Project description:We performed affymetrix gene expression profiling on mammary tumors from eight well-characterized genetically engineered Mouse (GEM) models of human breast cancer. The gene expression data will be combined with the miRNA gene expression data from the corresponding mammary tumors and tissues for integrated miRNA and mRNA gene expression analysis, which are useful in improving the identification of miRNA targets from potential targets identified in silico. mRNA expression data for 41 mouse primary mammary tumors and 5 mouse normal mammary glands

Project description:We report a mouse model that recapitulates expression of the ETV6-NTRK3 (EN) fusion oncoprotein, the product of the t(12;15)(p13;q25) translocation characteristic of human secretory breast carcinoma. Activation of EN expression in mammary tissues by Whey acidic protein (Wap) promoter-driven Cre leads to fully penetrant, multifocal malignant breast cancer with short latency. We provide genetic evidence that committed bipotent or CD61+ luminal alveolar progenitors, are targets of tumorigenesis. Furthermore, EN transforms these otherwise transient progenitors through activation of the AP1 complex. Given increasing relevance of chromosomal translocations in epithelial cancers, such mice serve as a paradigm for the study of their genetic pathogenesis and cellular origins, and generation of novel preclinical models. In this study, we generated microarray expression profiling data from both genetically marked, FACS-sorted tumor epithelial cells and from unfractionated mammary tumors and normal mammary glands. By using gene set enrichment analysis (GSEA) on these data, we have identified the AP1 transcriptional complex as the major downstream effector of the ETV6-NTRK3 signaling. Experiment Overall Design: Take the advantage of the Cre-lox system and a floxed lacZ reporter at the Rosa26 locus (Rosa-Stop-lacZ), we genetically marked mammary epithelial cells in which the conditional Etv6-NTRK3 knockin allele were turned on, and followed them to the hyperplasia and tumor stages. We then sorted lacZ+ hyperplastic mammary epithelial cells and lacZ+ tumor epithelial cells and collected their expression profiles by Affymetrix mouse whole genome MOE430.2 chips. We also collected expression profiles from unfractionated mammary tumors (unsorted tumors) derived from the same mouse model and mammary glands from normal mice.
Project description:Mouse mammary tumors are diverse neoplastic growths originating in the mammary glands of mice and serve as essential models for studying human breast cancer. These tumors can be classified into distinct molecular subtypes, including luminal, basal-like, HER2-enriched, and claudin-low, reflecting the heterogeneity observed in breast cancer. With unique genetic and molecular features, these subtypes are instrumental for understanding the complexities of tumor biology. Here we sequenced the mRNA of 13 treatment-na ve mouse mammary tumor lines for a total of 82 samples.
Project description:Adenoid cystic carcinoma (ACC) is an uncommon malignant neoplasm (incidence of 0.35 per 100,000) that occurs in different body sites. It most often arises in salivary glands and other secretory glands in the head and neck region. To find the gene expression signatures of ACC, we performed microarray experiments to compare ACC xenograft tumor models and normal tissue samples. We used microarrays to detail the global program of gene expression underlying adenoid cystic carcinoma and identified distinct classes of up- and down-regulated genes compared to non-neoplastic salivary tissue. Viable tumor tissue samples from human patients were used to establish xenograft tumor models in nude/nude immunodeficient mice. Tissue from these tumor models were compared to tissue samples of normal human salivary glands. Tissue samples were frozen, examined by cryostat histologic sectioning and macrodissected to obtain samples that represented at least 70% epithelial cells.
