Project description:25-hydroxycholesterol has been demonstrated to regulate SREBP processing, yet Ch25h-deficient mice have no cholesterol abnormalities. Using RNA-seq, we find that LPS-stimulated, Ch25h-deficient BMDMs have dysregulated SREBP target genes, demonstrating that 25-hydroxycholesterol is an induced repressor of SREBP during inflammatory settings. mRNA profiles from day 7 M-CSF bone marrow-derived macrophages (wild type, Ch25h-knockout, LXR-double knockout, were generated by deep sequencing on an Illumina HiSeq 2500. Sequence reads passing a quality filter were aligned to the mouse genome (mm10) using STAR.

Project description:Endothelial progenitor cells (EPCs) promote the maintenance of the endothelium by secreting vasoreparative factors. A population of EPCs known as early outgrowth cells (EOCs) are being investigated as novel cell-based therapies for the treatment of cardiovascular disease. We previously demonstrated that the absence of liver x receptors (LXRs) is detrimental to the formation and function of EOCs under hypercholesterolemic conditions. Here, we investigate whether LXR activation in EOCs is beneficial for the treatment of atherosclerosis. EOCs were differentiated from the bone marrow of wildtype (WT) and LXR-knockout (Lxrαβ-/-) mice in the presence of vehicle or LXR agonist (GW3965). This data set is a proteomic comparison of EPCs at day 1 after isolation and day 9 of GW39565 treatment compared to day 9 of vehicle treatment.
Project description:To further understanding the function of cullin 3 during inflammation in macrophages, we have employed mouse bone marrow derived macrophages microarray expression profiling to identify the gens that involve in regulationg inflammatory respones upon LPS challenge. Mouse BMM were stimulated with LPS for 6 hours and RNA was extracted for microarray. Ogt was indentified by comparison between wildtype and Cullin 3 knockout BMM.
Project description:Bone marrow derived macrophages were isolated from C57BL/6J (LXR positive) and LXRalpha +LXRbeta double knockout mice (DKO) that had been treated with vehicle, Kado2-lipid A (KLA, and inflammatory stimulus), acetylated LDL (AcLDL, a mimic of pro-atherogenic lipoprotein particles) and GW3965 (GW, a synthetic LXR agonist) for 17 hours. Following treatment RNA was isolated and RNA-Seq was performed.

Upon differentiation, BMDMs or iMacs were left untreated or stimulated as described below. 1) BMDM WT were stimulated with LPS, PGE2, IL-10, LPS+PGE2, LPS+IL-10 for 4 hours: each condition in triplicate.
