To dissect cellular responses toward heat stress in the plant pathogenic fungus F. graminearum, we compared transcriptomes of the fungal cultures incubated in normal temperature condition (25 ÂºC) and in high temperature condition (37 ÂºC) for 15 min. 6 samples examined: 24 h-old mycelia from complete medium (CM) of Fusarium graminearum wild-type Z-3639 were incubated in normal temperature condition (25 ÂºC) and in high temperature condition (37 ÂºC) for 15 min.

Proteome of extracellular vesicles from the filamentous fungal pathogen Fusarium graminearum

Project description:We performed a comparative study to determine the proteome of extracellular vesicles (EVs) from the cereal pathogen Fusarium graminearum (Fgr), recovered from in vitro cultures. Label-free quantitative proteomics was used to find significant enrichment of proteins between EV samples, the secretome (secreted-soluble proteins) and the cell lysate. Our results show that some proteins were exclusive to EVs and were upregulated compared to the secretome or cell lysate.

Project description:Fusarium graminearum (teleomorph Gibberella zeae) is a prominent pathogen that infects major cereal crops, such as wheat, barley, and maize. Conidiogenesis had been intensively studied in Aspergillus nidulans and regulatory pathway genes have been known to regulate conidiogenesis in stage specific manner. We reported the functional analyses of flbD, abaA, and wetA orthologs in F. graminearum. To understand genome-wide transcriptional profiling of conidiation, we employed RNA-seq of the wild-type Fusarium graminearum Z-3639 and each gene deletion mutants with three time courses (0 h, 6 h and 12 h after induction of conidiogenesis). Project description:Fusarium head blight (FHB) is a major disease of cereal crops caused by the fungus Fusarium graminearum (Fg). FHB affects the flowering heads (or spikes) and developing seeds. This study compare the gene expression profile in wheat spikelets (spk 2) inoculated with either water (mock treatment) or a pathogenic strain of Fusarium graminearum (WT); spikelets 2 were inoculated 24 hrs after a neighbour spikelet (spk 0) was treated with either water or F. graminerum mutant strain Tri6Δ or NoxABΔ. Spikelets 2 were sampled 8 and 24 hrs after the second treatment.
