Wastewater-based epidemiology has been revealed as a powerful approach for the survey of the population's health and lifestyle. In this context, proteins have been proposed as potential biomarkers that complement the information provided by those used up to now (small exogenous molecules, metabolites, and genomic material). However, few is known about the range of molecular species and dynamics of proteins in wastewater and the information hidden in these protein profiles is still to be uncovered. In previous research, we have described for the first time the proteome of wastewater using polymer probes immersed in wastewater at the entrance of a wastewater treatment plant (WWTP). Here, we studied the protein composition of wastewater from municipalities with diverse population and industrial activities. For this purpose, we collected water samples at the inlet of 10 different WWTPs in Catalonia at three different times of the year and the soluble fraction of this material was then analyzed by Liquid Chromatography High-resolution Tandem Mass Spectrometry using a shotgun proteomics approach. The complete proteomic profiles, the distribution among different organisms, and the semiquantitative analysis of the main constituents are described. Excreta (urine and feces) from humans, and blood and other residues from livestock were identified as the two main protein sources. Significant differences between the proteomes in the soluble phase and the particulate material, respectively dominated by eukaryote and bacterial proteins, were observed. Our findings provide new insights into the characterization of wastewater proteomics that allow proposing specific bioindicators for wastewater-based environmental monitoring, including human and animal population monitoring, most notably, for rodent pest control (immunoglobulins, amylases), and livestock processing industry monitoring (albumins).
Project description:The ammonia-oxidizing bacterium Nitrosomonas europaea has been widely recognized as an important player in the nitrogen cycle as well as one of the most abundant members in microbial communities for the treatment of industrial or sewage wastewater. Its natural metabolic versatility and extraordinary ability to degrade environmental pollutants enable it to thrive under various harsh environmental conditions. This model of N. europaea (iGC535) is the most accurate metabolic model for a nitrifying organism to date, reaching an average prediction accuracy of over 90% under several growth conditions. The manually curated model can predict phenotypes under chemolithotrophic and chemolithoorganotrophic conditions while oxidating methane and wastewater pollutants.
It is the first upload of the model.
Project description:Pure cultures of ammonia oxidizing bacterium, Nitrosomonas europaea, are exposed to cyanide in pseudo-steady state batch reactor in presence of ammonia. Nitrosomonas europaea are generally regarded as the most sensitive organism to various inhibitors commonly encountered in the wastewater treatment plants (WWTP). To find stress genes of Nitrosomonas europaea to cyanide known as inhibitor of respiratory process, whole-genome transcript response to cyanide was determined in this research using microarray and qRT-PCR. When 1 uM NaCN inhibits nitrification about 50 %, transcript levels of 35 genes were increased while transcript levels of 29 genes were decreased, showing more than 2-fold in total 2460 genes. moeZ (NE2353), homologue with rhodanases related to detoxification of CN-, showed 7-fold up regulation and gene cluster including moeZ also showed significant up regulation. Keywords: cyanide, stress response, moeZ
Project description:Nitrogen and arsenic contaminants often coexist in groundwater, and microbes show the potential for simultaneous removal of nitrogen and arsenic. Here, we reported that Hydrogenophaga sp. H7 was heterotrophic nitrification and aerobic denitrification (HNAD) and arsenite [As(III)] oxidation bacterium. Strain H7 presented efficient capacities for simultaneous NH4+-N, NO3--N, or NO2--N removal with As(III) oxidation during aerobic cultivation. Strikingly, the bacterial ability to remove nitrogen and oxidize As(III) has remained high across a wide range of temperatures, pH values, and shaking speeds, exceeding that of the most commonly reported HNAD bacteria. Additionally, the previous HNAD strains exhibited a high denitrification efficiency, but a suboptimal concentration of nitrogen remained in the wastewater. Here, strain H7 combined with FeCl3 efficiently removed 96.14% of NH4+-N, 99.08% of NO3--N, and 94.68% of total nitrogen (TN), and it oxidized 100% of As(III), even at a low nitrogen concentration (35 mg/L). The residues in the wastewater still met the Surface Water Environmental Quality Standard of China after five continuous wastewater treatment cycles. Furthermore, genome and proteomic analyses led us to propose that the shortcut nitrification-denitrification pathway and As(III) oxidase AioBA are the key pathways that participate in simultaneous nitrogen removal and As(III) oxidation.
