Project description:Doxorubicin is a widely used anti-cancer drug, and its toxicological effects on healthy tissues, especially the heart, remains challenging in therapy. To explore the potential targets that ameliorate doxorubicin-induced cardiotoxicity (DIC), we aimed to investigate the role that macrophages play in the DIC development. Senescence of RAW264.7 cells was induced through doxorubicin administration in the experiment, and the transcription of secretory components was analyzed in this study.

Project description:Epithelial senescence, specifically in the bronchiolized regions of the fibrotic lung, is apparent in IPF but not control tissue. In order to better understand the phenotype of these senescent cells we generated a in vitro culture model of senescence utillizing primary normal human bronchial epithelial cells. Cultures were treated with doxorubicin to induce senescence and the transcriptional phenotype of the senescent cultures was compared to DMSO-treated control cultures.

Project description:Oncogene-induced senescence provides a barrier against malignant transformation. Senescent cells secrete pro-inflammatory cytokines, chemokines and growth factors collectively known as the senescence-associated secretory phenotype (SASP). Paradoxically, the SASP can also negatively impact the neighbouring tissues through inducing an inflammatory environment that promotes tumorigenesis and aged-related pathologies. We have previously shown that the lncRNA MIR31HG is overexpressed and located in the cytoplasm during BRAF-induced senescence. In young proliferating cells, nuclear MIR31HG inhibits p16/CDKN2A expression through interaction with polycomb repressor complexes (PRC1/2). Here, we show that MIR31HG regulates the expression and secretion of a subset of SASP components during BRAF-induced senescence. The SASP secreted from senescent cells depleted for MIR31HG fails to induce paracrine invasion without affecting the growth inhibitory effect. Mechanistically, MIR31HG interacts with the translation factor YBX1 facilitating its phosphorylation at serine 102 (p-YBX1S102) by the kinase RSK. p-YBX1S102 induces IL1A translation which acts as upstream regulator inducing the transcription of the other SASP mRNAs. Our results suggest a dual role for MIR31HG in senescence depending on its cellular localization and place the lncRNA as a potential therapeutic target in the treatment of senescence-related pathologies.

Project description:Here, we investigate therapy-induced senescence (TIS) as a reversible mechanism of drug resistance in breast cancer cells. High-dose doxorubicin treatment was used to induce TIS in four distinct breast cancer cell lines and the drug resistance/sensitivity pattern of parental and TIS cells were investigated using a panel of FDA-approved anticancer molecules.
Proteome analysis confirmed the presence of the Senescence-Associated Secretory Phenotype (SASP), altered spliceosomal activity and proteins with significant role in immune evasion.

Project description:The senescence-associated secretory phenotype (SASP) has recently emerged as both a driver of -- and promising therapeutic target for -- a multitude of chronic age-related conditions, ranging from neurodegeneration to cancer. The complexity of the SASP, typically monitored by the secretion of a few proteins, has been greatly underappreciated, and a small set of factors cannot explain the diverse phenotypes it produces in vivo. Here, we present SASP Atlas, a comprehensive proteomic database of SASPs driven by multiple inducers of senescence in different human cell types. While there are common elements among all the SASPs we have documented so far, we discovered distinct senescence inducer- and cell type-specific secretory proteins. In all cases, the SASPs are comprised of hundreds of unique proteins secreted as both soluble proteins (sSASP) and exosomes (eSASP) with distinct and novel protein profiles. This resource will aid in identifying the proteins that drive senescence-associated phenotypes and provide comprehensive catalogs of potential senescence biomarkers that can be used to assess the burden and the originating stimuli and type of senescent cells in vivo.

Project description:Cells are subjected to dynamic mechanical environments which impart various forces and induce cellular responses. In age-related conditions like pulmonary fibrosis, there is both an increase in tissue stiffness and an accumulation of senescent cells, leading to elevated tension in fibroblasts among other cells. While senescent cells produce a senescence-associated secretory phenotype (SASP), the impact of physical stimuli on both cellular senescence and SASP is not well understood. Here, we show that mechanical tension, modeled using cell culture substrate rigidity, influences senescent cell markers like SA-beta-gal and secretory phenotypes. Comparing human primary pulmonary fibroblasts (IMR-90) cultured on physiological (2 kPa), fibrotic (50 kPa), and plastic (3 GPa) substrates followed by senescence induction using doxorubicin, we identified unique high-stiffness-driven secretory protein profiles using mass spectrometry and transcriptomic signatures, both showing an enrichment in collagen proteins. Computational meta-analysis of human interstitial lung disease single-cell RNA sequencing datasets confirmed these genes are highly expressed in disease samples and strongly correlate with mechanotransduction and senescence-related pathways. Thus, mechanical forces shape cell senescence and their secretory phenotypes.
