Project description:RNaseH2 is a heterotrimeric endoribonuclease that resolves RNA:DNA hybrids and mis-incorporated ribonucleotides, which are implicated in DNA replication stress and cancer development and potential therapeutic targets. Individual RNaseH2 subunit mRNA or protein levels are found elevated in some cancers, but little is known about the mechanisms behind and impact of RNaseH2 subunit upregulation. We report that RNaseH2 subunits are upregulated at the protein level in response to replication stress induced by oncogenes and chemotherapy drugs hydroxyurea, gemcitabine and camptothecin in human cancer and non-cancer cell lines. We show that inducible overexpression of the RNaseH2B subunit increases the levels of the whole active RNaseH2 complex and can counteract drug-induced RNA:DNA hybrid formation. RNaseH2B overexpression reduces replication fork stalling induced by camptothecin and hydroxyurea but can itself increase RNA:DNA hybrid levels and cause replication stress. RNaseH2 subunit levels do not strongly impact survival of chemotherapy drug treatments but may have more subtle effects on genomic instability and cytoplasmic DNA signalling. In contrast, increased RNaseH2 subunit levels in presence of oncogenic HRAS are required to limit not only RAS-induced replication fork stalling but also cell death. Our findings shed new light on the functions of RNaseH2 and suggest that upregulation of RNaseH2 subunits may be an important aspect of replication stress responses in cancer.

Pan-Cancer DNA Hypermethylation Leads to Transcriptional Upregulation of Homeobox Oncogene

Project description:Cancers have long been recognized to be not only genetically but also epigenetically distinct from their tissues of origin. Although genetic alterations underlying oncogene upregulation have been well studied, to what extent epigenetic mechanisms, such as DNA methylation, can also induce oncogene expression remains unknown. Here, through pan-cancer analysis of 4,182 genome-wide profiles, including whole-genome bisulfite sequencing (WGBS) data from 30 normal tissues and 35 solid tumors, we discovered a strong correlation between gene-body hypermethylation of DNA methylation canyons (broad under-methylated regions) and overexpression of ~43% homeobox genes, many of which are also oncogenes. To gain insights into the cause-and-effect relationship, we used a newly developed dCas9-SunTag-DNMT3A system to methylate genomic sites of interest. The locus-specific hypermethylation of gene-body canyon, but not promoter, of homeobox oncogene DLX1 and POU3F3, can direct increase its gene expression. Together, our pan-cancer analysis followed by functional validation reveals DNA hypermethylation as a novel epigenetic mechanism for homeobox oncogene upregulation.

Project description:Metabolism is tightly coupled with the process of aging, and tumorigenesis. However, the mechanisms regulating metabolic properties in different contexts remain unclear. Cellular senescence is widely recognized as an important tumor suppressor function and accompanies metabolic remodeling characterized by increased mitochondrial oxidative phosphorylation (OXPHOS). Here we showed retinoblastoma (RB) is required for the increased OXPHOS in oncogene-induced senescent (OIS) cells. Combined metabolic and gene expression profiling revealed that RB mediated activation of the glycolytic pathway in OIS cells, causing upregulation of several glycolytic genes and concomitant increases in the levels of associated metabolites in the glycolytic pathway. Knockdown of these genes by small interfering RNAs (siRNAs) resulted in decreased mitochondrial respiration, suggesting that RB-mediated glycolytic gene activation promotes metabolic flux into the OXPHOS pathway. These results suggest that coordinate transcriptional activation of metabolic genes by RB enables OIS cells to maintain metabolically bivalent states that both glycolysis and OXPHOS are highly active. Collectively, our findings demonstrated a previously unrecognized function of RB in OIS cells. To understand the role of RB, we investigated the effect of RB1-knockdown in the transcription profile of oncogene-induced senescent (OIS) cells. IMR90 ER:Ras cells were treated with 100 nM 4-OHT for 6 days to induce senescence. RNA was isolated 6 days after OHT treatment and hybridized to Affymetrix microarrays. SiRNA transfection (control siRNA or siRB1) was performed 4 days before RNA isolation.

Increased glucose availability sensitizes pancreatic cancer to chemotherapy

Project description:Pancreatic cancer (Pancreatic Ductal Adenocarcinoma; PDAC) is highly resistant to chemotherapy. Effective alternative therapies have yet to emerge, and therefore, chemotherapy remains the best available systematic treatment. The discovery of safe and available adjuncts that can improve chemotherapeutic efficacy could potentially improve survival outcomes. We show that a hyperglycemic state enhances the efficacy of conventional single- and multi-agent chemotherapies against PDAC. Molecular analyses of tumors exposed to relatively high glucose levels revealed that GCLC (catalytic subunit of glutamate-cysteine ligase), a key regulator of a metabolic pathway, glutathione biosynthesis, is diminished and underlies chemo-sensitization. Inhibition to GCLC phenocopied the effect of an induced hyperglycemic state in mouse models of PDAC, while rescuing the pathway abrogated the anti-tumor effects observed with chemotherapy treatment under high glucose conditions.

Project description:We used a cardiac-specific tamoxifen-dependent Gal4-ERT2/UAS model to achieve uniform but conditionally regulated expression of the oncogene HRASG12V in zebrafish cardiomyocytes. We found that the inhibition of TOR signaling using rapamycin treatment counteracts neoplasic growth. We analyse the transcriptome of neoplasic ventricles after exposure to DMSO and rapamycin and we applied bioinformatic analysis to determine common and distinctive molecular signatures of neoplasia versus regeneration of the heart.
