Project description:The Varroa mite represents the main threat of honey bees (Apis mellifera). Bees from some colonies can limit the proliferation of this parasite by detecting and removing parasitized brood, such behavior is defined as Varroa sensitive Hygiene (VSH). This is an important issue for selecting colonies that can survive Varroa outbreaks. We therefore study the molecular meachnisms underlying this behavior by comparing the antennae transcriptomic profile of VSH and non-VSH bees. Those profiles were further compared to to the profiles of nurses and forager profiles involved in brood care and food collection, respectively.

Project description:Apis mellifera syriaca is the native honeybee subspecies of Jordan and much of the Middle East. It expresses behavioral adaptations to a regional climate with very high temperatures, nectar dearth in summer, attacks of the Oriental wasp Vespa orientalis and in most cases it is resistant to varroa mites. The Thorax control sample of A. m. syriaca in this experiment was originally collected and stored since 2001 from Wadi Ben Hammad a remote valley in the southern region of Jordan. Using morphometric and Mitochondrial DNA markers it was proved that bees from this area had show higher similarity than other samples collected from the Middle East as represented by reference samples collected in 1952 by Brother Adam. The samples L1-L5 are collected from the National Center for Agricultural Research and Extension breading apiary which was originally established for the conservation of Apis mellifera syriaca. Goal was to use the genetic information in the breeding for varroa resistant bees and to determine the successfulness of this conservation program. Project description:Experiment was designed to study the effect of Deformed wing virus (DWV) and the mite Varroa destructor on on siRNA and miRNA composition using high-throughput sequencing of small RNA in developing worker honeybee (Apis mellifera). Newly hatched bee larvae (day 3 of bee development) were transferred from a Varroa-free colony with low DWV levels to a Varroa-infested colony with high levels of DWV in bees and Varroa mites. All transferred larvae were receiving the DWV strains present in this Varroa-infested colony with the food delivered by the nurse bees until their capping (day 8). About half of these larvae were capped with Varroa mite and were subjected to the mite piercing and feeding on their haemolymph during pupal development until sampling at purple eye stage (day 14). Exposure to the mite piercing and feeding resulted in about 1000-fold increase of the DWV levels in the majority of the mite-exposed pupae compared to the control pupae and the pupae not exposed to Varroa mites.

Project description:Previously, we attempted to identify the molecular basis for lateralization in honey bee workers by comparing protein expression between left and right antennae of hygienic bees using mass spectrometry-based quantitative proteomics (PXD005242). We identified 1,845 unique proteins but found no significant differences; therefore, we suggested this may be because of insufficient depth of coverage. To improve proteome coverage, we fractionated the peptides from left and right antennae from 4 highly hygienic colonies and repeated the comparison. Here, we identified 3,114 unique proteins (a 69% improvement), which is among the highest proteome coverage achieved in honey bees. However, we still did not identify significant differences. We also compared the proteomes of antennae that were injected with mock dsRNA, dsRNA targeting OBP18, and uninjected. We found a strong effect of microinjecting alone, but no knockdown of our target.

Project description:In other experiments, we found that hygienic honey bees show lateralized responses to odors associated with brood disease. Bees are also known to have more olfactory sensilla on their right antenna compared to their left. Following this, we performed proteomics on right and left antennae of hygienic honey bees in order to identify a molecular basis for lateralization (N = 5); however, we did not identify any significantly different proteins.

Project description:The honeybee brain is comprised of a nervous system that sufficiently regulates this life transition. Knowledge about how protein phosphorylation functions in regards to the neurobiological activities in the honeybee brain to drive the age-specific labor division is still lacking. Protein phosphorylation, the most common post-translational modification (PTM), is a key switch for rapid on-off control of signaling cascades that regulate cell differentiation and development, enzyme activity and metabolic maintenance in living cells. A fundamental mechanism for regulating signaling network and protein activity is the covalent PTM of serine (Ser), threonine (Thr), and tyrosine (Tyr) residues with phosphate. Fortunately, because of advances in phosphopeptide enrichment and improvements in mass spectrometry (MS) instrumentation and methods, phosphoproteomics has enabled large-scale identification of protein phosphorylation sites and phosphorylation networks in biological samples. Although the proteome has been mapped in the brain of nurse and forager bees, knowledge about age-specific effects of phosphorylation regulation on proteins in the honeybee brain is still lacking. Moreover, information in regards to the honeybee phosphoproteome is also very limited. Only very recently, in-depth phosphoproteomics analyses of protein phosphorylation networks in the hypopharyngealgland of the honeybee have been reported. Although the phosphoproteome analyses during the development of brood and salivary glands has been reported, only very limited proteins were phosphorylated and phosphorylation sites of those phosphoproteins were not assigned. Therefore, a comprehensive characterization of phosphoproteomics and changes in the brains of nurse and forager bees is key to understand the phosphorylation events underlying age-specific physiology to achieve the completion of biological missions in this well-organized social community of the honeybee. Honeybee (A. m. ligustica) colonies used for sampling were raised at the apiary of the Institute of Apicultural Research, Chinese Academy of Agricultural Science, Beijing. Newly emerged (<12 h after emergence) worker bees were marked on their thoraxes and placed back into the colonies to develop and then the marked nurse and forager bees were collected on days 10 and 20, respectively. There were 150 bees sampled from each of the five colonies which have queens at the same age. In brief, for each time point, worker bees were sampled from five colonies, and pooled all samples for further analysis. This procedure was repeated three times, so that we finally ended up with three independent biological replicates per time point, each consisting of 150 honeybees. Then their brains were dissected, and the brain samples were pooled and stored at −80 °C for further analysis. All the colonies were managed with almost identical population, food, and brood during the nectar flow of chaste berry (Vitexnegundo L.) in June.

Project description:The objective of the study was to assess the technical error due to blending of individual samples into pools in different experimental data sets. The blending error variance component corresponds to random effects for inaccuracies, which were modeled on the logarithmic scale of normalized gene expression. It's estimation based on a linear mixed model, fitted for each transcript. Honey bees showing hygienic behavior towards Varroa-parasitized brood were compared with controls and among each other. Tissues from mushroom body, antennal lobe and Antennae of honey bees were processed for microarray analysis. They were measured as single samples as well as pooled samples from 2 or 4 individuals.
