Project description:Tellurium (Te) is a metalloid that is growing as a challenge since its increasing environmental presence, as a result of mining activity and improper disposal of high-tech devices, leads to an increasing selective pressure against living organisms. How microorganims adapt to the presence of increasing concentrations of metalloids, such as Te, could increase the know-how to solve Te contamination problems. Two mining isolates, Bacillus sp. and Paenibacillus sp, were able to grow in high concentrations of Te(IV) and were able to reduce Te(IV) to Te(0) so the effects of growth in the presence of Te(IV) was determined, at the level of total protein expression, for this strains. It was determined that growth of these strains in the presence of Te(IV) affected the general protein expression pattern, and hence the metabolism.
Project description:A comparative genomic approach was used to identify large sequence polymorphisms among Mycobacterium avium isolates obtained from a variety of host species. DNA microarrays were used as a platform for comparing mycobacteria field isolates with the sequenced bovine isolate Mycobacterium avium subsp. paratuberculosis (Map) K10. ORFs were classified as present or divergent based on the relative fluorescent intensities of the experimental samples compared to Map K10 DNA. Map isolates cultured from cattle, bison, sheep, goat, avian, and human sources were hybridized to the Map microarray. Three large deletions were observed in the genomes of four Map isolates obtained from sheep and four clusters of ORFs homologous to sequences in the Mycobacterium avium subsp. avium (Maa) 104 genome were identified as being present in these isolates. One of these clusters encodes glycopeptidolipid biosynthesis enzymes. One of the Map sheep isolates had a genome profile similar to a group of Mycobacterium avium subsp. silvaticum (Mas) isolates which included four independent laboratory stocks of the organism traditionally identified as Maa strain 18. Genome diversity in Map appears to be mostly restricted to large sequence polymorphisms that are often associated with mobile genetic elements. Keywords: Comparative genomic hybridization
Project description:The pyrite mine of Lousal, Grândola, Portugal, was discovered in 1882 and extensively exploited from 1900 until its closure in 1988 when the mining ores were not viable economically. Aiming at becoming a cultural-touristic center, the mining area was rehabilitated raising a Mining Museum and a Science Centre along with the reopening of a mining gallery for public access. In recent times, aerobiological studies on subterranean environments such as caves, mines and archeological necropolis revealed to be an important source of novel bacteria, from which their biotechnological potential are under study. Here we present a Pseudomonas sp. isolated from the mine air with potential secondary metabolite biosynthesis, resistance to antibiotics and virulence factors.

Project description:Neisseria gonorrhoeae (GC) is a human-specific pathogen, and the agent of a sexually transmitted disease, gonorrhea. There is a critical need for new approaches to study and treat GC infections because of the growing threat of multidrug-resistant isolates and the lack of a vaccine. Despite the implied role of the GC cell envelope and membrane vesicles in colonization and infection of human tissues and cell lines, comprehensive studies have not been undertaken to elucidate their constituents. Accordingly, in pursuit of novel molecular therapeutic targets, we have applied isobaric tagging for absolute quantification coupled with liquid chromatography and mass spectrometry for proteome quantitative analyses. Mining the proteome of cell envelopes and native membrane vesicles revealed 533 and 168 common proteins, respectively, in analyzed GC strains FA1090, F62, MS11, and 1291.
Project description:The aim of the study was to carry out a CGH study utilizing a set of 39 diverse Bacillus isolates. Thirty four B. cereus and five B. anthracis strains and isolates were chosen so as to represent different lineages based on previous characterizations, including MLEE and MLST (Helgason, Okstad et al. 2000; Helgason, Tourasse et al. 2004). They represent the spectrum of B. cereus phenotypic diversity by including soil, dairy and periodontal isolates in addition to virulent B. anthracis strains.
