Project description:Regeneration of alveolar bone is an essential step in restoring healthy function following tooth extraction. Growth of new bone in the healing extraction socket can be variable and often unpredictable when systemic comorbidities are present, leading to the need for additional therapeutic targets to accelerate the regenerative process. This study examined the effect of signaling through the TAM (Tyro3, Axl, Mertk) family of receptors on alveolar bone regeneration. We performed RNA sequencing on human alveolar bone stem cells treated with a pan-TAM inhibitor in vitro. We also performed RNA sequencing on tissue isolated from the extraction socket of WT and Mertk-\- mice.

Expression data in the Cap Stage of the Human Embryonic Tooth Germ

Project description:Numerous genes that play important regulative roles during tooth development in mice have been identified. However, very little is known about gene expression and function in human odontogenesis. We used microarrays to detail the global programme of gene expression underlying tooth development and identified distinct classes of up-regulated genes during in the tooth germs. Tooth germs of molar, incisor, and canine at the cap stage were dissected, respectively, from 12-week-old human embryonic oral cavity for RNA extraction and hybridization on Affymetrix microarrays. We sought to screen for the genes that are strongly expressed in the dental tissues and analysis if these genes related to mammalian tooth development and tooth abnormalities.

Project description:Human bone marrow mesenchymal stromal cells (BM-MSC) could be committed toward a functional lymphoid-like stroma by a combination of TNFalpha (TNF) and Lymphotoxin alpha1/beta2 (LT) (Amé-Thomas et al Blood 2007). Bone marrow and lymph node stromal cells support FL malignant cell recruitment and growth in particular after comittment to a lymphoid-like differentiation in vitro. In addition, more than 70% of FL patients exhibit a bone marrow involvment at diagnosis. We delineate using Affymetrix U133+2.0 microarrays the gene expression profile of BM-MSC obtained from FL patients (FL-MSC) and age-matched healthy donors (HD-MSC) in order to identify a specific FL-MSC signature. In addition, we used Affymetrix microarrays to define the gene expression signature of lymphoid-like stromal cells obtained from HD-MSC by treatment with TNF/LT in vitro. This TNF/LT signature was then used to interpret the gene expression profile of FL-MSC. GEP was performed on 10 BM-MSC from FL patients and 6 from healthy donors, treated or not with TNF(10 ng/mL)/LT(100ng/mL)

Project description:Mesenchymal stem cells (MSCs) are multipotent progenitor cells present in various mesenchymal tissues that undergo strict lineage-specific differentiation programs, faithful to their unique tissue origins. However, the key regulators that activate dental pulp MSC commitment to odontogenesis remain unclear. In this study, we utilized an inducible Cre/loxP system to interrupt BMP signaling in apical MSCs at the onset of molar root formation in order to investigate the functional requirement for BMP signaling and its downstream targets in MSC cell fate determination during tooth morphogenesis.
