Project description:Cancer therapy is often limited by toxicity from pneumonitis. This often-lethal side effect is known to be impacted by innate immunity, and in particular the pathways regulated by the TRAIL death receptor DR5. We investigated whether DR5 agonists could rescue mice from the lethal effects of radiation. We found that two different agonists, parenteral PEGylated trimeric-TRAIL (TLY012) and oral TRAIL-Inducing Compound #10 (TIC10/ONC201), could achieve this goal. Both compounds could completely protect mice from lethality by reducing pneumonitis, alveolar-wall thickness, and oxygen desaturation. Protection extended to the late effects of radiation including less fibrosis at 22-weeks in TLY012-rescued survivors versus un-rescued surviving irradiated-mice. Wild-type orthotopic breast tumor-bearing mice receiving 20-Gy thoracic radiation were protected from pneumonitis with disappearance of tumors. At the molecular level, this protection appeared to be due to the inhibition of CCl22, a macrophage-derived chemokine previously associated with radiation pneumonitis and pulmonary fibrosis. The discovery that short-term treatment with TRAIL pathway agonists effectively rescues animals from high doses of radiation exposure has important translational implications.

Analysis of mRNA expression between irradiated and non-irradiated keloid-derived primary cells.

Project description:This study aims to compare mRNA expression between irradiated and non-irradiated keloid-derived primary cells by RNA-Seq. In this study, keloid-derived primary cells that were isolated from patients, and then divided into three groups and each group had four repeats. The first group of cells was treated without radiation, the second group of cells were received 4 Gy delivered in 2-Gy fractions once per day over 2 days and the third group of cells were received 8 Gy delivered in 2-Gy fractions once per day over 4 days.The last two groups of cells were irradiated at a fixed rate of 1.15 Gy/min using an X-ray linear accelerator (Rad Source Technologies Inc., Suwanee, GA, USA). 24 hours post irradiation of the third group of cells, all keloid-derived primary cells were collected and subjected to mRNA expression analysis.

Project description:We used microarrays to measure the expression levels of genes in irradiated immortalized B cells, lymphoblastoid cells, from members of Centre dEtude du Polymorphisme Humain (CEPH) Utah pedigrees. Data were collected for cells at baseline and 2 hour and 6 hour after exposure to 10 Gy of ionizing radiation (IR). Experiment Overall Design: We used microarrays to measure the expression levels of genes in irradiated immortalized B cells, lymphoblastoid cells, from members of 15 Centre dEtude du Polymorphisme Humain (CEPH) Utah pedigrees (CEPH 1333, 1341, 1346, 1362, 1408, 1416, 1420, 1421, 1423, 1424, 1444, 1447, 1451, 1454, 1582). Expression data was obtained for cell lines derived from 2 parents and 8 children per each family. Cells were irradiated at 10 Gy in a 137Cs irradiator. Cells were harvested prior to radiation and at 2 and 6 hours following exposure to IR.

Project description:Exposure to ionizing radiation around the time of neural tube closure results in a number of developmental defects, including neural tube defects (e.g. exencephaly, cleft palate) and eye defects (e.g. microphtalmos, anaphtalmos). These defects result from perturbed neural tube closure and development of the presumptive eye. In order to better understand the underlying molecular mechanisms, gene expression profiling was performed in heads of mouse embryos at embryonic day 9, after they had been irradiated at embryonic day 7.5 with 1 Gy of X-rays. Non-irradiated mice served as controls for differential gene expression.
