We analyzed mRNA expression pattern of 6 primary hepatocellular carcinomas and their corresponding nontumourous liver parenchyma. The specimens were obtained from a tissue bank in our hospital. All the clinical informations were de-linked from the specimens.

Project description:Tumor cells were microdissected from FFPE sections of hepatocellular carcinoma (HCC) samples. Samples were compared between different localisations either within or around the tumor. miRNA was labeled with the Affymetrix FlashTag Biotin HSR RNA Labeling Kit 6 microdissected hepatocellular carcinoma samples were compared to four different tumor localisations (LC: cirrhotic septa of the tumor-adjacent liver; LP: tumor-adjacent liver parenchyma;TC: fibrinous capsule of the tumor; TP: tumor parenchyma)
Project description:Liver cancer was the fourth leading cause of cancer-related deaths worldwide in 2018, of which Hepatocellular carcinoma accounts for 75-85% of all liver cancers and is therefore the most common type of liver cancer. In this study, total RNA were extracted from sections of formalin-fixed paraffin embedded (FFPE) archival human liver biopsies from 24 patients, including ten HCV-associated HCC (HCV-HCC), ten virus-unrelated HCC (VU-HCC) and four normal liver patients. By comparing the expression profiles of HCV-HCC and VU-HCC to that of the normal liver biopsies respectively, 18 lncRNAs were identified to be differentially expressed.

Project description:To identify the novel tumor suppressors in hepatocellular carcinoma (HCC), we have employed whole genome microarray expression profiling as a discovery platform in HCC and paired normal liver tissues to identify genes which down-regulated in HCC. Among which, INTS6 and its pseudogene, namely INTS6P1, were found to be dramatically down-regulated in HCC. The down-regulated expression of INTS6 and INTS6P1 in HCC was further confirmed by real-time PCR.
