Project description:This study investigates the epigenetic impact of Nicotinamide N-methyltransferase (NNMT) overexpression on human umbilical vein endothelial cells (HUVECs). HUVECs were transfected with adenovirus vectors, Ad-NC (negative control) or Ad-NNMT (overexpressing NNMT), for 48 hours. Subsequently, protein was collected for chromatin immunoprecipitation analysis using antibodies against H3K4me3 and H3K27me3 via Cut&Tag technology. To further elucidate the effects on DNA methylation, whole-genome methylation sequencing was performed. The resulting data provide insight into NNMT's regulatory role in histone modifications and methylation patterns.

Project description:The cellular heterogeneity within a tumor can be determined by core genetic and epigenetic programs that operate in certain cells (tumor initiating cells – TICs), providing them with the degree of plasticity needed for induction of metastasis and resistance to therapy. Here we show that the metabolic enzyme nicotinamide N-methyl transferase (NNMT) promotes TIC plasticity in Estrogen Receptor (ER) alpha negative breast cancer. NNMT downregulation delays tumor formation and its full depletion impairs metastasis formation in mice. Mechanistically, NNMT loss increases deposition of H3K9-me2/3 and H3K27-me3 at the promoter of genes involved in stem cell regulation, shutting down their expression and upregulating luminal differentiation genes. This study reveals a major function of NNMT in maintaining core epigenetic programmes that promote TIC self-renewal and metastasis and that repress luminal differentiation.

We therefore investigated the role of SREBP2 in EC function through comparing the transciptional profiling of human umbilical vein endothelial cells (HUVECs) that were infected with adenovirus overexpressing SREBP2 (Ad-SREBP2) or with empty vector (Ad-null) control. Gene ontology analysis revealed that SREBP2 not only regulates cholesterol biosynthesis, but also mediates the TGF, WNT, and cytoskeleton remodeling pathways. Among the responsive genes, SREBP2 exhibited the induction of mesenchymal transition.

Microarray analysis of NNMT overexpression and knockdown in stromal cells

Transcriptome Data of KLF4 Overexpression in Human Umbilical Vein Endothelial Cells

Project description:Atheroprotective flow (e.g., pulsatile shear stress) and statins drastically induce the expression of krüppel-like factor 4 (KLF4) in vascular endothelial cells (ECs). We therefore investigated the role of KLF4 in EC function through comparing the transciptional profiling of human umbilical vein endothelial cells (HUVECs) that were infected with adenovirus overexpression KLF4 (Ad-KLF4) or with empty vector (Ad-null) control. Gene ontology analysis revealed that KLF4 not only regulates EC homeostasis through the upregulation of nitric oxide synthesis and vascular development, but also mediates response to lipid. Among the lipid responsive genes, KLF4 exhibited induction of cholesterol efflux and oxidation [i.e., liver X receptor (LXR) and cholesterol 25-hydroxylase (Ch25h)], while suppressing cholesterol biosynthesis [i.e., sterol regulatory element-binding protein 2 (SREBP2)].

Optimizing transfection of primary human umbilical vein endothelial cells using commercially available chemical transfection reagents.

Project description:Primary cells, such as HUVEC, are notoriously difficult to transfect and are susceptible to the toxic effects of transfection reagents. A transfection reagent with a high transfection efficiency and low cytotoxicity was sought to retain sufficient viability of transfected HUVEC for subsequent assays. Nine chemical transfection reagents, currently commercially available, were compared for their ability to transfect HUVEC in vitro. A plasmid expressing the enhanced GFP (EGFP) was used for transfection, followed by flow cytometry of transfected HUVEC to determine the proportion of EGFP-expressing cells as a measure of transfection efficiency. Lipofectamine 2000 and Lipofectamine LTX (Invitrogen, Carlsbad, CA, USA) gave the highest transfection efficiencies of the reagents tested. Lipofectamine LTX was identified as the optimal transfection reagent as a result of its higher transfection efficiency at shorter periods of time following transfection when cytotoxicity was limited, allowing sufficient yield of transfected HUVEC for use in subsequent assays.

Proteomics reveals NNMT as a master metabolic regulator of cancer associated fibroblasts

Project description:High-grade serous carcinoma has a poor prognosis, owing primarily to its early dissemination throughout the abdominal cavity. Genomic and proteomic approaches have provided snapshots of the proteogenomics of ovarian cancer, but a systematic examination of both the tumour and stromal compartments is critical in understanding ovarian cancer metastasis. We developed a label- free proteomic workflow to analyse as few as 5,000 formalin-fixed, paraffin-embedded cells microdissected from each compartment. The tumour proteome was stable during progression from in situ lesions to metastatic disease; however, the metastasis-associated stroma was characterized by a highly conserved proteomic signature, prominently including the methyltransferase nicotinamide N-methyltransferase (NNMT) and several of the proteins that it regulates. Stromal NNMT expression was necessary and sufficient for functional aspects of the cancer-associated fibroblast (CAF) phenotype, including the expression of CAF markers and the secretion of cytokines and oncogenic extracellular matrix. Stromal NNMT expression supported ovarian cancer migration, proliferation and in vivo growth and metastasis. Expression of NNMT in CAFs led to depletion of S-adenosyl methionine and reduction in histone methylation associated with widespread gene- expression changes in the tumour stroma. This work supports the use of ultra-low-input proteomics to identify candidate drivers of disease phenotypes. NNMT is a central, metabolic regulator of CAF differentiation and cancer progression in the stroma that may be therapeutically targeted.
