Project description:Morchella sextelata, recognized for its medicinal properties and distinctive taste, is a highly valued edible mushroom. Despite its commercial importance, the mushroom's susceptibility to environmental factors and its reactions to herbicides commonly used in cultivation have not been extensively studied. This research aimed to explore how four prevalent herbicides—Trifluralin, Pendimethalin, S-metolachlor, and Acetochlor—affect the mycelial growth of Morchella sextelata. The study involved cultivating the mycelium in a Potato Dextrose Agar (PDA) medium with concentrations of herbicides at ten times the standard application rate, followed by measuring the degree of mycelial growth inhibition. Employing transcriptomic analysis, RNA sequencing was conducted on the mycelium exposed to the herbicides. The subsequent transcriptome profiling indicated that the application of these herbicides impacted various biological pathways within Morchella sextelata, with a notable influence on glycolysis/gluconeogenesis and the glyoxylate cycle, which are crucial for energy production and metabolic processes. While Trifluralin and Pendimethalin demonstrated a less pronounced negative impact on mycelial growth, the study concluded that all tested herbicides were detrimental to Morchella sextelata. It is thus recommended that, in agricultural practice, the use of herbicides be approached with caution, and that application rates be strictly adhered to in order to ensure the healthy cultivation of Morchella sextelata and minimize potential pesticide residue issues.

Project description:True morels (Morchella spp., Morchellaceae, Ascomycota), a delicious edible mushroom, has rapidly expanded in recent years, especially in China. However, a severe disease of morels, red fruitbody disease, led to very low production of fruiting bodies. The cause reason and the mechanisms under red fruitbody are unclear. Herein, we integrated the transcriptomics and metabolomics data of M. sextelata from red fruitbody group (R) and normal group (N), which was artificial cultivation in Fujian province, China. Transcriptome data revealed the differentially expressed genes (DEGs) between R group and N group were significantly enriched in the pathways of tyrosine metabolism, riboflavin metabolism, and glycerophospholipid metabolism. Similarly, the differential accumulated metabolites (DAMs) were mainly assigned to metabolism categories, including tyrosine metabolism, biosynthesis of plant secondary metabolites, biosynthesis of amino acids, and others. Then, combined analysis of the transcriptome data and metabolome traits revealed that the most enriched pathway was tyrosine metabolism, followed by ABC transporters, alanine, aspartate and glutamate metabolism, and others. In summary, this integration of transcriptomics and metabolomics data of M. sextelata during fruitbody redness implicated several key genes, metabolites, and pathways involved in this disease. We believe that these findings will help us understand the mechanisms under fruitbody redness of M. sextelata and provide new clues for optimizing the methods for its cultivation application.

Project description:Morchella are macrofungi and are also called morels, as they exhibit a morel-like upper cap structure. Morels contain abundant essential amino acids, vitamins and biologically active compounds, which provide substantial health benefits. Approximately 80 species of Morchella have been reported, and even more species have been isolated. However, the lack of wild Morchella resources and the difficulties associated with culturing Morchella have caused a shortage in the morels available for daily consumption. Additionally, in-depth genomic and morphological studies are still needed. In this study, to provide genomic data for further investigations of culturing techniques and the biological functions of Morchella sextelata (M. sextelata), de novo genome sequencing was carried out on the Illumina HiSeq. 4000 platform using both the Illumina 150 and PacBio systems. The final estimated genome size of M. sextelata was 52.93 Mb, containing 59 contigs and a GC content of 47.37%. A total of 9,550 protein-coding genes were annotated. In addition, the repeat sequences, gene components and gene functions were analyzed using various databases. Furthermore, the secondary metabolite gene clusters and the predicted structures of their products were analyzed. Finally, a genomic comparison of different species of Morchella was performed.

Project description:In this project, we have performed a comparative analysis of the proteomes of the mycelium and cysts of P. capsici. The identified proteins were functionally classified, which provides the basis for future research on these developmental stages of P. capsici and closely related species. Many of the proteins enriched in either the mycelium or cysts were found to contribute specific functions in these two life stages. Proteins enriched in mycelium were essentially associated with vegetative growth, whereas those enriched in cysts were primarily related to development-specific processes associated with the initiation of infection. The data provide a better understanding of the metabolic pathways underlying the mycelial and cyst developmental stages of P. capsici and point to potential targets of anti-oomycete compounds at each of these stages.

Project description:Mushrooms of the Morchella genus exhibit a variety of biological activities. Two polysaccharides (MSP1-1, 389.0 kDa; MSP1-2, 23.4 kDa) were isolated from Morchella sextelata by subcritical water extraction and column chromatography fractionation. Methylation and nuclear magnetic resonance analysis determined MSP1-1 as a glucan with a backbone of (1→4)-α-D-glucan branched at O-6, and MSP1-2 as a galactomannan with coextracted α-glucan. Light scattering analysis and transmission electron microscopy revealed that MSP1-1 possessed a random coil chain and that MSP1-2 had a network chain. This is the first time that a network structure has been observed in a polysaccharide from M. sextelata. Despite the differences in their chemical structures and conformations, both MSP1-1 and MSP1-2 possessed good thermal stability and showed antioxidant activity. This study provides fundamental data on the structure-activity relationships of M. sextelata polysaccharides.
