Gene Expression Patterns in the Preoptic Area of Mice with Defective FGF Signaling in GnRH Neurons

Project description:Sexual interactions have previously been shown to improve reproductive health through unknown mechanisms. In this study, we used RNA-Seq to examine sex-induced gene expression changes in the preoptic area (POA), a critical reproductive brain region. Using a mouse model defective in fibroblast growth factor signaling (dnFGFR mouse), previously shown to disrupt the gonadotropin-releasing hormone (GnRH) system, we examined the impact of opposite sex (OS) housing on gene expression in the POA of a reproductively compromised animal. Bulk RNA-Seq followed by gene set enrichment analysis (GSEA) were used to analyze changes in gene expression and biological processes in control and dnFGFR mice after 300 days of cohabitation with a same sex or OS partner. OS housing of dnFGFR mice, but not control mice, significantly improved reproductive anatomy and gonadotropins in dnFGFR mice. These changes occurred concomitantly with novel biological processes related to estradiol metabolism and neuron excitation. Our results suggest a new role of neuron- or astrocyte-derived estradiol in the plasticity of the GnRH neuron population and offer a promising new direction for the treatment of reproductive disorders stemming from GnRH deficiency.

Prenatal Androgenization Alters the Development of GnRH Neuron and Preoptic Area RNA Transcripts in Female Mice (II)

Project description:Translating Ribosome Affinity Purification results in non specific RNA binding to the immunoprecipitation matrix, even in mice not expressing cell specific GFP labelled ribosomal protein L10a. This creates false positive enriched transcripts in our datasets. To determine which RNA transcripts bind non specifically to the immunoprecipitation matrix, preoptic area punches were collected from young and adult mice that do not express L10a-GFP. Also, included was a positive control (GnRH CRE x floxed L10a-GFP mice) to ensure that the TRAP procedure was enriching for GnRH neuron specific transcripts as previously described. Four GnRH depleted supernatant RNA samples from the previous study were also included to aid comparison of the two sequencing runs.

Project description:The effect of photoperiod on gene expression was assessed in punch samples of the bed nucleus of the stria terminalis and medial preoptic area. Long day mice were housed in 16L:8D and short day mice were housed in 8L:16D. Experiment Overall Design: For the medial preoptic area, independent samples were collected from long day (n=3) and short day (n=3) mice. For the bed nucleus of the stria terminalis independent samples were collected from long day (n=3) and short day (n=2) mice.

RNA-sequencing of preoptic area leptin receptor-expressing neurons from heat acclimated LepRCreHTB mice

Project description:This experiment intended to look for the transcriptome changes that happen in neuronal cells from the preoptic area of the hypothalamus expressing leptin receptors during heat acclimation conditions. 3 groups of animals were used; long-term heat-acclimated animals constantly exposed to 36˚C for 28-35 days, short-term heat-acclimated animals constantly exposed to 36˚C for 5 days, and non-habituated (non-acclimated) animals which were not exposed to heat. The animal line used was LepRCreHTB.
