Project description:Arbuscular mycorrhizal symbiosis is a predominant relationship between plant and arbuscular mycorrhizal fungi. To idendify arbuscular mycorrhiza responsive miRNAs, small RNA libraries were constructed in tomato roots colonized with Rhizophagus irregularis and without Rhizophagus irregularis. We identify miRNAs in tomato roots and provide a new profile of tomato miRNAs. And we found that some miRNAs were responsive to arbuscular mycorrhiza by comparing miRNAs in treatment with that in control.

From root to fruit: RNA-Seq analysis reveals that arbuscular mycorrhizal symbiosis may affect the tomato fruit metabolism

Project description:The crop species Solanum lycopersicum establishes a beneficial root- symbiosis with the widespread group of arbuscular mycorrhizal (AM) fungi. The mycorrhiza establishment leads to a modulation of the plant gene expression which is not restricted to the root compartment but spreads at the organism-wide level. To understand the systemic effect of the fungal presence on the tomato fruit, we performed global transcriptome profiling through RNA-Seq analysis on Moneymaker tomato fruits sampled at the turning ripening stage. Gene expression data were obtained from fruits sampled at 55 days after flowering. Fruits were collected from Funneliformis mosseae colonized plants and from control plants which were fertilized in order to avoid responses related to nutrient deficiency.

Effect of Penicillium chrysogenum, Penicillium minioluteum, and Serendipita indica on gene expression in Solanum lycopersicum plants under control and drought stress conditions

Project description:Plants coexist in close proximity with numerous microorganisms in their rhizosphere. With certain microorganisms, plants establish mutualistic relationships that can confer physiological benefits to the interacting organisms, including enhanced nutrient assimilation or increased stress tolerance. The root-colonizing endophytic fungi Penicillium chrysogenum, Penicillium minioluteum, and Serendipita indica have been reported to enhance the drought stress tolerance of plants. However, to date, the molecular mechanisms triggered by these fungi in plants remain unexplored. This study presents a comparative analysis of the effects on mock- and fungus-infected tomato plants (var. Moneymaker) under drought stress conditions (40% field capacity) and control conditions (100% field capacity). The findings provide evidence for the induction of common response modules by the fungi.

Effect of Mac1 deletion on copper homoestasis and virulence in Fusarium oxysporum f. sp. lycopersici [RNA-seq]

Project description:The transcription factor Mac1 is a key regulator of copper homeostasis, controlling the transcriptional response to copper-limiting conditions in fungi. It has been described as a virulence factor in human pathogens such as Candida albicans and Aspergillus fumigatus, but surprisingly its role in plant pathogenesis is poorly understood. Targeted deletion of Mac1 in the soilborne pathogen Fusarium oxisporum results in an inability to grow under copper-limited conditions and kills tomato plants. The aim of this RNA-seq analysis is to elucidate the role of Mac1 in gene regulation under copper starvation and during infection of tomato plants by F. oxysporum.

Effect of Fusarium sp. strain K-23-infection on the gene expression in Arabidopsis thaliana plants under control and salt stress conditions

Project description:Several fungi have been reported to form intimate bonds with plants. These close mutual relationships can provide physiological benefits to the interacting organisms, including the improvement of nutrient assimilation or enhanced stress tolerance. The root-colonizing fungus Fusarium sp. strain K-23 is known to successfully colonize tomato roots. Previous studies highlighted the promotion of plant biomass production and stress tolerance of tomato plants infected with the K-23. However, up to data, nothing is known about the effect of this fungus on the model plant Arabidopsis thaliana. We here report the comparative analysis of the effect of mock- and K-23-infected wild-type Arabidopsis plants (Col-0) in the abscence and presence of moderate salt stress conditions (50 mM NaCl). Our data provide evidence for a profound effect of K-23 on root hair elongation through the activation of a regulatory network that includes several plant hormones. Intriguingly, the fungus suppresses gibberellic acid biosynthesis which, in turn, promotes root hair elongation.

Proteomic profiling and alteration in Tuta absoluta upon infection with entomopathogenic fungi, Beauveria bassiana and Purpureocillium lilacinum

Project description:Tomato leafminer (Tuta absoluta) is a major pest of solanaceous plant species and causes heavy economic losses to the tomato crop. T. absoluta have been developed resistance to many chemical insecticides and have serious impacts on human health as well as environment. To overcome the ill effects of insecticides, ecofriendly approachis needed and among various biological microbes, entomopathogenic fungi is reliable and cost effective. The entomopathogenic fungi, like Beauveria bassiana and Purpureocillium lilacinum have ability to control pest population. The present study was conducted to study the interaction of B. bassiana and P. lilacinum at proteomic level to understand post infection effects. The analysis showed that in fourth instar larvae of T. absoluta, and the 442 and 423 proteins were significantly dysregulated upon infection with P. lilacinum and B. bassiana respectively. The present study concluded entomopathogenic fungal infection decline the immune system of T. absoluta larvae and can be adopted in for control of T. absoluta insect pest.

A pathogen-responsive gene cluster for the production of highly modified fatty acids in tomato

Project description:In response to biotic stress, plants produce suites of highly modified fatty acids that bear unusual chemical functionality. Despite their chemical complexity and proposed roles in pathogen defense, little is known about the biosynthesis of these decorated fatty acids in plants. Falcarindiol is a prototypical acetylenic lipid present in carrot, tomato, and celery that inhibits growth of fungi and human cancer cell lines. Using a combination of untargeted metabolomics and RNA sequencing, we discovered a biosynthetic gene cluster in tomato (Solanum lycopersicum) that is required for the production of falcarindiol in response to an adapted fungal pathogen, Cladosporium fulvum. By reconstituting the initial biosynthetic steps in a heterologous host (Nicotiana benthamiana) and generating transgenic pathway mutants in tomato, we demonstrate a direct role for three genes in the cluster in falcarindiol biosynthesis. This work reveals a mechanism by which plants sculpt their lipid pool in response to pathogens, and provides critical insight into the complex biochemistry of alkynyl lipid production.
