Project description:The V. dahliae could cause host obvious disease symptoms. Moreover, it was difficult to control, because of the rapidly genetic variation. Due to the rapidly genetic variation and complexity of the genetic background, the mechanism of pathogenicity differentiation of V. dahliae has still been unknown. To solve the problem, we isolated 24 strains of V. dahliae from a farmland of Shihezi city, Xinjiang province. Through ITS sequences and VCGs assay, the strains of SHZ-4, SHZ-5, and SHZ-9 had closer kinship. Moreover, the large difference was presented on the pathogenicity of SHZ-4, SHZ-5, and SHZ-9. Furthermore, a number of differentially expressed genes was identified through comparative transcriptome sequencing method and the genes were mainly centered on CWDEs and transport relative proteins. It was well understand that the CWDEs determined the source of nutrients of V. dahliae and the organic substance of host cell degraded by the CWDEs would induce the hyperosmosis of intercellular liquid. According to these, we proposed a hypothesis that the hyperosmosis induced the dehydration and death of host cell and contributed to absorb the nutrition for V. dahliae. The hyperosmosis promoting the nutrients scramble indicated that the V. dahliae could resist the hyperosmosis and the pathogenicity was positive associating with the tolerance. With regards to this, the sugar-induced-hyperosmotic resistance of the three V. dahliae isolates and cotton leaf tissue were analyzed through changing the sugar concentration. The result was entirely consistent with our hypothesis. It was firstly found that the pathogenicity of V. dahliae was relative to the resistance of sugar-induced-hyperosmosis, which could provide a new idea for cultivating a new varieties to resistant to the V. dahliae.

Project description:Verticillium dahliae is a soil-borne vascular pathogen that causes severe wilt symptoms in a wide range of plants. Co-culture of the fungus with Arabidopsis roots for 24 hours induces many changes in the gene expression profiles of both partners, even before defense-related phytohormone levels are induced in the plant. Both partners reprogram sugar and amino acid metabolism, activate genes for signal perception and transduction, and induce defense and stress responsive genes. Furthermore, analysis of Arabidopsis expression profiles suggests a redirection from growth to defense. The plant and fungal genes that rapidly respond to the presence of the partner might be crucial for early recognition steps and the future development of the interaction. Thus they are potential targets for the control of V. dahliae-induced wilt diseases.

Project description:Transcriptome sequencing is a powerful approach to globally delineate both the transcriptional and post-transcriptional genome regulation in human and other higher eukaryotes. This study sequenced the transcriptomes of two pathogenicity-differential strains of the plant wilt pathogen Verticillium dahliae. Although they showed no growth difference in vitro, hundreds of V. dahliae genes including those synthesizing aflatoxin were preferentially expressed at in the high-virulence strain. Using both the Pfam and GO annotation strategies, some of these putative virulence genes were ambiguously clustered into several known pathogenic mechanisms including hydrophobins secreted from fungal cells and acting as phytotoxin, biosynthesis of melanin protecting the fungal pathogen against host immune responses, membrane proteins of CFEM and major facilitator superfamily MFS1 with known functions in pathogenesis and multi-drug resistance, respectively. These results suggest that some pathogenicity pathways are pre-activated at the transcriptional level prior to the fungal infection of host plants. We developed two algorithms to confidently identify 1518 alternative splicing events in 1,259 Verticillium genes, representing 15.1% of multi-exonic genes. Among the events, 43.5% involving in novel splice sites were classified into nine AS types, the others belong to intron retention. Verticillium AS genes were exclusively enriched in the regulatory biological processes such as mycelium development, reproduction, morphogenesis, cell communication and signal transduction, predicting a primary function of AS regulation when the pathogen infecting its host. This work presents a transcriptome-wide approach for identifying the fungal virulence genes and pathogenicity mechanisms; both the methodology and mechanisms could be generally applicable to other fungal pathogens.

