The regional distribution of resident immune cells shapes distinct immunological environments along the murine epididymis

Project description:Previously, the epididymal regions were demonstrated to react strikingly different to inflammatory stimuli. We therefore aimed at untangling the heterogeneity of extravascular CD45+ cells throughout epididymal regions and identifying their transcriptional profile. Using an unbiased scRNASeq approach, 12 distinct populations were identified in total showing striking differences in their distribution along the epididymal duct. Our data indicate that resident immune cell populations are strategically positioned within the organ to provide distinct immunological environments required to maintain tissue integrity and to adequately tackle invading pathogens ascending through the urogenital tract. This study provides an important common research platform and starting point for future functional studies of distinct populations under physiological and pathological conditions.

Characterization of resident CD8+ Trm cell populations using whole-genome biomarker analysis

Project description:This study seeks a better understanding of the functional differences between naturally acquired murine CD8+ T resident memory (Trm) cells of three distinct mucosal environments; the small intestine, the lung and the liver. CD8+ Trm cells of healthy young adult mice were retrieved from their respective organ environments by automated organ disintegration and MACS sorting, and compared with each other by whole genome gene expression profiling. Peripheral blood derived circulating CD8+ Tem cells were also analyzed, serving as nonresident peripheral memory T cell controls, in an attempt to identify common characteristics of resident mucosal CD8+ Trm subsets, as well.

Treg cells from human blood differentiate into non-lymphoid tissue-resident effector cells upon TNFR2 costimulation

Project description:Regulatory T (Treg) cells can facilitate transplant tolerance and attenuate autoimmune- and inflammatory diseases. Therefore, it is clinically relevant to stimulate Treg cell expansion and function in vivo and to create therapeutic Treg cell products in vitro. We report that TNF receptor 2 (TNFR2) is a unique costimulus for naïve, thymus-derived (t)Treg cells from human blood that promotes their differentiation into non-lymphoid tissue (NLT)-resident effector Treg cells, without Th-like polarization. In contrast, CD28 costimulation maintains a lymphoid tissue (LT)-resident Treg cell phenotype. We base this conclusion on transcriptome and proteome analysis of TNFR2- and CD28-costimulated CD4+ Treg cells and conventional T (Tconv) cells, followed by bioinformatic comparison with published transcriptomic Treg cell signatures from NLT and LT in health and disease, including autoimmunity and cancer. These analyses illuminate that TNFR2 costimulation promotes Treg cell capacity for survival, migration, immunosuppression and tissue regeneration. Functional studies confirmed improved migratory ability of TNFR2-costimulated tTreg cells. Flow cytometry validated the presence of the TNFR2-driven Treg cell signature in effector/memory Treg cells from the human placenta as opposed to blood. Thus, TNFR2 can be exploited as driver of NLT-resident Treg cell differentiation for adoptive cell therapy or antibody-based immunomodulation in human disease.

Project description:This study seeks a better understanding of the functional differences emerging between mouse CD8+ T effector (Teff) cells infiltrating three distinct mucosal environments; the small intestine, the lung and the liver. CD8+ Teff cells immigrating in the target organs were generated using a murine experimental acute graft versus host disease (aGVHD) model. For this purpose, congenially labeled CD45.1 OT-I CD8+ Teff cells were transplanted to and activated in a lethally irradiated allogeneic CD45.2 host constitutively expressing their cognate antigen, the chicken ovalbumin protein. CD8+ Teff cells were retrieved from their respective organ environments by automated organ disintegration and MACS sorting, and compared with each other by whole genome gene expression profiling.
