Project description:Plant cyclophilins are widely involved in a variety of abiotic stress regulation processes. Thirty cyclophilin members were identified from apple and their patterns of response to abiotic stress were examined. Apple seedlings with consistent growth were selected, cultivated under drought stress conditions simulated by PEG6000, sampled according to different time periods, and quickly frozen in liquid nitrogen.Total RNAs of apple plantlets were isolated using an RNA extraction kit (Tiangen, Beijing, China).

Project description:EGCG, as an active oxygen scavenger, has a significant effect on inhibiting the greasiness of apples. However, the impact of greasiness on fruit storage and how EGCG eliminates this effect, as well as its influence on apple quality formation, have not been clarified. In this study, we found that exogenous application of EGCG effectively improved a series of nutritional growth indicators, including seedling growth status and photosynthetic activity. Additionally, it significantly increased indicators of reproductive growth, such as fruit diameter, soluble solids content, surface wax, and surface smoothness. Through our experiments, we further discovered that greasiness primarily affects fruit respiration, accelerating fruit decay and causing harm during storage, while EGCG mitigates this process. We also found that the application of tea residue had similar effects to exogenous EGCG. Collectively, these studies suggest that EGCG could be used as a novel foliar fertilizer to enhance overall fruit quality. Moreover, the application of tea residue has similar effects to EGCG, providing a cost-effective solution for apple production.

Project description:New mechanisms-of-action of anthocyanins (ACNs) provided by a red-fleshed apple compared with a white-fleshed apple ACN-poor, and with an ACN-rich extract on the proteome profile of aorta and heart as cardiovascular key tissues were determined. Hypercholesterolemic Wistar rats were separated into the corresponding groups to analyze the proteomic profile of the aorta and heart tissues using nano-liquid chromatography coupled to mass-spectrometry. Red-fleshed apple downregulated CRP, C1QB and CFP related-inflammation. White-fleshed apple reduced C1QB, CFB, CFD, C3, and C9 related to the complement system, reduced MB and CP related to iron metabolism, and increased ME1, PKM, and PC related to energy homeostasis. ACN-rich extract increased FMOD, TAGLN, and CAP1 related to cellular structure and decreased PRKACA, IQGAP1, and HSP90AB1 related to cellular signaling. Red-fleshed apple rich in ACNs suggested an anti-inflammatory effect while white-fleshed apple reduced the complement system protein-related. An apple matrix effect reduced inflammatory proteins regardless their ACN content.

Project description:Anaerobic digestion (AD) is a core technology in management of urban organic wastes, converting a fraction of the organic carbon to methane and the residual digestate, the biorest, have a great potential to become a major organic fertilizer for agricultural soils in the future. At the same time, mitigation of N2O-emissions from the agricultural soils is needed to reduce the climate forcing by food production. Our goal was therefore to enrich for N2O reducing bacteria in AD digestates prior to fertilization, and in this way provide an avenue for large-scale and low-cost cultivation of strongly N2O reducing bacteria which can be directly introduced to agricultural soils in large enough volumes to alter the fate of nitrogen in the soils. Gas kinetics and meta-omics (metagenomics and metaproteomics) analyses of the N2O enriched digestates identified populations of N2O respiring organisms that grew by harvesting fermentation intermediates of the methanogenic consortium.

Project description:Tomato seeds (Solanum lycopersicum Mill., cultivar Marion) were germinated in vermiculite and 10-day old seedlings were transplanted into 4-inch plastic pots containing a mixture of composted pine bark and vermiculite (3:1) amended with lime and fertilizer. Plants were incubated in a growth chamber (constant 30oC, 14-h photoperiod; 180 ?mol s-1 m-2 light intensity) for 18-21 days before inoculation. Seedlings received a complete liquid fertilizer approximately 7 and 14 days after transplanting. Unwounded roots were inoculated with R. solanacearum by drenching the soil in each pot with 40 ml of water containing 1´107 CFU ml-1 of the wild type or 1´108 CFU ml-1 of the hrp mutant. Non inoculated plants received 40 ml of water. Four days after inoculation, a piece of each stem from about 1-cm above the soil line to just below the cotyledon node was excised, immediately plunged into liquid nitrogen, and stored at -80oC. A separate 0.5-cm stem section from the cotyledon node was removed aseptically from each plant and the number of bacteria in the section determined by dilution plating. Stem pieces from plants systemically colonized by R. solanacearum were pulverized in a mortar under liquid nitrogen, and total RNA was extracted with the Tri-Reagent (Molecular Research Center) and then treated with RNase-free recombinant DNase (Ambion) according to the recommended protocols from TIGR and stored at -70oC. Project description:We performed Illumina sequencing of sRNA libraries prepared from juvenile and reproductive phase buds from the apple trees. A large number of sRNAs exemplified by 33 previously annotated miRNAs and 6 novel members displayed significant differential expression (DE) patterns in juvenile and reproductive stages. The study provides new insight into our understanding of fundamental mechanism of poorly studied phase transitions in apple and other woody plants and important resource for future in-depth research in the apple development.

Project description:miRNAs are key players in multiple biological processes, therefore analysis and characterization of these small regulatory RNAs is a critical step towards better understanding of animal and plant biology. In apple (Malus domestica) two hundred microRNAs are known, which most probably represents only a fraction of miRNAome diversity. As a result, more effort is required to better annotate miRNAs and their functions in this economically important species. We performed deep sequencing of twelve small RNA libraries obtained for fire blight resistant and fire blight sensitive trees. In the sequencing results we identified 116 novel microRNAs and confirmed a majority of previously reported apple miRNAs. We then experimentally verified selected candidates with RT-PCR and stem-loop qPCR and performed differential expression analysis. Finally, we identified and characterized putative targets of all known apple miRNAs. In this study we considerably expand the apple miRNAome by identifying and characterizing dozens of novel microRNAs. Moreover, our data suggests that apple microRNAs might be considered as regulators and markers of fire blight resistance.
