Project description:The goal of this study was to compare the skin transcriptomes of non-irradiated and 30 Gy irradiated Pelophylax nigromaculatus and to find the polypeptide sequences that were changed before and after irradiation. Skin mRNA profiles of non-irradiated and 30Gy irradiated Pelophylax nigromaculatus were generated by deep sequencing,using Illumina HiSeq.The sequenced sequences were assembled into transcripts, and the transcripts were hierarchically clustered using the Corset program. Subsequent analysis was performed using the clustered sequences as references:CDS prediction, SNP and InDel analysis, gene expression level analysis, SSR analysis, RNA-seq overall quality assessment, differential expression analysis, differential gene GO enrichment analysis and KEGG enrichment analysis.Through differential analysis, 875 down-regulated mRNAs and 536 up-regulated mRNAs were identified, and 271 significant changes in peptides were found through the rank sum test, which provided the basis for the next study.

A novel antimicrobial peptide found in Pelophylax nigromaculatus.

Project description:BackgroundMany active peptides have been found in frog skin secretions. In this paper, our research focused on Pelophylax nigromaculatus and found a broad-spectrum antimicrobial peptide Nigrocin-PN based on the molecular cloning technique. Thereafter, the "Rana box" function was briefly studied by two mutated peptides (Nigrocin-M1 and Nigrocin-M2). Furthermore, in vitro and in vivo assays were used to characterize the peptide's biofunctions, and the peptide's function in treating multidrug-resistant pathogens was also studied.ResultsNigrocin-PN not only displayed potent antimicrobial abilities in vitro but also significantly ameliorated pulmonary inflammation induced by Klebsiella pneumoniae in vivo. By comparing, leucine-substituted analogue Nigrocin-M1 only displayed bactericidal abilities towards gram-positive bacteria, while the shorter analogue Nigrocin-M2 lost this function. More strikingly, Nigrocin-PN exhibited synergistic effects with commonly used antibiotics; in vitro evolution experiments revealed that coadministration between Nigrocin-PN and ampicillin could delay Staphylococcus aureus antibiotic resistance acquisition. Kinetics and morphology studies indicate that antibacterial mechanisms involved membrane destruction. Furthermore, toxicities and anticancer abilities of these peptides were also studied; compared to two analogues, Nigrocin-PN showed mild haemolytic activity and indistinctive cytotoxicity towards normal cell lines HMEC-1 and HaCaT.ConclusionsA broad-spectrum antimicrobial peptide Nigrocin-PN was discovered from the skin secretion of Pelophylax nigromaculatus. Structurally, "Rana box" played a crucial role in reducing toxicities without compromising antibacterial abilities, and Nigrocin-PN could be a desired therapeutic candidate.

Physiological Response of Pelophylax nigromaculatus Adults to Salinity Exposure.

Project description:Many freshwater ecosystems are becoming more saline, and amphibians, which have permeable skin, are sensitive to this change. We studied the physiological responses to high salinity and recovery from saline exposure in adult frogs (Pelophylax nigromaculatus). Frogs that experienced severe salinity were exposed to saline conditions for 6 days, while those in the moderate group were exposed to saline conditions for 40 days, followed by a recovery period in freshwater for 20 days. Our data showed that during exposure to saline conditions of severe and moderate groups, serum electrolytes increased, protein concentrations decreased, and creatinine, an indicator of renal function, sharply increased. However, renal tissue sampled after exposure did not show renal dysfunction. In addition, serum components that changed during exposure to salinity returned to their initial values during the recovery period. Thus, adult anurans can be resilient, to some extent, to saline conditions in habitats that experience either rapid or slow salinity changes.
