Project description:How the complex interaction between Mycobacterium tuberculosis (Mtb) and the host is regulated during infection is still not well understood. Using a systems biology approach, we demonstrate here that miR-155 is one of several microRNAs that regulate host gene expression over the first 48 hours of Mtb infection in macrophages. miR-155 regulates the cell survival of Mtb-infected macrophages through SHIP1/AKT signaling. Using timecourse gene expression data, we constructed a miRNA regulatory network for the innate immune response to Mtb infection by WT macrophages. The network suggested a role for seven miRNAs in regulating the host response to Mtb, with miR-155 being one of them. We then validated a role for miR-155 by comparing the response between WT and miR-155-/- macrophages.

Bone marrow from either wild type or miR-155-/- C57Bl/6 mice was differentiated into dendritic cells by incubating with GM-CSF. These cells were then stimulated with LPS, and gene expression was performed.

Project description:Mammalian microRNAs (miRNAs) are emerging as key regulators of the development and function of the immune system. Here, we report a strong but transient induction of miR-155 in mouse bone marrow after injection of bacterial lipopolysaccharide (LPS) correlated with granulocyte/monocyte (GM) expansion. Demonstrating the sufficiency of miR-155 to drive GM expansion, enforced expression in mouse bone marrow cells caused GM proliferation in a manner reminiscent of LPS treatment. However, the mir-155-induced GM populations displayed pathological features characteristic of myeloid neoplasia. Extending possible relevance to human disease, miR-155 was overexpressed in the bone marrow of patients with acute myeloid leukemia (AML). Furthermore, miR-155 repressed a subset of genes implicated in hematopoietic development and disease. These data implicate miR-155 as a contributor to physiological GM expansion during inflammation and to certain pathological features associated with AML, emphasizing the importance of proper miR-155 regulation in developing myeloid cells during times of inflammatory stress. Project description:Human bone marrow mesenchymal stromal cells are capable of limited self-renewal and multi-lineage differentiation in vitro. Several studies have demonstrated that microRNAs, post-transcriptional modifiers of protein expression, play crucial roles in the regulation of these complex processes. To gain knowledge regarding the role of microRNAs in human adipocyte regulation, we examined the microRNA expression profile of the immortalized human bone marrow-derived stromal cell line hMSC-Tert20. We identified 12 microRNAs that were differentially expressed during adipogenesis, of which several have previously been shown to play important roles in adipocyte biology. The expression of miR-155, miR-221 and miR-222 decreased during the adipogenic program, suggesting that they act as negative regulators of differentiation. Interestingly, adenovirus-mediated expression of either miR-155 alone or miR-221 plus miR-222 significantly inhibited adipogenesis and repressed induction of the master regulators C/EBP α and PPARgamma Our study provides the first experimental evidence that miR-155, miR-221 and miR-222 function in human adipocyte differentiation.

However, the molecular mechanisms that underlie the oncogenic role of miR-155 in B-cell lymphoma are not well understood. To identify the miR-155 targets relevant for B-cell lymphoma, we performed RNA immunoprecipitation of Argonaute 2 in Hodgkin lymphoma (HL) cells upon miR-155 inhibition and in BL cells upon ectopic expression of miR-155. We identified 54 miR-155-specific target genes in BL cells and confirmed miR-155 targeting of DET1, NIAM, TRIM32, HOMEZ, PSIP1 and JARID2. Five of these targets are also regulated by endogenous miR-155 in HL cells. Both overexpression of miR-155 and inhibition of expression of the novel miR-155 target gene NIAM increased proliferation of BL cells. In primary B-cell lymphoma NIAM-positive cases have significant lower levels of miR-155 as compared to NIAM-negative cases, suggesting that NIAM is also regulated by miR-155 in primary B-cell lymphoma. Thus, our data indicate an oncogenic role for miR-155 in B-cell lymphoma which involves targeting the tumor suppressor NIAM.
