Molecular interplay between dormant bone marrow-resident cells (BMRCs) and CTCs in breast cancer.

Project description:Despite widespread knowledge that bone marrow-resident breast cancer cells (BMRCs) affect tumor progression, signaling mechanisms of BMRCs implicated in maintaining long-term dormancy have not been characterized. To overcome these hurdles, we developed a novel experimental model of tumor dormancy employing circulating tumor cells (CTCs) derived from metastatic breast cancer patients (de novo CTCs), transplanted them in immunocompromised mice, and re-isolated these cells from xenografted mice bone marrow (ex vivo BMRCs) and blood (ex vivo CTCs) to perform downstream transcriptomic analyses. Here we report that the balance between mTORC1 vs mTORC2 signaling regulates BMRC mitotic and/or dormancy characteristics.

RNA-seq analysis of freshly isolated and ex-vivo cultured human cord blood CD34+ cells

Project description:RNA-seq analysis was performed to compare differentially expressed genes in freshly isolated and ex-vivo cultured human cord blood CD34+ cells. Mitochondrion related genes are upregulated in CD34+ hematopoietic stem and progenitor cells upon ex vivo culture. In vivo transplantation experiments demonstrate that stemness of CD34+ cells is significantly decreased due to oxidative stress induced by ex vivo culture.

Identification of a stable molecular signature in breast tumor endothelial cells that persists ex vivo

Project description:Long-term, in vitro propagation of tumor-specific endothelial cells (TEC) allows for ex vivo functional studies and gene expression profiling of clonally-derived, well-characterized subpopulations. Using a genetically-engineered mouse model (GEMM) of mammary adenocarcinoma (C3-TAg), we have optimized an isolation procedure and defined growth conditions for long-term propagation of breast TEC. Based on expression of EC-selective markers, the isolated TEC maintain their endothelial specification and phenotype in culture. Furthermore, gene expression profiling of multiple TEC subpopulations revealed striking, persistent overexpression of several candidate genes including Irx2 and Zfp503 (transcription factors), Cd166 and Cd133 (cell-surface markers), Ccl12 and neurotensin (angiocrine factors), and Gpr182 and Cnr2 (G protein-coupled receptors). Taken together, we have described an effective method for isolating and culture-expanding breast TEC from a GEMM and we have uncovered several new TEC-selective genes whose overexpression persists even after long-term in vitro culture. Breast cancer remotely imposes a myeloid bias on hematopoietic stem cells by reprogramming the bone marrow niche

Project description:Increased number of circulating myeloid cells is a hallmark of several cancers, however it remains unclear how primary tumors impact on myelopoiesis. Here we show that non-metastatic breast tumors remotely instruct the fate of long-term hematopoietic stem cell (HSCLT) in the bone marrow. We found that HSCLT from tumor bearing mice acquire a myeloid bias persisting upon primary and secondary HSCLT transfer in lethally-irradiated tumor-free animals. By imaging the bone marrow HSC niche, we found that the tumor-bearing status is associated with increased physical interactions between mesenchymal stem/stromal cells (MSC) and HSCLT. Moreover, ex vivo co-culture experiments demonstrate that MSC isolated from tumor-bearing mice increase myeloid differentiation of HSCLT isolated from tumor free mice. In summary, our data reveal that breast cancer remotely promotes myelopoiesis at the earliest stages of hematopoietic differentiation in the bone marrow mesenchymal niche.

Gene expression profile of A549 Circulating Tumor Cells from 4D Model.

Project description:CTCs in cancer patients are thought to be responsible for metastasis. Currently, there is no ex vivo model that can isolate this group of cells. We have developed an ex vivo 4D lung cancer model that forms perfusable tumor nodules and form CTCs. Gene array analyses show 2504 differentially expressed genes when comparing CTCs from the 4D model seeded with A549 cells to the same cells grown on a petri dish (2D). We compared the gene expression profile (Human OneArray v5 chip) of A549 cells, human lung cancer cell line, grown on petri dish (2D) and same cells circulating as tumor cells in our ex vivo model (4D/CTC).
