Project description:Dendrite aberration is a common feature of neurodegenerative diseases caused by protein toxicity, but the underlying mechanisms remain largely elusive. Here, we show that nuclear polyglutamine (polyQ) toxicity resulted in defective terminal dendrite elongation accompanied with a loss of Golgi outposts (GOPs) and decreased supply of plasma membrane (PM) in Drosophila Class IV da (C4da) neurons. mRNA sequencing revealed that down-regulated genes by polyQ proteins included many secretory pathway-related genes, including COPII genes regulating GOP synthesis. Transcription factor enrichment analysis identified CREB3L1/CrebA, which regulates COPII gene expression. CrebA overexpression in C4da neurons restored the dysregulation of COPII genes, GOP synthesis, and PM supply. The ChIP-PCR assay revealed that CrebA expression was regulated by CBP, which was sequestered by polyQ proteins. Furthermore, co-overexpression of CrebA and Rac1 synergistically restored the polyQ-induced dendrite pathology. Collectively, our results suggest that GOPs impaired by polyQ proteins contribute to dendrite pathology through the CBP-CrebA-COPII pathway.

Project description:Recycling and supply of plasma membrane (PM), referred to as PM turnover, are essential for maintaining integrity of neurons undergoing continuous dynamic morphological changes. Yet, the link of PM turnover to neuronal pathophysiology remains largely elusive. Here we showed that a subset of toxic polyglutamine proteins markedly compromised local targeting-recycling of PM components, reflecting a unique feature of early neuropathies. Such disruption of PM turnover in dendrites was ascribed primarily to alterations in dendritic endosomes and Golgi outposts accounting for local PM recycling and supply, respectively. Genome-wide mRNA-sequencing analysis identified CREB3L1/CrebA-COPII pathway to be responsible for the disruption of PM turnover. Indeed, CrebA overexpression reversed PM turnover impairment caused by these toxic proteins through restoring COPII pathway. Thus we demonstrate how PM turnover is linked to aberration of neuronal integrity in pathological conditions as well as its near-complete restoration through genetic engineering of CREB3L1/CrebA-COPII pathway.

Project description:Loss of function of dystonin cytoskeletal linker proteins causes neurodegeneration in the sensory ataxia, dystonia musculorum (dt). While much investigation has focused on understanding dt pathology, divergent functions of dystonin isoforms are still unknown. Here, we highlight a novel function of the dystonin-a2 isoform in mediating microtubule (MT) stability, golgi organization and flux through the secretory pathway. Using dystonin-null mice combined with isoform-specific loss of function analysis, we find dystonin-a2 is bound to MAP1B in the area surrounding the centrosome, where it maintains MT acetylation. In dt, the absence of the MAP1B-dystonin-A2 interaction results in a loss of MAP1B perinuclear localization, leading to MT deacetylation and instability. Deacetylated MTs lead to golgi fragmentation and prevent anterograde trafficking of motor proteins. Maintenance of MT acetylation through TSA administration or MAP1B overexperssion in vitro, mitigates the observed defect. These aberrations are apparent in pre-phenotype dorsal root ganglia (DRG) and primary sensory neurons, suggesting they are causal in the dt disorder. P4 dorsal root ganglia (DRG) tissue from 3 WT and 3 dt27J animals from 2 separate litters was subjected to RNA extraction and analyzed.

Project description:Local translation within excitatory and inhibitory neurons is known to be involved in neuronal development and synaptic plasticity. Despite the extensive dendritic and axonal arborizations of monoaminergic neurons, the subcellular localization of protein synthesis has not been well-characterized in these populations. Here, we investigated mRNA localization in midbrain dopaminergic (mDA) neurons, cells with enormous axonal and dendritic projections, both of which can release dopamine (DA). Using highly-sensitive sequencing and imaging approaches in mDA axons, we found no evidence for axonal mRNA localization or translation. In contrast, we found that mDA neuronal dendritic projections into the substantia nigra reticulata (SNr) contain ribosomes and mRNAs encoding the DA synthesis, release, and reuptake machinery. Surprisingly, we found dendritic localization of mRNAs encoding synaptic vesicular release proteins in mDA neurons. Our results are consistent with a role for local translation in the regulation of DA transmission from dendrites, but not striatal axons. Finally, we defined a molecular signature of sparse mDA neurons in the SNr, including enrichment of an ER calcium pump previously undescribed in mDA neurons.

Project description:Spinocerebellar ataxia type 3 (SCA3) is one of the polyglutamine (polyQ) diseases, which are caused by a CAG repeat expansion within the coding region of the associated genes. The CAG repeat specifies glutamine, and the expanded polyQ domain with mutation confers dominant toxicity on the protein. Traditionally, studies have focused on protein toxicity in polyQ disease mechanisms. Recent findings, however, demonstrate that the CAG repeat RNA, which encodes the toxic polyQ protein, also contributes to the disease in Drosophila. To provide insight into the nature of the RNA toxicity, we extracted brain-enriched RNA from flies expressing a toxic CAG repeat mRNA (CAG100) and a non-toxic interrupted CAA/G mRNA repeat (CAA/G105) for microarray analysis. This approach identified a set of genes that are differentially expressed specifically in CAG100 flies.

