Microbial community dynamics during aerobic granulation in a sequencing batch reactor (SBR).

Project description:Microorganisms in aerobic granules formed in sequencing batch reactors (SBR) remove contaminants, such as xenobiotics or dyes, from wastewater. The granules, however, are not stable over time, decreasing the removal of the pollutant. A better understanding of the granule formation and the dynamics of the microorganisms involved will help to optimize the removal of contaminants from wastewater in a SBR. Sequencing the 16S rRNA gene and internal transcribed spacer PCR amplicons revealed that during the acclimation phase the relative abundance of Acinetobacter reached 70.8%. At the start of the granulation phase the relative abundance of Agrobacterium reached 35.9% and that of Dipodascus 89.7% during the mature granule phase. Fluffy granules were detected on day 43. The granules with filamentous overgrowth were not stable and they lysed on day 46 resulting in biomass wash-out. It was found that the reactor operation strategy resulted in stable aerobic granules for 46 days. As the reactor operations remained the same from the mature granule phase to the end of the experiment, the disintegration of the granules after day 46 was due to changes in the microbial community structure and not by the reactor operation.

Proteomic investigation of a mixed microbial culture synthesizing polyhydroxyalkanoates from fermented dairy manure under aerobic feast-famine conditions

Project description:Polyhydroxyalkanoates (PHAs) are bio-based, biodegradable polyesters that can be produced from organic-rich waste streams using mixed microbial cultures. To maximize PHA production, mixed microbial cultures may be enriched for PHA-producing bacteria with a high storage capacity through the imposition of cyclic, aerobic feast-famine conditions in a sequencing batch reactor (SBR). Though enrichment SBRs have been extensively investigated a bulk solutions-level, little evidence at the proteome level is available to describe the observed SBR behavior to guide future SBR optimization strategies. As such, the purpose of this investigation was to characterize proteome dynamics of a mixed microbial culture in an SBR operated under aerobic feast-famine conditions using fermented dairy manure as the feedstock for PHA production. At the beginning of the SBR cycle, excess PHA precursors were provided to the mixed microbial culture (i.e., feast), after which followed a long duration devoid of exogenous substrate (i.e., famine). Two-dimensional electrophoresis was used to separate protein mixtures during a complete SBR cycle, and proteins of interest were identified.

Shifts in microbial community structure and diversity in a novel waterfall biofilm reactor combined with MBBR under light and dark conditions.

Project description:In this study, a novel, low-cost, easy-maintenance and effective waterfall aeration biofilm reactor (WFBR) was designed to treat wastewater with MBBR. The chemical oxygen demand (COD), nitrogen removal efficiency, and the microbial community structure in this novel system were evaluated for 70 days under light and dark conditions. The COD and ammonium nitrogen (NH3-N) removal efficiency remained at approximately 90% and 100% respectively after 25 days, even if the influent substrate concentration and illumination condition changes. High-throughput sequencing was used to investigate the composition and function of the microbial community in different fillers in the treatment system. Dark padding, illuminate carrier and fabric play the good performance in nitrogen nitrification, denitrification and fixation respectively. The major classes present were Betaproteobacteria (30.2% on average), Cytophagia (19.8%), Gammaproteobacteria (11.7%), Alphaproteobacteria (11.2%), Sphingobacteriia (5.1%), Flavobacteriia (2.6%), Deltaproteobacteria (2.4%), Verrucomicrobiae (0.7%), Chloroplast (0.6%) and Clostridia (0.5%). These results could provide important guidance for the improvement of MBBR or other tradition wastewater treatment process, and could also enrich our theoretical understanding of microbial ecology.
