Identifying differential gene expression in TBT-treated dog whelk, Nucella lapillus

Project description:Imposex, the superimposition of male sexual characteristics in females, is caused by tributyltin (TBT) and provides one of the best ecological examples of anthropogenically-induced endocrine disruption in aquatic ecosystems. We used combinations of 454 Roche pyrosequencing and microarray technologies to understand the functional genomic basis of imposex in the prosobranch gastropod, Nucella lapillus, a recognised sentinel for TBT-induced imposex.

Imposex in the dogwhelk (Nucella lapillus): 22-year monitoring around England and Wales.

Project description:Six imposex surveys in the dogwhelk (Nucella lapillus) have been conducted over the past two decades to assess legislation effectiveness controlling the use of tri-butyl tin (TBT) by the maritime shipping industry. This study firstly analysed the results of the 2014 survey and secondly carried out a trend assessment of the same 88 sampled sites between 1997 and 2014 of which 65 showed statistically significant reductions. To highlight the magnitude of change, the Vas Deferens Sequence stages (VDS) of the same 56 sites sampled in 1997 and 2010 showed that the Vas Deferens Sequence Index (VDSI) reduced statistically significantly from 2.89 and 0.42, respectively. These data confirm that the legislation enacted, latterly through the International Maritime Organization (IMO) during 2003 to 2008, has been effective in progressively reducing the impact of TBT in the marine environment. Nevertheless, the 2014 results show that two of 18 sampled sites (Gurnard Bay and St. Mawes) are still above the Ecotoxicological Assessment Criteria (EAC) derived for TBT (VDSI ≤ 2).

AFLPs reveal different population genetic structure under contrasting environments in the marine snail Nucella lapillus L.

Project description:Dispersal has received growing attention in marine ecology, particularly since evidence obtained with up-to-date techniques challenged the traditional view. The dogwhelk Nucella lapillus L., a sedentary gastropod with direct development, is a good example: dispersal was traditionally assumed to be limited until studies with microsatellites disputed this idea. To shed some light on this controversy, the genetic structure of dogwhelk populations in northwest Spain was investigated with highly polymorphic AFLP markers giving special attention to the influence of hydrodynamic stress. In agreement with the expectations for a poor disperser, our results show a significant genetic structure at regional (<200 km) and areal scales (<15 km). However, the spatial genetic structure varied with wave-exposure in the present case study: IBD was evident under sheltered conditions but absent from the exposed area where genetic differentiation was stronger. Our results provide evidence that differences in wave-exposure can exert a detectable influence on the genetic structure of coastal organisms, even in species without a planktonic larva.

First visual record of rare purple-colored dogwhelks ( Nucella lapillus) on the Atlantic coast of Nova Scotia, Canada.

Project description:The dogwhelk Nucella lapillus is a rocky intertidal gastropod of the North Atlantic coast. Individual shell color varies. Common colors range between white and brown, with darker dogwhelks being more affected by heat stress than lighter-colored conspecifics. Other reported shell colors are purple, black, mauve, pink, yellow, and orange from UK coasts, red and gray from the Bay of Fundy coast of New Brunswick and Nova Scotia (Canada), and purple, black, gray, yellow, and orange from the coasts of Maine and Massachusetts (USA), with purple being considered as a rare color. On the Atlantic coast of Nova Scotia, dogwhelks are active from April until November, but information on dogwhelk shell color is missing for this coast. On 16 June 2016, we found two purple-colored dogwhelks in the mid-to-high intertidal zone of a moderately wave-exposed rocky shore near Duncans Cove, on the Atlantic coast of Nova Scotia while collecting dogwhelks (n= 1000) during low tide for manipulative field experiments. All other dogwhelks collected on that day were of common white and brown colors. During earlier dogwhelk collections in Atlantic Nova Scotia (between 2011-2013) and field surveys in Duncans Cove (between 2014-2016), we did not find any purple-colored dogwhelks, indicating the rareness of this color in that region. Apparently, our observations provide the first visual record of rare purple-colored dogwhelks on the Atlantic coast of Nova Scotia, Canada.

Plastic and heritable components of phenotypic variation in Nucella lapillus: an assessment using reciprocal transplant and common garden experiments.

Project description:Assessment of plastic and heritable components of phenotypic variation is crucial for understanding the evolution of adaptive character traits in heterogeneous environments. We assessed the above in relation to adaptive shell morphology of the rocky intertidal snail Nucella lapillus by reciprocal transplantation of snails between two shores differing in wave action and rearing snails of the same provenance in a common garden. Results were compared with those reported for similar experiments conducted elsewhere. Microsatellite variation indicated limited gene flow between the populations. Intrinsic growth rate was greater in exposed-site than sheltered-site snails, but the reverse was true of absolute growth rate, suggesting heritable compensation for reduced foraging opportunity at the exposed site. Shell morphology of reciprocal transplants partially converged through plasticity toward that of native snails. Shell morphology of F(2)s in the common garden partially retained characteristics of the P-generation, suggesting genetic control. A maternal effect was revealed by greater resemblance of F(1)s than F(2)s to the P-generation. The observed synergistic effects of plastic, maternal and genetic control of shell-shape may be expected to maximise fitness when environmental characteristics become unpredictable through dispersal.