Project description:The soil-borne ascomycete Verticillium dahliae which causes wilt disease in many important crops results a severe reduction in crop yield and quality. During infection, the V.dahliae will secrete a lot of secondary metabolites which act as toxic factors to promote the infection process. However, the mechanism underlying how V.dahliae secondary metabolites regulate cotton infection remains largely unexplored. BRE1 is a highly conserved ubiquitin ligase (E3) enzymes and regulate expression of specific genes related to the production of secondary metabolites. To get an insight into the genes regulated by VdBre1, the comparative transcriptomic analysis was conducted between VdBre1 deletion mutant and the wild type V592. The conidia of wilde type and mutant were collected and total RNA was extracted using fungal RNA kit, sequencing was performed on an BGISEQ-500 platform.

Project description:Magnaporthe oryzae is notorious for its efficient evolution as a plant pathogen. Analysis of well organised fungal pathogenic mechanisms would enable the scientific understanding to fight against pathogens. Histidine phosphotransferase in fungal systems are good targets for antifungals since they are component of SAPK pathway, which is essential for survival of the fungus. The histidine Phosphotransferase YPD1 in M. oryzae is known to be light responsive and important for both asexual reproduction as well as pathogenicity. In this study, the knock-down (KD) mutant of YPD1 was highly sensitive to cell wall perturbing agents and were highly deficient in laccase expression and activity. A RNA-seq transcriptome screening of KD mutant compared to wild type revealed altered expression of about 146 genes important in biotrophic and necrotrophic phases of the pathogen. About 96 genes were found to be down regulated, of which several plant cell wall degrading genes vital for pathogenicity and fungal cell wall maintenance genes important for growth and fungal development were identified. Most of the genes identified are involved in biotrophic invasion and metabolism. Our results explain the reason for the deficiency of the KD mutant in asexual reproduction as well as pathogenicity. Analysis of differentially expressed genes in the KD YPD1 mutant suggests that YPD1 can be a good target for development of novel antifungal compounds, as YPD1 expression affects genes required for pathogenicity.

Project description:Purpose: Verticillium dahliae is a fungal plant pathogen. The goals of the study are to examine gene networks involved in controlling resistance to V. dahliae in potato using expression quantitative trait loci (eQTL) mapping. Methods: 91 clones from a diploid potato mapping population 15143 and the parental lines 07506-01 and 12120-03 from the Agriculture and Agri-Food Canada in Fredericton, New Brunswick, Canada were propagated in a V. dahliae-infested field in triplicate plots of five plants in a randomized complete block design. Tetraploid varieties Shepody, Hindenburg, Kennebec, Russet Burbank, Atlantic and Green Mountain were planted similarly and included in the analysis. Green Mountain was also planted in additional plots and used a guard row clone. Leaf samples were taken at 70 days after planting. Two µg of total RNA per sample was used to construct DeepSAGE tag libraries using a modification to facilitate direct sequencing of the amplicons by Illumina sequencing. The samples from 91 clones were bar-coded with a different identification key. Samples were diluted to a final concentration of 10 nM and were pooled. Each of the three biological replicates was done in a separate pool. Final pool concentrations were estimated using Quant-iT-PicoGreen prior to template DNA hybridization and sequencing on an Illumina Genome Analyzer II according to the manufacturer's instructions. Results: Tag count data were transformed to the relative unit of counts per million. Tags included in the analysis had to be present in at least one biological replicate. Clonal variation between samples was detected (p-value ≤ 0.05) using the exact test for a negative binomial distribution . Tags that did not have significant clonal variation were removed, leaving 6248 tags. The LS mean of tag count was calculated for each tag for each clone. The 21-nt tag sequences were aligned against PGSC S. tuberosum group Phureja DM1-3 transcripts (v3.4) using blastn. Hits with E-value ≤ 0.5 were filtered in. Where there were multiple blast hits meeting E-value criteria, only the one with the longest alignment was included. 4198 tags were assigned single-hit annotations in this manner and were used for eQTL analysis. Conclusions: The results indicated that for most genes the eQTL controlling expression was mapped near the gene (cis-eQTL). There were also many trans-eQTLs, where were eQTLs for a gene that were found on another chromosome particularly on chromosome 5.
