FABP4 as a critical mediator in osteoporosis: inhibition strategies and therapeutic potential

Project description:Fatty acid-binding protein 4 (FABP4), a key lipid protein in metabolism and inflammation, has been suggested to be linked to osteoporosis (OP), though direct evidence is scarce. Here, we present the first clear evidence of FABP4's significant role in OP, supported by clinical data and comprehensive in vivo and in vitro experiments. Elevated serum FABP4 in OP patients inversely correlates with bone mineral density (BMD), with similar trends observed in OVX mice. While FABP4 does not influence osteoblast differentiation, it promotes osteoclast formation and bone resorption. The FABP4 inhibitor BMS309403, with an IC50 of 0.89 μM, inhibits osteoclast differentiation by modulating calcium ions and suppressing the Ca2+-Calcineurin-NFATc pathway. Oral BMS309403 increased BMD in OVX mice, albeit less effectively than alendronate, whereas bone-targeted PLGA nanoparticles showed comparable efficacy to alendronate. This research identifies FABP4 as a promising therapeutic target for OP, with significant clinical implications.

Fabp4 is Essential for the Maintenance of Leukemia Stem Cells while Sparing Hematopoietic Stem Cells

Project description:Fatty acid-binding protein 4 (FABP4) plays a crucial role in the uptake, transport, and metabolic regulation of fatty acids. Previous research has found that the abnormal expression of FABP4 is associated with certain cancers, and it may become a new target for tumor therapy. We discovered that a haploinsufficient or absent dosage of Fabp4 does not affect the function of hematopoietic stem cells in mice, but significantly impacts the maintenance of leukemia stem cells (LSC). Our findings indicate that while Fabp4 does not influence the initiation of acute myeloid leukemia (AML) transformed by MLL-AF9 (MA9) in mice, it markedly promotes the progression of AML. A haploinsufficient or absent dosage of Fabp4 significantly extends the survival time of MA9 mice. Our research reveals the key role and mechanism of Fabp4 in MA9-LSC, suggesting that Fabp4 is a potential target for selectively eliminating MLL-rearranged leukemia.

Altered gene expression in multiple key pathways in MSCs from people with osteoporosis compared to people who do not have osteoporosis

Project description:Osteoporosis is caused by imbalanced bone remodelling homeostasis and is highly prevalent in post-menopausal women. The impact of osteoporosis on mesenchymal stromal cells (MSCs) is a key area of research as they are the progenitors of osteoblasts and are therefore critical to bone homeostasis. TBioinformatic analysis of transcriptional profiling data revealed specific misregulation of genes involved in the processing and trafficking of cellular components, as well as downregulation of Neo1 which has an important role in controlling cell fate choice. These data indicate that the therapeutic targeting of bone marrow MSCs to increase their numbers, to support the appropriate processing of cellular contents, or to redirect cell fate choice may provide new methods for the treatment of osteoporosis, thereby reducing fracture-associated morbidity and improving quality of life for the 200 million people living with osteoporosis globally.

The RNA Polymerase II factor RPAP1 is critical for Mediator-driven transcription and cell identity

Project description:The RNA Polymerase II Associated Protein 1 (RPAP1) is conserved across metazoa and critical for stem cell differentiation in plants, however, very little is known about its mechanism of action or its role in mammalian cells. Here, we report that RPAP1 is essential for the expression of lineage specifying factors and for viability. Accordingly, inhibition of RPAP1 triggers somatic cell de-differentiation and facilitates reprogramming into pluripotent stem cells. Conversely, interfering with RPAP1 in ES cells severely impairs their differentiation capacity. Mechanistically, we show that RPAP1 is essential for the interaction between Pol II and Mediator, as well as for the recruitment of important regulators, such as the Mediator-specific Pol II factor POLR2M/Gdown1 and the CTD phosphatase RPAP2. In agreement, depletion of RPAP1 disturbs the loading of Pol II and Pol II Ser5 phosphorylation levels and impairs expression of super-enhancer-driven genes. We conclude that Mediator-RPAP1-Pol II is an ancient module, conserved from plants to mammals, critical for establishing and maintaining cell identity.
