Project description:Brain metastatic disease occurs in 10-30% of metastatic breast cancer cases. The incidence of brain metastases is increasing with median overall survival < 2 years for patients. In order to better characterize oncogenic pathway activity pertinent to breast cancer brain metastasis, exome capture RNA sequencing was carried out on patient matched primary breast with brain metastatic tumor samples for 45 cases of breast cancer brain metastasis (N= 90 samples). Here, exome capture RNA sequencing data is deposited as sequencing batch corrected log2 transformed trimmed M of means (TMM) normalized counts per million (CPM) (log2(TMM-CPM +1) gene expression values (n=16,714 protein coding genes; N=90 tumor samples).

SIVA mutation, a candidate metastasis gene identified from clonally related bilateral breast cancers, promotes breast cancer cell aggressiveness in vitro and in vivo

Project description:Our purpose is to identify candidate genes involved in the early steps of breast cancer metastasis and examine their pro-invasive functions both in vitro and in vivo. A percentage of bilateral breast cancers were clonally related based on copy number variation profiling. Whole exome sequencing and comparative sequence analysis revealed that a limited number of somatic mutations were acquired in this “breast to breast” metastasis. These mutations might promote breast cancer distant spread. The pro-invasive functions of a candidate metastasis gene were assessed in vitro by its abilities to promote proliferation, migration and invasion and in vivo as tumor xenografts in immunocompromised mice or a syngeneic orthotopic mouse breast cancer model. RNAseq analysis was performed to probe the transcription programs modulated by this candidate metastasis gene. SIVA1-D160N was one somatic mutation acquired in the breast to breast metastasis. Over-expression of SIVA1-D160N promoted migration and invasion of human MB-MDA-231 breast cancer cells in vitro, consistent with a dominant negative interfering function. When introduced via tail vein injection, 231 cells over-expressing SIVA1-D160N displayed enhanced distant spread on IVIS imaging. Over-expression of SIVA1-D160N promoted anchorage independent growth of mouse 4T1 breast cancer cells in vitro. When introduced orthotopically via mammary fat pad injection in syngeneic Balb/c mice, over-expression of SIVA1-D160N in 4T1 cells increased mammary gland tumor growth as well as liver metastasis. We conclude clonally related bilateral breast cancers represent a novel system to investigate metastasis and revealed a role of SIVA1-D160N in breast cancer metastasis.

RET overexpression leads to increased brain metastatic competency in luminal breast cancer patients

Project description:Breast cancer brain metastasis is a rising occurrence, necessitating a better understanding of the mechanisms involved for effective management. Breast cancer brain metastases diverge significantly from the primary tumor, with gains in kinase and concomitant losses of steroid signaling observed. In this study, we explored the role of the kinase receptor RET in promoting breast cancer brain metastasis and provide a rationale for targeting the receptor in this patient cohort. Here, exome capture RNA sequencing data is deposited as sequencing batch corrected log2 transformed trimmed M of means (TMM) normalised counts per million (CPM) (log2(TMM-CPM +1) gene expression values for the patients described in this study (n=19,622 protein coding genes; N=16 tumour samples).

Integrative analysis of mesenchymal stromal cell conversion by breast cancer secretomes in blank slate 3D microenvironments

Project description:The ability of primary tumour cells to invade and metastasise is responsible for over 90% of cancer patient deaths. During tumour growth and progression, cancer cells secrete factors that recruit and reprogram otherwise healthy cells to facilitate the formation of a favourable tumour microenvironment. Here, we have investigated how breast cancer cells convert normal mesenchymal stromal cells (MSCs) into tumour-associated MSCs (TA-MSCs) using unbiased global approaches. We compared the secretomes produced by non-invasive MCF-7 cells with invasive MDA-MB-231 cells, and identified extracellular matrix and exosome components associated with invasion. We then treated MSCs cultured in fully-defined, synthetic 3D hydrogels with invasive/non-invasive cancer secretomes and analysed their responses by kinase activity profiling and RNA sequencing, which led us to identify an invasion-associated signature of MSC conversion. Finally, we investigated whether there is an organ-specific metastasis-associated reprogramming of MSCs, and found that breast cancer cells from different metastatic sites have different secretome profiles that induce reprogramming of MSCs. These data describe at a systems-level how breast cancer cells with different invasion and metastatic abilities secrete molecules that activate MSCs and convert them into TA-MSCs.
