Project description:Background: Vicia sativa (the common vetch) possesses a predominant zygomorphic flower and belongs to the subfamily Papilionoideae, which is related to Arabidopsis thaliana in the eurosid II clade of the core eudicots. Each vetch flower consists of 21 concentrically arranged organs: the outermost five sepals, then five petals and ten stamens, and a single carpel in the center. Methodology/Principal Findings: We explored the floral transcriptome to examine a genome-scale genetic model of the zygomorphic flower of vetch. mRNA was obtained from an equal mixture of six floral organs, leaves and roots. De novo assembly of the vetch transcriptome using Illumina paired-end technology produced 71,553 unigenes with an average length of 511 bp. We then compared the expression changes in the 71,553 unigenes in the eight independent organs through RNA-Seq Quantification analysis. We predominantly analyzed gene expression patterns specific to each floral organ and combinations of floral organs that corresponded to the traditional ABC model domains. Comparative analyses were performed in the floral transcriptomes of vetch and Arabidopsis, and genomes of vetch and Medicago truncatula. Conclusions/Significance: Our comparative analysis of vetch and Arabidopsis showed that the vetch flowers conform to a strict ABC model. We analyzed the evolution and expression of the TCP gene family in vetch at a whole-genome level, and several unigenes specific to three different vetch petals, which might offer some clues toward elucidating the molecular mechanisms underlying floral zygomorphy. Our results provide the first insights into the genome-scale molecular regulatory network that controls the evolution and development of the zygomorphic flower in Papilionoideae. Project description:Drought is a major environmental stress that limits growth and development of cool-season annual grasses. Drought transcriptional profiles of resistant and susceptible lines were studied to understand the molecular mechanisms of drought tolerance in annual ryegrass (Lolium multiflorum L.). A total of 4,718 genes exhibited significant differential expression in two L. multiflorum lines. Additionally, up-regulated genes associated with drought response in the resistant lines were compared with susceptible lines. Gene ontology enrichment and pathway analyses revealed that genes partially encoding drought-responsive proteins as key regulators were significantly involved in carbon metabolism, lipid metabolism, and signal transduction. Comparable gene expression was used to identify the genes that contribute to the high drought tolerance in resistant lines of L. multiflorum.

Project description:Recent studies have shown that several plant species require microbial associations for stress tolerance and survival. In this work, we show that the desert endophytic bacterium Enterobacter sp. SA187 enhances yield and biomass of alfalfa in field trials, revealing a high potential for improving desert agriculture. To understand the underlying molecular mechanisms, we studied SA187 interaction with Arabidopsis thaliana. SA187 colonized surface and inner tissues of Arabidopsis roots and shoots and conferred tolerance to salt and osmotic stresses. Transcriptome, genetic and pharmacological studies revealed that the ethylene signaling pathway plays a key role in mediating SA187-triggered abiotic stress tolerance to plants. While plant ethylene production is not required, our data suggest that SA187 induces abiotic stress tolerance by bacterial production of 2-keto-4-methylthiobutyric acid (KMBA), known be converted into ethylene in planta. These results reveal a part of the complex molecular communication process during beneficial plant-microbe interactions and unravel an important role of ethylene in protecting plants under abiotic stress conditions.

Project description:B. napus, a widely cultivated oilseed crop spanning roughly 35 million hectares world-wide (Faostat , 2022), faces various stress factors including salt stress which reduces plant height, size, and yield (Shahza d et al., 2022; Naheed et al., 2021). Endophytic microorganisms are known to promote plant growth and biomass production (Rho et al., 2018, Azad et al., 2016, Zhang et al., 2019). In this study, inoculation with endophyte Acremonium alternatum increased both fresh and dry weight under salt stress conditions. Further molecular analyses provided insights into potential mechanisms involved, highlighting a putative role of abscisic acid in mediating ROS metabolism and ion sequestering. These findings contribute to our understanding of plant-fungi interactions and offer promising leads for developing novel biological agents to improve crop production under the challenges posed by climate change.

Project description:Soybean is one of the main sources of oil worldwide. Salinity severely affect its yield. GmSIN1 is a NAC transcription factor coding gene. Its overexpression (OE) transgenic lines greatly improved the yield in both common and saline fields. This study focuses on founding changes genes between GmSIN1 OE transgenic seedlings and control seedlings under salt stress or non-salt stress conditions. Illumina Solexa sequencing platform was used for the comparative analysis of transcriptome profiles in the roots and leaves of GmSIN1 OE transgenic seedlings and WEI6823 (control) seedlings under mock or 150 mM NaCl treatment for 6 hrs.
