Project description:Colon tissues from 6-8 week BALB/c mice and STAT6-/- mice having 3% DSS for 3 days were obtained. Total RNAs were isolated with Trizol (invitrogen) and the RNA-seq library was prepared by the Beijing Genomics Institute. Sequence reads were obtained using BGIseq500 and successfully mapped to mouse genome. Reads counts were normalized based on FPKM, fold changes were calculated for all possible comparisons.

Project description:To investigate the roles of SIRT6 and IL4/IL13-STAT6 pathway in intestinal epithelium homeostasis, we generated IEC-specifc Sirt6 knockout mice and transgenic mice with IEC-specific overexpression of constitutively activated STAT6 (STAT6vt). IECs from these mice were isolated and RNA were extracted and performed RNAseq analysis.

Gene expression in WT and Stat6 KO intestinal stem cells

Project description:Purpose: To examine the expression profile of WT and Stat6 KO intestinal stem cells. Methods: We isolated Lgr5+ ISCs from the intestine tissue, and generated Stat6 KO ISCs through CRISPR/Cas9 approach. Results: Many genes are changed upon Stat6 KO cells. We pay special attention on Wnt/β-catenin signaling that is the most critical pathway in ISCs, and discovered Stat6 drove the expression of Wnt target genes. Conclusions: Stat6 crosstalks with Wnt signaling to drive ISC self-renewal.

Project description:We sought to elucidate the molecular mechanisms whereby LIN28B functions by comparing the gene expression profile of cells constitutively expressing LIN28B to empty vector controls. Accordingly, we performed microarray analysis on total RNA isolated from empty vector LoVo and LIN28B-expressing LoVo colon cancer cell lines. Constitutive LIN28B expression was achieved in the LoVo (ATCC #CCL-229) colon cancer cell line via retroviral transduction of MSCV-PIG-LIN28B. Contol = empty vector MSCV-PIG.
