Knockdown studies of TAPIR lncRNAs in LNCaP cells

Project description:To unravel the molecular function of TAPIR-1 and -2, four different specific siRNAs were used to knockdown the TAPIR-transcripts in LNCaP-cells. Gene expression changes upon knockdown of TAPIR was assessed by Agilent SurePrint G3 Human Gene Expression Arrays at 24h and 48h after treatment.

Analysis of copy number variations among Chinese cattle breeds

Project description:Here we describe a genome-wide analysis of copy number variations (CNVs) in Chinese domestic cattle by using array comparative genomic hybridization (array CGH) and quantitative PCR (qPCR). We conducted array CGH analysis on 30 male cattle individuals, animals from consisting of 12 breeds of Bos taurus/Bos indicus, 1 Bos grunniens and and two ones of Bubalus bubalis breeds for with beef, and/or dairy or dual purpose. We identified over 470 candidate CNV regions (CNVRs) in Bos B. taurus/B. indicus; 118 candidate CNV regions (CNVRs) in B. grunniens, 139 CNVRs in B. bubalis. Furthermore, based on the Y haplotypes of B. taurus/ B. indicus, Wwe also identified 69, 337, and 251 candidate CNV regions (CNVRs) in the sub-groups of Y1, Y2 and Y3 haplotypes.

Exploring molecular dynamics indicators associated with reproductive performance of Bos indicus cattle in plasma samples through data-independent acquisition mass spectrometry

Project description:Aim of the study was to profile the blood plasma proteome of healthy Bos indicus cattle and use it as a baseline against which to compare different conditions in Bos indicus cattle, such as disease, reproduction associated proteins and pregnancy specific proteins etc. Data-independent acquisition (DIA) bases quantitative proteomics approach coupled to LC-MS/MS methods were used to achieve this aim. An Orbitrap Fusion Tribrid mass spectrometer (Thermo Fischer Scientific, Bremen, Germany) connected to the Easy- nLC-1200 nanoflow liquid chromatography system (Thermo Scientific) was used for data acquisition.