Project description:Local translation is a conserved molecular mechanism. It allows a cell with a complex shape to bypass somatic protein synthesis and transport, and thus to respond quickly to a local stimulus. Local translation also contributes to the establishment of molecular and functional polarity. In the brain, it has been extensively studied in neurons and has also been described more recently in astrocytes - a type of glial cell with specialized extensions that contact vessels and synapses. Here, we studied perisynaptic astrocytic processes (PAPs) in the dorsal hippocampus and showed that they contain RNAs, ribosomes, the endoplasmic reticulum-Golgi intermediate compartment, particles of the Golgi apparatus, and protein synthesis events. We used our recently refined polysome immunoprecipitation technique to characterize the pool of polysomal mRNAs in PAPs (which we refer to as the “PAPome”) from the dorsal hippocampus and compared it with the polysomal mRNAs found in the astrocyte as a whole. The polysomal transcripts that were enriched in the PAPome encoded mostly cytoplasmic proteins and defined an unexpected molecular repertoire with the most enriched transcripts coding for proteins involved in iron homeostasis, translation, cell cycle and cytoskeleton. Interestingly, among them Erz, Fth1, and Rplp1 were enriched in PAPs compared to perivascular astrocytic processes indicating that local translation differ at these two interfaces and may sustain distinct molecular properties. The PAP-enriched transcripts Flt1, Fth1, Ccnd2, Mdm2, Gnb2l1 and Eef1a1 code for proteins involved in memory and learning mechanisms. We therefore studied their local translation in the context of fear conditioning (i.e. behavior involving the dorsal hippocampus). We observed changes in the density and/or distribution of these mRNAs in astrocytes processes as well as a drop of their translation specifically in PAPs. Our results highlight unexpected molecular properties of hippocampal PAPs and suggest for the first time that local translation in this perisynaptic compartment is linked to fear-related memory.

Project description:Hexanucleotide repeat expansion in the C9ORF72 gene is the most frequent inherited cause of amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). Poly-GR is one of the most toxic dipeptide repeat (DPR) proteins translated from the RNA repeats. It has been shown to affect protein synthesis, but how this contributes to neurodegeneration is not clear. Here, we found that poly-GR inhibits global translation by perturbing translation elongation. We identified that the transcripts with relatively slow elongation rate tend to be further stalled by poly-GR in iPSC-differentiated neurons. This increases ribosome collision and ZAKα-mediated ribotoxic stress response (RSR), which elevates the phosphorylation of p38 and promotes cell death. Knockdown of ZAKα or pharmacological inhibition of p38 can ameliorate the GR toxicity, and improve the survival of C9ORF72-ALS/FTD patient-derived iPSC-neurons. Our study reveals molecular mechanism of poly-GR mediated toxicity on global translatome, and identifies RSR as a potential therapeutic target for C9ORF72-ALS/FTD.

Project description:G4C2 hexanucleotide repeat expansions in a non-coding region of the C9orf72 gene are the most common cause of familial amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). G4C2 insertion length is variable, and patients can carry up to several thousand repeats. Dipeptide repeat proteins (DPR) translated from G4C2 transcripts are thought to be a main driver of toxicity. Experiments in model organisms with relatively short DPR have shown that arginine-rich DPR are most toxic, while polyGlycine-Alanine (GA) DPRs cause only mild toxicity. However, GA is the most abundant DPR in patient brains, and experimental work in animals has generally relied on the use of low numbers of repeats, with DPRs often tagged for in vivo tracking. Whether repeat length, or tagging, affect the toxicity of GA has not been systematically assessed. Therefore, we generated Drosophila fly lines expressing GA100, GA200 or GA400 specifically in adult neurons. Consistent with previous studies, expression of GA100 and GA200 caused only mild toxicity. In contrast, neuronal expression of GA400 drastically reduced climbing ability and survival of the flies, indicating that long GA DPRs can be highly toxic in vivo. This toxicity could be abolished by tagging GA400. Proteomics analysis of fly brains showed a repeat-length-dependent modulation of the brain proteome, with GA400 causing earlier and stronger changes than shorter GA proteins. PolyGA expression up-regulated proteins involved in ER to Golgi trafficking, and down-regulated proteins involved in insulin signalling. Experimental down-regulation of Tango1, a highly conserved regulator of ER-to Golgi transport, partially rescued GA400 toxicity, suggesting that misregulation of this process contributes to polyGA toxicity. Experimentally increasing insulin signaling also rescued GA toxicity. In summary, our data show that long polyGA proteins can be highly toxic in vivo, and that they may therefore contribute to ALS/FTD pathogenesis in humans patients.

Project description:To find differentially expressed genes by comparing Fuss neurons and mutant Fuss neurons we utilised targeted DamID in Drosophila melanogaster. This not only predicts many genes expressed and nonexpressed in adult Fuss neuron populations, but also reveals Fuss being expressed in gustatory neurons, especially bitter gustatory neurons. Comparison of control and mutant datasets reveals a reduced expression of gustatory receptor gene Gr66a. Gr66a is an important component of the gustatory receptors compounds responsible for toxic compound sensation. Behaviourial tests reveal an impaired toxic compound sensation of adult Fuss mutant flies.
