Project description:To determine functional differences between the corpus luteum (CL) of the estrous cycle and pregnancy in cows, gene expression profiles between the CL of the estrous cycle and pregnancy were investigated. A 15 K bovine oligo DNA microarray detected 138, 265 and 455 differentially expressed genes (>2-fold; P<0.05) in the bovine CL of 20-25, 40-45, and 150-160 days of pregnancy compared with 10-12 days of the estrous cycle. The different gene expression profiles may contribute to functional differences between the CL of pregnancy and the CL of the estrous cycle in cows. Chemokines including eotaxin and lymphotactin may regulate CL function during pregnancy.

The day of artificial insemination was designated as day 1.

The aim was to compare the expression of miRNA between the early-cycle (Day4) and midcycle (days9-12) CL The three cows designated for early-cycle time point received an injection of gonadotropin releasing hormone (GnRH; Factrel; Zoetis, Florham Park, NJ) to synchronize the ovulation of a dominant follicle, and were slaughtered 4 days later to collect a day 4 CL. The three cows designated for midcycle time point were observed after CIDR removal to determine the onset of estrus, and on days 9-12 of the estrous cycle, the CL was collected by culpotomy.

CL were collected from heifers at 0, 4 and 24 h following PG injection on d 4 and 11 of the estrous cycle (n = 5 animals/treatment) and isolated RNA labeled and hybridized to Affymetrix GeneChip Bovine Genome Arrays. At 4 and 24 h post PG respectively, 221 (d 4) and 661 (d 11) and 248 (d 4) and 1419 (d 11) regulated genes were identified.

Project description:The aim of the present study is to determine possible roles of chemokines in regulating bovine endometrial function during early pregnancy. A 15 K bovine oligo DNA microarray detected 344 and 1336 differentially expressed genes (>2-fold; P<0.05) in the bovine endometrium of 15 and 18 days of pregnancy compared with these days of the estrous cycle (cyclic, n=4). The expression of six chemokines, including CCL2, CCL8, CCL11, CCL14, CCL16 and CXCL10 was higher in the endometrium of early pregnancy than that in the cyclic stage.

Our model of the bovine estrous cycle is a set of ordinary differential equations which generates hormone profiles of successive estrous cycles with several follicular waves per cycle. It describes the growth and decay of the follicles and the corpus luteum, as well as the change of the key reproductive hormones, enzymes and processes over time. In this work we describe recent developments of this model towards the administration of prostaglandin F2α. We validate our model by showing that the simulations agree with observations from synchronization studies and with measured progesterone data after single dose administrations of synthetic prostaglandin F2α.
