Project description:High light (HL)-induced chloroplast retrograde signaling originates from the photosynthetic apparatus and regulates nuclear gene expression to enhance photoprotection and coordinate cell metabolism. Here, we analyzed the transcript profiles and accumulation of reactive oxygen species (ROS), stress hormones and small molecule antioxidants to investigate the signaling mechanisms operating under HL stress, and in particular during plant recovery (R) from stress under growth conditions. Exposure to HL for 15 min induced a number of singlet oxygen (1O2) and hydrogen peroxide (H2O2)-responsive genes and accumulation of an oxidative form of glutathione (GSSG) and ascorbate (DHA), the hallmarks of oxidative stress in cells. Prolonged exposure to HL resulted in accumulation of transcripts encoding oxylipin biosynthesis enzymes, leading to accumulation of 12-oxo-phytodienoic acid (OPDA) and jasmonic acid (JA). However, the expression of several JA-responsive genes, already induced by HL, peaked during the R-phase (R-sustained) together with accumulation of JA, reduced glutathione (GSH) and ascorbate (AsA), highlighting the critical role of JA signaling in restoring chloroplast redox balance following HL stress. The involvement of JA signaling in R-sustained gene expression was further confirmed by conducting experiments with JA receptor mutants (coi1). HL exposure of only 2 min was sufficient to induce some R-sustained genes, indicating rapid response of plants to changing light conditions. We propose that ROS production at HL induces the signaling cascade for early oxylipin biosynthesis and OPDA accumulation, while increased accumulation of JA in R-phase activates the genes that fully restore the glutathione metabolism, and ultimately allow recovery from short-term HL stress.

Project description:The stability of mRNA is an important determinant of its abundance and, consequently, protein production. There has been extensive research on the pathways governing mRNA stability and translation, however, it is unclear the extent to which these processes are modulated by environmental conditions. We previously modelled rapid recovery gene down-regulation (RRGD) following light stress in Arabidopsis thaliana (Arabidopsis) using mathematical calculations to account for transcription in order to predict half-lives and led to the hypothesis of recovery-specific transcript destabilisation. Here, we test this hypothesis by quantifying changes in transcription, mRNA stability, and translation in leaves of mature Arabidopsis undergoing light stress and recovery and investigate processes regulating transcript abundance and fate. Compared to juvenile plants from prior work, here we find that stability is altered for a range of transcripts that encode proteins involved in post-transcriptional processes in mature leaves. We also observe transcript destabilisation during light stress, followed by re-stabilisation upon recovery. Alongside this, we observe fast transcriptional shut-off in recovery that, when paired with transcript destabilisation, promotes rapid down-regulation of stress-induced genes. Translation was dynamic over the course of light stress and recovery, with substantial transcript-specific increases in polysome loading observed during late stress independently of total mRNA abundance. Taken together, we provide evidence for the combinatorial regulation of transcription, mRNA stability, and translation that occurs during light stress and recovery.

Project description:Environmental stimuli-triggered stomatal movement is a key physiological process that regulates CO<sub>2</sub> uptake and water loss in plants. Stomata are defined by pairs of guard cells that perceive and transduce external signals, leading to cellular volume changes and consequent stomatal aperture change. Within the visible light spectrum, red light induces stomatal opening in intact leaves. However, there has been debate regarding the extent to which red-light-induced stomatal opening arises from direct guard cell sensing of red light versus indirect responses as a result of red light influences on mesophyll photosynthesis. Here we identify conditions that result in red-light-stimulated stomatal opening in isolated epidermal peels and enlargement of protoplasts, firmly establishing a direct guard cell response to red light. We then employ metabolomics workflows utilizing gas chromatography mass spectrometry and liquid chromatography mass spectrometry for metabolite profiling and identification of Arabidopsis guard cell metabolic signatures in response to red light in the absence of the mesophyll. We quantified 223 metabolites in Arabidopsis guard cells, with 104 found to be red light responsive. These red-light-modulated metabolites participate in the tricarboxylic acid cycle, carbon balance, phytohormone biosynthesis and redox homeostasis. We next analyzed selected Arabidopsis mutants, and discovered that stomatal opening response to red light is correlated with a decrease in guard cell abscisic acid content and an increase in jasmonic acid content. The red-light-modulated guard cell metabolome reported here provides fundamental information concerning autonomous red light signaling pathways in guard cells.

It was found that UV light inhibited transcription elongation and that recovery of RNA synthesis occurred as a wave in the 5’-3’ direction with slow recovery and TC-NER at the 3’ end of long genes. RNA synthesis resumed fully at the 3’-end of genes after a 24-hour recovery in wild-type fibroblasts, but not in cells deficient in transcription-coupled nucleotide excision repair (TC-NER) or global genomic NER (GG-NER). Different transcription recovery profiles were found for individual genes but these differences did not fully correlate to differences in DNA repair of these genes. Our study gives the first genome-wide view of how UV-induced lesions affect transcription and how the recovery of RNA synthesis of large genes are particularly delayed by the apparent lack of resumption of transcription by arrested polymerases.

Project description:Seeds were germinated on 1/2 MS plates plus 1% sucrose and placed at 4°C for 3 days and then transferred to white light for 12 h before placed in blue light for another 4 days. Total RNA was extracted from samples including WT, agb1, cry1 cry2 and hy5 with RNAprep plant kit (TIANGEN), and followed by DNase I (Takara) treatment. This study reveals the molecular basis for light and G-protein crosstalk by analyzing gene expression profile under control of both signals.

Abstract:
Light/dark cycles are probably the most important environmental signals that regulate plant development. Light is essential for photosynthesis, but an excess, in combination with the unavoidable presence of atmospheric oxygen inside the chloroplast, leads to excessive reactive oxygen species production. Among the defense mechanisms that activate plants to cope with environmental stress situations, it is worth noting the ascorbate-glutathione cycle, a complex metabolic pathway in which a variety of photochemical, chemical and enzymatic steps are involved.We herein studied the dynamic behavior of this pathway under light/dark conditions and for several consecutive days. For this purpose, a mathematical model was developed including a variable electron source with a rate law proportional to the intensity of solar irradiance during the photoperiod, and which is continuously turned off at night and on again the next day. The model is defined by a nonlinear system of ordinary differential equations with an on/off time-dependent input, including a parameter to simulate the fact that the photoperiod length is not constant throughout the year, and which takes into account the particular experimental kinetics of each enzyme involved in the pathway. Unlike previous models, which have only provided steady-state solutions, the present model is able to simulate diurnal fluctuations in the metabolite concentrations, fluxes and enzymatic rates involved in the network.The obtained results are broadly consistent with experimental observations and highlight the key role played by ascorbate recycling for plants to adapt to their surrounding environment. This approach provides a new strategy to in vivo studies to analyze plant defense mechanisms against oxidative stress induced by external changes, which can also be extrapolated to other complex metabolic pathways to constitute a useful tool to the scientific community in general.
