Project description:Cotton (Gossypium hirsutum L.) is an important economic crop, used mainly for the production of textile fiber. Using a space mutation breeding technique, a novel photosensitive genetic male sterile mutant CCRI9106 was isolated from the wild-type upland cotton cultivar CCRI040029. To use CCRI9106 in cotton hybrid breeding, it is of great importance to study the molecular mechanisms of its male sterility. Here, histological and iTRAQ-facilitated proteomic analyses of anthers were performed to explore male sterility mechanisms of the mutant. Scanning and transmission electron microscopy of the anthers showed that the development of pollen wall in CCRI9106 was severely defective with a lack of exine formation. At the protein level, 6,121 high-confidence proteins were identified and 365 of them showed differential expression patterns between mutant and wild-type anthers. The proteins up- or down-regulated in MT anthers were mainly involved in exine formation, protein degradation, calcium ion binding and etc. These findings provide valuable information on the proteins involved in anther and pollen development, and contribute to elucidate the mechanism of male sterility in upland cotton.

Project description:Cotton (Gossypium spp.) is one of the most important economic crops and exhibits yield-improving heterosis in specific hybrid combinations. The genic male-sterility system is the main strategy used for producing heterosis in cotton. To better understand the mechanisms of male sterility in cotton, we carried out label-free quantitative proteomics analysis in the anthers of two near-isogenic lines, the male-sterile line 1355A and the male-fertile line 1355B, the two isogenic lines. These experiments, along with bioinformatics analyses, identified 124 proteins from the anthers at uninucleate pollen stage (stage 8) that were significantly differentially expressed between these two lines.

Project description:We studied the causes of sterility underlying multiple-allele-inherited male sterility in Chinese cabbage by identifying differentially expressed genes (DEGs) related to pollen sterility between fertile and sterile flower buds. In this work,we performed transcriptome analysis of mRNA isolated from fertile and sterile buds using Illumina HiSeq 2000 platform sequencing. Approximately 80% of ~229 million high quality paired-end reads were uniquely mapped to the reference genome. In sterile buds, 699 genes were significantly up-regulated and 4,096 genes were down-regulated. Among the DEGs, 28 pollen cell wall-related genes, 53 transcription factor genes, 45 phytohormone-related genes, 20 anther and pollen-related genes, 212 specifically expressed transcripts (SETs), and 417 DEGs located in linkage group R07 were identified. Six transcription factor genes BrAMS, BrMS1, BrbHLH089, BrbHLH091, BrAtMYB103, and BrANAC025, were identified as putative sterility-related genes. The weak auxin signal that is regulated by BrABP1 may be one of the key factors causing pollen sterility observed here. Moreover, several significantly enriched GO terms “cell wall organization or biogenesis” (GO:0071554), “intrinsic to membrane” (GO:0031224), “integral to membrane” (GO:0016021), “hydrolase activity, acting on ester bonds” (GO:0016788) and one significantly enriched pathway “starch and sucrose metabolism” (ath00500) were obtained in this work. qRT-PCR and in situ hybridization validated that our RNA-seq transcriptome analysis was accurate and reliable. This study will lay the foundation for elucidating the molecular mechanism(s) underlying sterility, and provide valuable information for studying multiple-allele-inherited male sterility in Chinese cabbage line ‘AB01’.

Project description:We identified the nup1 mutant rice that exhibited specific male sterility due to the absence of Ubisch body formation and pollen grain production. We cloned the causal gene and demonstrated that NUP1, a class III peroxidase predominantly expressed in the anther wall, mediated ROS scavenging. The loss-of-function of NUP1 resulted in ROS burst in anthers, which disrupted the oxidation-reduction process and carbohydrate and lipid metabolisms, and thereby led to loss of tapetal Ubisch bodies and ultimately resulted in male sterility in rice. Our study expands the understanding of the molecular mechanisms by which ROS homeostasis regulates cell metabolism, tapetal Ubisch body formation and male fertility in plants.

Project description:To reveal the possible molecular mechanism underpinning the male sterility induced by a sulfonyflurea herbicide tribenuron-methyl (TBM), a comparative transcriptome analysis between rapeseed plants exposed to a TBM solution of 0.2 mg/L concentration and the control (spray with water) was conducted by using Illumina digital gene expression tag profiling technology (DGE). A transcriptomic analysis of the flower buds of TBM treatment identified 208 up-regulated and 164 down-regulated differential expression genes in comparison with the control. It was showed by the genetic network that the main groups of GO terms including response to chemical and hormone stimulus (response to stress), developmental process (pollen wall and pollen exine formation), lipid metabolism, and cellular amino acid derivative biosynthesis process were significantly influenced by the TBM treatment. The cytological and transcriptomic alterations in the treated plants were in accordance with reduction of chlorophyll, photosynthetic rate, and elevation of ethylene release.
