Project description:Sequencing of the Aedes aegypti genome has enabled genome-wide studies of gene expression in this mosquito. The large quantities of data produced from such studies require efficient cataloguing in order for new insight to be made into gene expression patterns and the underlying molecular mechanisms for producing these patterns. Our study provides a comprehensive catalogue of genes whose transcription products increase or decrease in abundance in adult females following blood feeding. We developed a publicly-accessible database and data-mining tool, aeGEPUCI, that integrates 1) stage-specific microarray analyses of gene expression in Ae. aegypti, 2) functional gene annotation, 3) genomic sequence data, and 4) computational sequence analysis tools. Project description:Investigation of whole genome gene expression level changes during a 44 hour time period in Aedes aegypti observed under light/dark and constant dark conditions. This is a 12-plex high defination NimbleGen array design. The Higgs White Eye (Wh) strain of Aedes aegypti was investigated for rhythmic expression during the light/dark phase and constant darkness. Samples were collected every 4 hours for 44 hours in each treatment, and RNA collected from whole head tissue.
Project description:We conducted a whole transcriptome analysis of testes from a meiotic drive-carrying strain (T37) in comparison with a drive-sensitive strain (RED) using microarrays based on the complete annotated Ae. aegypti gene set. The T37 strain, which carries a strong meiotic drive gene (Mori et al., 2004 (PMID 15605641)), was established from mosquitoes collected in Trinidad. The RED strain is highly sensitive to the meiotic drive gene (Hickey and Craig, 1966 (PMID ); Mori et al., 2004 (PMID 15605641)). A six-chip study using total RNA recovered from three biological samples of the T37 strain and another three biological samples of the Red strain of Aedes aegypti. Each chip measures the expression level of 16,092 genes annotated from the Aedes aegypti genome sequence, with twenty 60-mer probe pairs (PM/MM) per gene, with three-fold technical redundancy.
