Project description:Analysis of testis-specific over-expression of Musashi-1 and Musashi-2 at gene expression level. The hypothesis tested in the present study was that testis-specific over-expression of Musashi-1/Musashi-2 would result in differential gene expression profiles from the Wildtype control. Results provide important information of the genes affected by Musashi-2 overexpression during spermatogenesis. Total RNA obtained from whole adult testis of transgenic testis-specfic Musashi-1 and Musashi-2 overexpression mice compared to Wildtype (control) littermates. 4 replicates, 16 samples, 2 comparisons: Wt (Musashi-1) vs. TgMsi1 & Wt (Musashi-2) vs. TgMsi2

Project description:To explore the mechanism of pachytene pirna biogenesis, we employed a proteomics approach using flag-tagged adad2 transgenic mice expressed fully functional N-terminal epitope-tagged HA-ADAD2 and performed anti-FLAG immunoprecipitation coupled with quantitative mass spectrometry (IP-MS) from testis extracts of adult wt and transgenic mice.
Project description:To futher find the gene expression due to the aromatase overexpression in the mouse testis,we have employ whole genome microarray expression profiling as a discovery platform to identify genes compared to the wild type mice. The testes were taken from 5 month old aromatase overexpression mice and WT mice as control. We investigated genes that were significantly upregulated or downregulated in AROM+ mouse testis compared with WT testis. Two groups: 1) total RNA from three 5 month old AROM+ overexpression transgenic mice testes were collected and processed for microarray;2) total RNA from three 5 month old WT mice testes as control.

Project description:We have employed whole genome microarray expression profiling as a discovery platform to identify the effect of plin1 on gene expression during testis development. Testis of day 21, day 40, and day 60 in plin1-null mice and wild type mice were analyzed. Expression of a list genes (including cyp51, prm1, tnp2) from this signature was quantified in the same RNA samples by real-time PCR, confirming the influence of plin1 on testis development.
